A series of programmed cell death-1 (PD-1)/programmed cell death ligand 1 (PD-L1) inhibitors based on the resorcinol diphenyl ether scaffold were discovered by incorporating hydrophilic moieties into the side chain and converting into the corresponding hydrochloride salt. Among these compounds, P18 showed the highest inhibitory activity against PD-1/PD-L1 with an IC50 value of 9.1 nM in a homogeneous time-resolved fluorescence binding assay. Besides, P18 promoted HepG2 cell death dose dependently in a HepG2/PD-L1 and Jurkat/PD-1 coculture cell model. Further, P18 demonstrated significantly higher water solubility (17.61 mg/mL) and improved pharmacokinetics (e.g., t(1/2) of similar to 20 h and oral bioavailability of 12%) than the previous analogues. Moreover, P18 was highly effective in suppressing tumor growth in an immune checkpoint humanized mouse model without apparent toxicity. Collectively, these results suggest that compound P18 represents a promising PD-1/PD-L1 inhibitor worthy of further investigation as a potential anticancer agent.

Here we present a protocol for analyses of axon regeneration and density in unsectioned adult mouse spinal cord. This includes methods for injury and tracing of dorsal column sensory and corticospinal axons; clearing and staining of unsectioned spinal cord; visualization of axon degeneration and regeneration in cleared and uncleared specimens using two-photon microscopy; and either manual or semi-automatic analysis of axon density and regeneration in 3D space using Imaris and ImageJ software. This protocol can be used to elucidate the molecular and cellular mechanisms underlying nervous system degeneration and regeneration and to establish the therapeutic efficacy of candidate neuroregenerative treatments. Because tissue sectioning is not required, this protocol enables unambiguous evaluation of regeneration and greatly accelerates the speed at which analyses can be conducted. Surgical procedures take < 30 min per mouse, with a wait period of 2 weeks between axonal injury and tracing and 2-8 weeks between tracing and tissue processing. Clearing and immunolabeling take similar to 1-2 weeks, depending on the size of the sample. Imaging and analysis can be performed in 1 d. All these procedures can be accomplished by a competent graduate student or experienced technician. Dynamic self-assembly represents an effective approach to form energy-dissipative structures by introducing interactions among multiple building blocks with a continuous energy supply. Time-dependent magnetic fields are treated as convenient energy inputs to construct such a dynamic self assembled system. The induced interactions can be further tuned by modulating the input field, resulting in a diversity of assembled patterns. However, formation of a functional dynamic pattern with controllability remains a challenge. Herein, we report the formation and pattern control of dynamically self-assembled magnetic droplets at an air liquid interface, energized by a precessing magnetic field. The formation process involves the assembly of magnetic microparticles into particle chains inside droplets, and then highly ordered patterns are generated by balancing the induced interactions among droplets. By modulating the input field, the interactions and collective behaviors are adjusted and the pattern can be reversibly tuned, i.e., expand and shrink, in a controlled manner. Furthermore, the assembled droplets are able to be steered in two-dimensional as an entity by applying a magnetic field gradient. Utilizing dynamic pattern control and steerability of the assembled structure, cargoes are successfully trapped, transported, and released in a noncontact fashion, indicating that the dynamically assembled droplets can act as a reconfigurable untethered robotic end-effector for manipulation.

Optical tissue clearing using dibenzyl ether (DBE) or BABB (1 part benzyl alcohol and 2 parts benzyl benzoate) is easy in application and allows deep-tissue imaging of a wide range of specimens. However, in both substances, optical clearing and storage times of enhanced green fluorescent protein (EGFP)-expressing specimens are limited due to the continuous formation of peroxides and aldehydes, which severely quench fluorescence. Stabilisation of purified DBE or BABB by addition of the antioxidant propyl gallate efficiently preserves fluorescence signals in EGFP-expressing samples for more than a year. This enables longer clearing times and improved tissue transparency with higher fluorescence signal intensity. The here introduced clearing protocol termed stabilised DISCO allows to image spines in a whole mouse brain and to detect faint changes in the activity-dependent expression pattern of tdTomato.

We report herein the possibility to perform the hydrosilylation of carbonyls using actinide complexes as catalysts. While complexes of the uranyl ion [UO2](2+) have been poorly considered in catalysis, we show the potentialities of the Lewis acid [UO2(OTf)(2)] (1) in the catalytic hydrosilylation of a series of aldehydes. [UO2(OTf)(2)] proved to be a very active catalyst affording distinct reduction products depending on the nature of the reductant. With Et3SiH, a number of aliphatic and aromatic aldehydes are reduced into symmetric ethers, while (Pr3SiH)-Pr-i yielded silylated alcohols. Studies of the reaction mechanism led to the isolation of aldehyde/uranyl complexes, [UO2(OTf)(2)(4-Me2N-PhCHO)(3)], [UO2(mu-kappa(2)-OTf)(2)(PhCHO)](n), and [UO2(mu-kappa(2)-OTf(kappa(1)-OTf)(PhCHO)(2)](2), which have been fully characterized by NMR, IR, and single-crystal X-ray diffraction.

Electron-rich arenes are oxidatively photochlorinated in the presence of catalytic amounts of bromide ions, visible light, and 4CzIPN as organic photoredox catalyst. The substrates are brominated in situ in a first photoredox-catalyzed oxidation step, followed by a photocatalyzed ipso-chlorination, yielding the target compounds in high ortho/para regioselectivity. Dioxygen serves as a green and convenient terminal oxidant. The use of aqueous hydrochloric acid as the chloride source reduces the amount of saline by-products.

