The reaction mixture was heated to 60 C for 12 h. After 12 h the reaction mixture was diluted with water (1000 mL) and washed with EtOAc (400 mLx2). The combined organics were washed with brine (100 mL), dried over anhydrous Na2SC>4, and concentrated under reduced pressure. The residue was purified by flash chromatography on silica (petroleum ether : EtOAc = 50: 1 to 20: 1) to afford 1-(4-bromophenyl)-3,3-dimethoxycyclobutanecarbonitrile. MS (ESI) Calc’d [M+H]+, 296; found, 296.

Intermediate 314A: 1-(4-Bromophenyl)-3,3-dimethoxycyclobutanecarbonitrile To a suspension of NaH (0.733 g, 18.32 mmol, 60% in mineral oil) in DMF (14 mL) was added 2-(4-bromophenyl)acetonitrile (1.796 g, 9.16 mmol) followed by 1,3-dibromo-2,2-dimethoxypropane (2.0 g, 7.64 mmol) and the reaction mixture was heated to 60 C. and stirred for 12 h. The reaction mixture was cooled to RT, poured into water (150 mL) and extracted with EtOAc (3*40 mL). The combined organic layer was washed with brine, dried over Na2SO4, filtered and the filtrate concentrated. The crude product was purified by silica gel chromatography (24 g REDISEP column, eluting with 5% EtOAc in hexane). Fractions containing the product were combined and evaporated to afford Intermediate 314A as a white solid (2 g, 88%). 1H NMR (300 MHz, DMSO-d6) delta ppm 7.68-7.57 (m, 2H), 7.46-7.40 (m, 2H), 3.18 (s, 3H), 3.08 (s, 1H), 3.07 (s, 3H), 3.05-3.02 (m, 1H), 2.74-2.71 (m, 1H), 2.70-2.67 (m, 1H).

The reaction was quenched with water (100 mL) at 0 C and extracted with EtOAc (2 x 100 mL). The organic extracts werecombined and washed with brine (2 x 50 mL), dried over Na2504, filtered, and concentrated under reduced pressure. The residue was purified by silica gel chromatography using petroleumn ether/EtOAc (4/1) as eluting solvents to afford the title compound as a yellow solid (12 g, 45% yield). ?H NMR(400 MHz, DMSO-d6)oe 7.36-7.28(m, 2H), 3.18(s, 3H), 3.05(s, 3H), 3.03 (d, J= 13.6 Hz, 2H), 2.77 (d, J= 13.6 Hz, 2H), 2.24 (d, J= 1.6 Hz, 3H). The resulting mixture was heated at 140 0C for 24 h and then cooled to RT. Saturated NH4Cl aqueous (400 ml) was added and the mixture extracted with hexane (2 x 190 ml). The combined organic phases were washed with H2O (2 x 250 ml) saturated NaHCO3 (2 x 250 ml), H2O (2 x 250 ml) and brine (250 ml). The organic phase was dried (Na2SO4), filtered and concentrated at reduced pressure to provide an orange oil. Purification by in vacuo distillation to remove excess of l,3-dibromo-2,2-dimethoxypropane (b.p. 60-62 0C, 0.1 mniHg) and diisopropylmalonic ester (b.p 78-80 0C, 0.1 mmHg) provided the title compound (20 g, 52%) as orange oil in the distillation flask. MS data: Calculated MH+ (289); Found 100% (MH+) m/z 289.NMR data: 1H NMR (360 MHz, CHLOROFORM- d) delta ppm 5.07 (2 H, spt, J=6.3 Hz), 3.16 (6 H, s), 2.70 (4 H, s), 1.24 (12 H, d, J=6.4 Hz).

The organic layers were combined, washed with brine, dried over Na2SC>4, filtered and concentrated in vacuo to afford a residue, which was purified by column chromatography on silica gel (PE : ethyl acetate =40: 1 to 30: 1) to give the title compound. MS (EI) m/z 296 [M+H]+.
