The comprehensive identification of organic species existing in indoor environments is a key issue to understand their impact in human health. This study proposes the analysis of condensed water samples, collected with portable dehumidifiers, to characterize semi-volatile compounds in the gas phase of confined areas. Water samples are concentrated by solid-phase extraction (SPE). The obtained extracts are analysed by gas chromatography (GC) time-of-flight mass spectrometry (TOF-MS), following a nontarget screening data mining approach. In first term, spectra of deconvoluted compounds are compared with those in NIST low resolution library; thereafter, tentative identifications are verified using an inhouse database of accurate electron ionization (EI) MS spectra. Chromatographic (retention index) and spectral data are combined for unambiguous species identification. The potential of condensed water samples to reflect changes in the composition of indoor atmospheres, the match between data obtained using different dehumidifiers, and the relative concentration efficiency of condensed water compared to that attained by active sampling of moderate air volumes are discussed. A total of 141 semi-volatile compounds were identified (98 confirmed against authentic standards) in a set of 21 samples obtained from different homes and working places. This list contains more than 40 fragrances (including several potential allergens), solvents and intermediates in the production of polymeric materials, plasticizers and flame retardants. Microtubule targeting agents represent a very active arena in the development of anticancer agents. In particular, compounds binding at the colchicine site in tubulin are being deeply studied, and the structural information recently available on this binding site allows structure-directed design of new ligands. Structural comparison of our recently reported high resolution X-Ray structure of the cyclohexanedione derivative TUB075 bound to tubulin and the tubulin-DAMA-colchicine complex has revealed a conformational change in the alpha T5 loop. By a grid-based computational analysis of the tubulin-DAMA-colchicine binding site, we have identified a new favourable binding area in the colchicine-site that was unexplored by our lead TUB075. Thus, based on a structure-guided design, new cyclohexanedione derivatives have been synthesized and tested for tubulin binding and in cellular assays. As a result, we have identified diphenyl ether derivatives with IC50 values around 10-40 nM against three different tumor cell lines and affinity constants for tubulin similar to that of colchicine around 10(7) M-1. As expected, they halted the cell cycle progression at G2/M phase at concentrations as low as 0.08 mu M. (C) 2019 Elsevier Masson SAS. All rights reserved.

Significant advancements in C-N bond formationviaC-H bond functionalisation have made it a staple in the production of nitrogen-containing compounds in both industry and academia. However, transition metal-free synthesis, particularly in the case of C(sp(3))-N formation, has remained a significant challenge to the synthetic community. Herein we report a procedure for alpha-C(sp(3))-H amination of ethereal compounds through use of azodicarboxylates as the nitrogen source and freely-available atmospheric oxygen to access ethereal radical intermediatesviaaerobic C-H activation. The use of fluorinated alcohols as solvent is observed to greatly increase the efficiency of the reaction and we show experimentally and theoretically the key role of H-bonding between fluorinated alcohols and azodicarboxylates. Calculations of the condensed Fukui functions of a H-bonded fluorinated alcohol-azodicarboxylate complex correlates with a significantly increased susceptibility of azodicarboxylates to undergo reaction with radicals, which informs a number of recent reports in the literature.

In the present study, we report the synthesis of a series of copper(II) complexes with a wide range of ligands and their testing in the copper catalyzed Chan-Evans-Lam (CEL) coupling of aniline and phenylboronic acid. The efficiency of the coupling was directly connected with the ease of the reduction of Cu(II) to Cu(I) of the complexes. The most efficient catalyst was derived from 4-t-butyl-2,5-bis[(quinolinylimino)methyl]phenolate and two Cu(II) ions. Depending on the counter-anion nature and the concentration of the reaction mixture, the reaction can be directed to predominant C-N-bond formation. Forty-three derivatives of diphenylamine were prepared under the optimized conditions. The proposed mechanism of the catalysis was based on the reduction potential of a series of complexes, molecular weight measurements of the catalytic complex in MeOH and the kinetic studies of aniline and phenylboronic acid coupling. In addition, an H-1 NMR experiment in a sealed NMR tube, without external oxygen supply available, proved that no complete Cu(II) to Cu(I) conversion was observed under the condition, ruling out the usually accepted mechanism of the C-N coupling, which included the oxygenation of the intermediately formed Cu(I) complexes after the key step of C-N conversion had already been completed. Instead, a mechanism was proposed, involving an oxygen molecule coordinated to two copper ions in the key C-N bond formation without any detectable conversion of the Cu(II) complexes to Cu(I).

Thiobenzoic acid (TBA) can serve as a single-electron reducing agent under photoirradiation from a blue light-emitting diode, in the presence of appropriate electron acceptors, and the resulting sulfur-centered radical species undergoes hydrogen atom abstraction. This dual-role catalysis by TBA enables regioselectivie C alpha-H arylation of benzylamines, benzyl alcohols, and ethers, as well as dihydroimidazoles, with cyano(hetero)arenes in good yield, without the need for a transition-metal photocatalyst and/or synthetically elaborated organic dyes.