Nanosized self-assemblies built from inorganic nanoparticles and polymer ligands have the potential to generate personalized theranostics systems for diagnostic imaging and cancer therapy. However, most of the theranostics systems suffer from poor targeting activity, insensitive diagnosis and drug leakage, leading to poor treatment results. In this study, a hierarchical tumor acidity-responsive magnetic nanobomb (termed HTAMN) was developed for photodynamic therapy and diagnostic imaging. The HTAMNs were formed through the self-assembly of chlorin e6 (Ce6)-functionalized polypeptide ligand, methoxy poly (ethyleneglycol)-block-poly (dopamineethylenediamine-2,3-dimethylmaleic anhydride)-L-glutamate-Ce6 [mPEG-b-P (Dopa-Ethy-DMMA)LG-Ce6] and superparamagnetic iron oxide nanoparticles (SPIONs). Negatively charged HTAMNs circulate in the blood for prolonged periods and promote tumor retention by passive targeting to the tumor. Once the HTAMNs arrive at the tumor location, the acidic extracellular tumor environment reverses the surface charge of the HTAMNs, resulting in tumor accumulation and cellular uptake. Moreover, in response to the more acidic environment inside cells, the photosensitizers are activated resulted in enhanced diagnostic imaging and cancer treatment. The in vitro and in vivo results indicate the effective tumor accumulation, internalization, diagnostic sensitivity and superior photodynamic therapy effect of the HTAMNs. Therefore, designing smart HTAMNs can promote the rapid development of cancer theranostics for clinical implementation.

Non-equilibrium plasma technology provides an unconventional but promising solution for the cleaning of tar contaminated bio-syngas in biomass gasification. This work is focused on the reforming of mixed naphthalene (C10H8) and toluene (C7H8) as typical single ring and double ring tar model components using a gliding arc discharge (GAD) reactor. The influence of naphthalene content, steam/carbon molar ratio and discharge power on the destruction of C10H8 and C7H8 was evaluated to understand their effects on the tar conversion, gas yield, and the energy consumption. Adding H2O to the plasma process forms OH radicals, creating additional reaction routes for the step-wised oxidation of naphthalene, toluene and their fragments towards the production of CO, CO2, and water. The highest conversion of naphthalene and toluene (similar to 80% overall) was obtained when the steam/carbon molar ratio changed between 1.0 and 1.5, which was dependent on the balance between two opposite effects due to the presence of steam: positive effect of OH radicals and negative effect of electron attachment on water molecules. The highest energy efficiency (53.6 g/kWh) was obtained for the conversion of mixed tar compounds, which is by far the highest in previously reported works. CO, H-2 and C2H2 were the major gases with the highest CO yield of 38.6% and H-2 yield of 39.1%. Emission spectroscopic diagnostics was employed to understand the contribution of chemically reactive species to the GAD reaction. Possible reaction pathways in the plasma reforming of mixed naphthalene and toluene were proposed based on the plasma spectroscopic diagnostics coupled with a comprehensive analysis of major gas products and condensable by-products.

Many experimental studies have demonstrated that titanium dioxide nanoparticles (TiO2 NPs) could induce adverse health effects in vivo and in vitro. But epidemiological evidence and biomarkers related to early health effects are still lacking. This study aimed to explore biomarkers in the urine samples of workers occupationally exposed to a relatively low concentration of TiO2 NPs. A cross-sectional study was conducted in Jinan, China, involving 132 employees of a TiO2 NP manufacturing plant, among which the exposed group and control group were 1 : 1 matched by confounding factors such as gender, age, BMI, smoking and drinking. Untargeted metabolomics was performed in urine samples using high performance liquid chromatography-mass spectrometry (HPLC-MS) technology. The differential metabolites between the TiO2 NP exposed group and the control group were analyzed and then screened for potential biomarkers using bioinformatics methods. Metabolomics found a total of 1760 differentially expressed metabolites in the TiO2 NP exposed group, of which 60 differential metabolites were simultaneously confirmed by one-dimensional and multi-dimensional statistical analysis. Among these 60 differential metabolites, the relative expression of 27 metabolites increased, and the remaining 33 decreased. Pathway enrichment analysis further found that the metabolic pathway of long chain acyl-coa dehydrogenase deficiency (Lcad) was significantly enriched. Ten differential metabolites were selected as potential biomarkers of occupational exposure to TiO2 NPs using machine learning methods, including dibenzyl ether, quassimarin, tryptophan, etc. The receiver operating characteristic curves (ROCs) of these potential biomarkers showed good sensitivity and specificity. These potential biomarkers also had biological basis for occupational exposure to TiO2 NPs. Therefore, urine metabolites represented by dibenzyl ether are considered as good biomarkers of early health effects for occupational exposure to TiO2 NPs.

A novel ferrierite type zeolite named as IEZ-Al-PLS-3(S4R) with pore sizes of 14 x 12-ring (R) was post synthesized by simply interlayer silylation of lamellar precursor Al-PLS-3 taking 1,3,5,7-tetramethylcyclosilone (TMCS) as silane. Its catalytic properties were tested by isomerization/disproportionation of m-xylene, alkylation/acylation of anisole with benzyl alcohol/acetic anhydride. The catalytic performance of IEZ-Al-PLS-3(S4R) was compared with Al-PLS-3 (10 x 8-R) and IEZ-Al-PLS-3(1Si) (12 x 10-R) which were prepared by calcination or silylation using dimethoxydimethylsilane (DEDMS) as silane of as made Al-PLS-3 respectively. Since these materials have the same laminates composition, the differences of catalytic performance can be considered as the result of different pore sizes. We also prepared conventional 10-R zeolite ZSM-5 and 12-R zeolite Beta for control experiments. IEZ-Al-PLS-3(S4R) showed even higher catalytic activities than Beta as a result of its enlarged 14-R pore size.
