Summary: Naturally occurring peptides often possess macrocyclic and N-methylated backbone. These features grant them structural rigidity, high affinity to targets, proteolytic resistance, and occasionally membrane permeability. Because such peptides are produced by either nonribosomal peptide synthetases or enzymic posttranslational modifications, it is yet a formidable challenge in degenerating sequence or length and preparing libraries for screening bioactive mols. Here, we report a new means of synthesizing a de novo library of “natural product-like” macrocyclic N-methyl-peptides using translation machinery under the reprogrammed genetic code, which is coupled with an in vitro display technique, referred to as RaPID (random nonstandard peptides integrated discovery) system. This system allows for rapid selection of strong binders against an arbitrarily chosen therapeutic target. Here, we have demonstrated the selection of anti-E6AP macrocyclic N-methyl-peptides, one of which strongly inhibits polyubiquitination of proteins such as p53.

Experiments were carried out with the objective of enhancing shelf life and maintain quality of fresh-cut radish slices during storage at 8°C. Dip treatment of radish slices in 20 g/L glucose solution for five minutes retained the quality attributes viz., surface color, sensory properties and antioxidant capacity of the slices till six days of storage. Isothiocyanates were also retained better due to glucose dip. Biplot generated through principal component anal. of head space volatiles from fresh and stored radish slices showed that fresh and glucose treated slices grouped together with 4-Me thio-3-butenyl isothiocyanate. Accumulation of sulphurous volatiles such as Me disulfide, di-Me trisulfide, 2-pentanethiol was observed in control (undipped) radish slices, indicating their probable role as spoilage indicator volatiles. Thus, glucose pre-treatment can be considered as a practical method for quality retention of fresh-cut radish.

In the present work, a set of anthracene maleimide monomers with different aliphatic side groups obtained by Diels Alder reactions were used as precursors for a series of polymers of intrinsic microporosity (PIM) based homo- and copolymers that were successfully synthesized and characterized. Polymers with different sizes and shapes of aliphatic side groups were characterized by size-exclusion chromatog. (SEC), (NMR) 1H-NMR, thermogravimetric (TG) anal. coupled with Fourier-Transform-IR (FTIR) spectroscopy (TG-FTIR) and d. measurements. The TG-FTIR measurement of the monomer-containing Me side group revealed that the maleimide group decomposes prior to the anthracene backbone. Thermal treatment of homopolymer methyl-100 thick film was conducted to establish retro-Diels Alder rearrangement of the homopolymer. Gas and water vapor transport properties of homopolymers and copolymers were investigated by time-lag measurements. Homopolymers with bulky side groups (i-propyl-100 and t-butyl-100) experienced a strong impact of these side groups in fractional free volume (FFV) and penetrant permeability, compared to the homopolymers with linear alkyl side chains. The effect of anthracene maleimide derivatives with a variety of aliphatic side groups on water vapor transport is discussed. The maleimide moiety increased the water affinity of the homopolymers. Phenyl-100 exhibited a high water solubility, which is related to a higher amount of aromatic rings in the polymer. Copolymers (methyl-50 and t-butyl-50) showed higher CO2 and CH4 permeability compared to PIM-1. In summary, the introduction of bulky substituents increased free volume and permeability while the maleimide moiety enhanced the water vapor affinity of the polymers.

Osteoporosis affects millions of people worldwide. As such, this study assessed the macrophage-dependent in vitro anti-osteoporosis, phytochem. profile and hepatotoxicity effects in zebrafish larvae of the stem bark extracts of P. africana. Mouse bone marrow macrophages (BMM) cells were plated in 96-well plates and treated with P. africana methanolic bark extracts at concentrations of 0, 6.25, 12.5, 25, and 50 μg/mL for 24 h. The osteoclast tartrate-resistant acid phosphatase (TRAP) activity and cell viability were measured. Lipopolysaccharides (LPS) induced Nitrite (NO) and interleukin-6 (IL-6) production inhibitory effects of P. africana bark extracts (Methanolic, 150 μg/mL) and β-sitosterol (100 μM) were conducted using RAW 264.7 cells. Addnl., inhibition of IL-1β secretion and TRAP activity were determined for chlorogenic acid, catechin, naringenin and β-sitosterol. For toxicity study, zebrafish larvae were exposed to different concentrations of 25, 50, 100, and 200 μg/mL P. africana methanolic, ethanolic and water bark extracts DMSO (0.05%) was used as a neg. control and tamoxifen (5 μM) and dexamethasone (40 μM or 80 μM) were pos. controls. The methanolic P. africana extracts significantly inhibited (p < 0.001) TRAP activity at all concentrations and at 12.5 and 25 μg/mL, the extract exhibited significant (p < 0.05) BMM cell viability. NO production was significantly inhibited (all p < 0.0001) by the sample. IL-6 secretion was significantly inhibited by P. africana methanolic extract (p < 0.0001) and β-sitosterol (p < 0.0001) and further, chlorogenic acid and naringenin remarkably inhibited IL-1β production The P. africana methanolic extract significantly inhibited RANKL-induced TRAP activity. The phytochem. study of P. africana stem bark revealed a number of chem. compounds with anti-osteoporosis activity. There was no observed hepatocyte apoptosis in the liver of zebrafish larvae. In conclusion, the stem bark of P. africana is non-toxic to the liver and its inhibition of TRAP activity makes it an important source for future anti-osteoporosis drug development.

The single-layer light-emitting diode was fabricated in a configuration of ITO (indium-tin oxide)/polymer/Al. Electroluminescence (EL) emission of POFB-PF and POFB-PK were shown at 475 and 458 nm, resp., corresponding to the pure blue emissions. And, a dialkoxy-Ph containing POFB-MEH-PPV showed greenish blue light at 494 nm. But, LED devices from synthesized polymers showed poor device performance and high turn on voltage. So, we fabricated light-emitting diodes (LEDs) from blend polymers composed of poly[2-methoxy-5-(2-ethylhexyl-oxy)-1,4-phenylenevinylene] (MEH-PPV) and POFB-MEH-PPV (POFB-PF or POFB-PK) as the emitting layers. The EL emission maxima of each blend polymers were in the range of 573-591 nm, which indicates that the emission is mainly due to MEH-PPV and POFB-MEH-PPV (POFB-PF or POFB-PK) contributes to the enhancement of the luminescence. And each blend polymers exhibited higher EL quantum efficiency compared with MEH-PPV at the same c.d.

The development of a robust amide-bond formation remains a critical aspect of N-methylated peptide synthesis. In this study, we synthesized a variety of dipeptides in high yields, without severe racemization, from equivalent amounts of amino acids. Highly reactive N-methylimidazolium cation species were generated in situ to accelerate the amidation. The key to success was the addition of a strong Broensted acid. The developed amidation enabled the synthesis of a bulky peptide with a higher yield in a shorter amount of time compared with the results of conventional amidation. In addition, the amidation can be performed by using either a microflow reactor or a conventional flask. The first total synthesis of naturally occurring bulky N-methylated peptides, pterulamides I-IV, was achieved. Based on exptl. results and theor. calculations, we speculated that a Broensted acid would accelerate the rate-limiting generation of acyl imidazolium cations from mixed carbonic anhydrides.
