Decabromodiphenyl ether (BDE-209) is a persistent environmental pollutant that poses great risks to human health and has been associated with glucose and lipid metabolism. However, the mechanisms by which BDE-209 disturbs glycolipid metabolism in the liver remain unclear. Therefore, this study sought to confirm the effects of BDE-209 on glycolipid metabolism in mice livers and L02 cells to elucidate potential mechanisms of action. In vivo BDE-209 exposure caused histological damage and lipid accumulation, elevated glucose, low-density lipoprotein, total cholesterol, and triglyceride levels, and decreased glycogen and high-density lipoprotein levels in mice livers. Moreover, in vitro BDE-209 exposure not only induced L02 cells cytotoxicity (i.e., reduced cell viability and increased LDH leakage and ROS generation) but also increased glucose and triglyceride concentrations in L02 cells. Furthermore, IdF-1, an activator of the PI3K-AKT pathway, markedly inhibited BDE-209-incluced glucose concentration increase in L02 cells and antagonized the inhibitory effect of BDE-209 on the PI3K/AKT/GLUT4 pathway by counteracting the changes in the expression levels of p-IRS, AKT, Pl3K, p-AKT, and GLUT4. Moreover, GW9662, a PPAR gamma inhibitor, blocked lipid accumulation and the upregulation of the mTOR/PPAR gamma/RXRa. pathway in L02 cells induced by BDE-209 by relieving the increases in p-mTOR, PPAR gamma, and RXR alpha protein expression levels. In summary, this study revealed that BDE-209 disrupted glycol ipid metabolism by inhibiting the Pl3K/AKT/GLUT4 pathway and activating the mTOR/PPAR gamma/RXR alpha pathway. (C) 2020 Elsevier B.V. Strengthened, conductivity-tunable, and low solvent-sensitive flexible conductive rubber films with a Zn2+-crosslinked one-body segregated network

A high-performance flexible conductive material based on cheap carboxymethyl chitosan (CMCS) and carboxylated styrene-butadiene rubber (XSBR) was fabricated via latex film-forming technique. Nano zinc oxide (ZnO) was introduced to react with CMCS, generating a Zn2+-crosslinked CMCS one-body segregated (Zn2+-COBS) network. Differing from reported conductive fillers with segregated structures, this Zn2+-COBS network was a continuous framework structure, which could be turned into conducting network by absorbing moisture. Superior tensile strength of 22.6 MPa was achieved with 30 wt% CMCS and 2.5 wt% ZnO owing to the reinforced concrete structure effect of Zn2+-COBS architecture. The order of volume resistance (Rv) of the films could be tailored between x 10(8) and x 10(4) Omega cm by changing the water content from 0 to similar to 20%, although at 20% water content the strength decreased to about 2 MPa. Particularly, the material with 30 wt% CMCS and 2.5 wt% ZnO achieved a Rv of 7.8 x 10(3) Omega cm with 18.7% water content. Thanks to the abundant hydrogen bonding interactions between CMCS chains, this special architecture provided a strong swelling restriction on the rubber matrix and dramatically improved the conducting stability of film in solvents. These results demonstrate a promising method to fabricate an economical, conductivity-tunable, and flexural film material with potential applications in humidity detectors or in solvent environments.

The combination of tetrahydrocurcumin (THC) and Centella asiatica extract (CAE) is expected to improve various parameters of premature aging. This study used pre-test post-test control group design. Mice were randomized into 5 test groups. The dorsal part of the mice was shaved, and then various anti-aging parameters, including skin sensitivity, moisture content, collagen levels, elasticity, and large pores, were measured using a skin analyzer. Furthermore, the dorsal was smeared with a cream base for the negative control (NC) group, a cream containing ethyl ascorbyl ether for the positive control (PC) group, a cream containing THC for THC group, a cream containing CAE for the CAE group, and a cream containing THC and CAE for the Combination group, each with a dose of 2 mg/cm2 before and after UV irradiation. After treatment for four weeks, the anti-aging parameters were measured again. The combination of THC and CAE could increase the collagen and elasticity levels compared to THC and CAE itself, but the increase was not significant. Based on all anti-aging parameters scores, THC and CAE’s combination did not provide more significant anti-aging activity than single THC and single CAE.

A polyphenolic biomacromolecule prepared from a flavonoid: Catechin as degradable microparticles

Catechin (CAT) was crosslinked with trimethylolpropane triglycidyl ether (TMPTGE) to obtain degradable poly(CAT) particles in a single step. Spherical p(CAT) particles with tens of micrometer size range and an isoelectronic point at pH 1.2 were obtained. The hydrolytic degradation of p(CAT) particles provided sustainable and extended release with 264 mg/g CAT release within 10 days at pH 7.4. Blood clotting and hemolysis assays demonstrated dose-dependent blood compatibility revealing higher blood compatibility for p(CAT) particles up to 10 mu g/ml concentration. The cytotoxicity results show that p(CAT) particles have almost no toxicity for CCD841 normal colon cells at 250 mu g/ml concentration in 24 h incubation time giving similar to 97% cell viability, whereas CAT molecules only provide similar to 34% cell viability.