Background Despite tremendous progress has been achieved in tumor theranostic over the past decade, accurate identification and complete eradication of tumor cells remain a great challenge owing to the limitation of single imaging modality and therapeutic strategy. Results Herein, we successfully design and construct BiVO4/Fe3O4@polydopamine (PDA) superparticles (SPs) for computed tomography (CT)/photoacoustic (PA)/magnetic resonance (MR) multimodal imaging and radiotherapy (RT)/photothermal therapy (PTT) synergistic therapy toward oral epithelial carcinoma. On the one hand, BiVO4 NPs endow BiVO4/Fe3O4@PDA SPs with impressive X-ray absorption capability due to the high X-ray attenuation coefficient of Bi, which is beneficial for their utilization as radiosensitizers for CT imaging and RT. On the other hand, Fe3O4 NPs impart BiVO4/Fe3O4@PDA SPs with the superparamagnetic property as a T-2-weighted contrast agent for MR imaging. Importantly, the aggregation of Fe3O4 NPs in SPs and the presence of PDA shell greatly improve the photothermal conversion capability of SPs, making BiVO4/Fe3O4@PDA SPs as an ideal photothermal transducer for PA imaging and PTT. By integrating advantages of various imaging modalities (CT/PA/MR) and therapeutic strategies (RT/PTT), our BiVO4/Fe3O4@PDA SPs exhibit the sensitive multimodal imaging feature and superior synergistic therapeutic efficacy on tumors. Conclusions Since there are many kinds of building blocks with unique properties appropriating for self-assembly, our work may largely enrich the library of nanomateirals for tumor diagnosis and treatment.

Lignin valorization is significantly influenced by the degrees of lignin condensation measured by the amount of cleaved beta-O-4 ether linkages and interunit C-C linkages formed during biomass fractionation. To minimize the cleavage of aryl ether and formation of C-C linkages, a p-toluenesulfonic acid (p-TsOH)/choline chloride (ChCl) solvent system with hydrotropic properties was evaluated. The ChC1 was chosen to control the H+ concentration of the p-TsOH/ChCl solvent system. Compared with p-TsOH/H2O, the degradation of beta-O-4 was kept in a similar rate during p-TsOH/ChCl fractionation; however, the formation of C-C linkage was slower. Under 75/25 p-TsOH/ChCl mass ratio at 80 degrees C for 20 min, 76.6 % of miscanthus lignin, with beta-O-4 content of 30.9 %, and 88.9 % of wheat straw lignin, with beta-O-4 content of 33.6 %, can be dissolved. In addition, the low polar and layered structure of hydrotropic p-TsOH/ChCl system has a strong ability to dissolve lignin, which resulted in lignin with larger particle size and lower surface charge. The results also indicated that lignin from p-TsOH/ChCl solvent system has great potential for lignin depolymerization to aromatics for valorization.

Introduction: Gestational exposure to chemical mixtures, which is prevalent among pregnant women, may be associated with adverse childhood neurodevelopment. However, few studies have examined relations between gestational chemical mixture exposure and children’s cognitive abilities. Methods: In a cohort of 253 pregnant women and their children from Cincinnati, OH (enrolled 2003-2006), we quantified biomarker concentrations of 43 metals, phthalates, phenols, polybrominated diphenyl ethers, organophosphate and organochlorine pesticides, polychlorinated biphenyls, perfluoroalkyl substances, and environmental tobacco smoke in blood or urine. Using k-means clustering and principal component (PC) analysis, we characterized chemical mixtures among pregnant women. We assessed children’s cognitive abilities using the Wechsler Preschool and Primary Scale of Intelligence-III and Wechsler Intelligence Scale for ChildrenIV at ages 5 and 8 years, respectively. We estimated covariate-adjusted differences in children’s cognitive ability scores]=cross clusters, and with increasing PC scores and individual biomarker concentrations. Results: Geometric mean biomarker concentrations were generally highest, intermediate, and lowest among women in clusters 1, 2, and 3, respectively. Children born to women in clusters 1 and 2 had 5.1 (95% CI: 9.4,-0.8) and 2.0 (95% CI: 5.5, 1,4) lower performance IQ scores compared to children in cluster 3, respectively. PC scores and individual chemical biomarker concentrations were not associated with cognitive abilities. Conclusions: In this cohort, combined prenatal exposure to phenols, certain phthalates, pesticides, and perfluoroalkyl substances was inversely associated with children’s cognition, but some individual chemical biomarker concentrations were not. Additional studies should determine if the aggregate impact of these chemicals on cognition is different from their individual effects.
