Photodynamic and Cold Atmospheric Plasma Combination Therapy Using Polymeric Nanoparticles for the Synergistic Treatment of Cervical Cancer

Integrating multi-modal therapies into one platform could show great promise in overcoming the drawbacks of conventional single-modal therapy and achieving improved therapeutic efficacy in cancer. In this study, we prepared pheophorbide a (Pheo a)/targeting ligand (epitope analog of oncoprotein E7, EAE7)-conjugated poly(gamma-glutamic acid) (gamma-PGA)/poly(lactide-co-glycolide)-block-poly(ethylene glycol) methyl ether (MPEG-PLGA)/hyaluronic acid (PPHE) polymeric nanoparticles via self-assembly and encapsulation method for the photodynamic therapy (PDT)/cold atmospheric plasma (CAP) combinatory treatment of human papillomavirus (HPV)-positive cervical cancer, thereby enhancing the therapeutic efficacy. The synthesized PPHE polymeric nanoparticles exhibited a quasi-spherical shape with an average diameter of 80.5 +/- 17.6 nm in an aqueous solution. The results from the in vitro PDT efficacy assays demonstrated that PPHE has a superior PDT activity on CaSki cells due to the enhanced targeting ability. In addition, the PDT/CAP combinatory treatment more effectively inhibited the growth of cervical cancer cells by causing elevated intracellular reactive oxygen species generation and apoptotic cell death. Moreover, the three-dimensional cell culture model clearly confirmed the synergistic therapeutic efficacy of the PDT and the CAP combination therapy using PPHE on CaSki cells. Overall, these results indicate that the PDT/CAP combinatory treatment using PPHE is a highly effective new therapeutic modality for cervical cancer.

Hypothesis: The alpha-sulfo alkyl ester, AES, surfactants are a class of anionic surfactants which have potential for improved sustainable performance in a range of applications, and an important feature is their enhanced tolerance to precipitation in the presence of multivalent counterions. It is proposed that their adsorption properties can be adjusted substantially by changing the length of the shorter alkyl chain, that of the alkanol group in the ester. Experiments: Surface tension and neutron reflectivity have been used to investigate the variation in the adsorption properties with the shorter alkyl chain length (methyl, ethyl and propyl), the impact of NaCl on the adsorption, the tendency to form surface multilayer structures in the presence of AlCl3, and the effects of mixing the methyl ester sulfonate with the ethyl and propyl ester sulfonates on the adsorption. Findings: The variations in the critical micelle concentration, CMC, the adsorption isotherms, the saturation adsorption values, and the impact of NaCl illustrate the subtle influence of varying the shorter alkyl chain length of the surfactant. The non-ideal mixing of pairs of AES surfactants with different alkanol group lengths of the ester show that the extent of the non-ideality changes as the difference in the alkanol length increases. The surface multilayer formation observed in the presence of AlCl3 varies in a complex manner with the length of the short chain and for mixtures of surfactants with different chains lengths. Base-Free Cross-Couplings of Aryl Diazonium Salts in Methanol: Pd-II-Alkoxy as Reactivity-Controlling Intermediate

Pd-catalyzed cross-coupling reactions of aryl diazonium salts are generally assumed to proceed via cationic Pd-II intermediates which in turn would be highly reactive in the subsequent transmetalation step. Contrary to this belief, we herein report our observation and rationalization of opposing reactivities of ArN2+ in Suzuki (=effective) and Stille (=ineffective) cross-couplings in MeOH. Our systematic experimental and computational studies on the roles of transmetalating agent, solvent, base and the likely involvement of in situ formed diazoether derivatives challenge the currently accepted mechanism. Our data suggest that the observed solvent dichotomy is primarily due to Pd-II-methoxy intermediates being formed, which are unreactive with arylstannanes, but highly reactive with arylboronic acids, complementing the Suzuki Pd-oxy mechanism with the direct demonstration of transmetalation of a Pd-II-alkoxy complex. Lewis acids were found to circumvent this reactivity divergence, promoting efficient couplings regardless of the employed conditions or coupling partners.

A series of titanium compounds containing bidentate ketiminate ligands are synthesized and structurally characterized. There is no previous work that utilizes the multi-solvent extraction and structure elucidation of Lactobacillus helveticus cell-free supernatant (CFS). In this study, the CFS of Lb. helveticus CNRZ 32 was extracted by ethyl acetate, diethyl ether, dichloromethane, and n-hexane solvents. Tumor-Activated Photosensitization and Size Transformation of Nanodrugs

Effective intratumoral distribution of anticancer agents with good tumor penetration is of practical importance for photo-chemotherapy. Herein, a metal-organic framework (MOF) assisted strategy is reported for smart delivery of aggregation-induced emission photosensitizer (AIE PS) and chemodrug for deep tumor penetration to realize effective image-guided photo-chemotherapy. A newly designed AIE PS is loaded inside an iron(III) carboxylate-based MOF, MIL-100, to produce PS@MIL-100, which is encapsulated by doxorubicin (Dox) conjugated poly(ethylene glycol) methyl ether (PEG) to yield Dox-PEG-PS@MIL nanoparticles (NPs) with a diameter of 120 nm. After Dox-PEG-PS@MIL NPs reached the tumor site, intratumoral H2O2 can cause the release of the loaded PS at the tumor surface for activatable photodynamic therapy (PDT). The Dox-PEG segment is simultaneously triggered to self-assemble into ultrasmall Dox NPs. Under light irradiation, PDT is activated at the tumor surface, synergistically enhancing the tumor penetration of Dox NPs along with their ultrasmall size. After endocytosis of Dox NPs, free Dox is released from Dox NPs under low pH to enter cell nuclei for effective chemotherapy. Accompanied by bright far-red/near-infrared emission from the PS, image-guided photo-chemotherapy with enhanced efficacy is achieved.

Chitosan nanoparticles based on their derivatives as antioxidant and antibacterial additives for active bioplastic packaging

Chitosan nanoparticles (CSNPs) based on their different derivatives were proposed as antioxidant and antimicrobial additives for active bioplastic packaging. Chitosan was modified with polyethylene glycol methyl ether methacrylate (PEGMA), stearyl methacrylate (SMA) and deoxycholic acid (DC) using radiation-induced graft polymerization and chemical conjugation. The CSNPs-DC derivative has superior antioxidant activity and the CSNPs-g-pSMA derivative exhibited outstanding antibacterial activity against growth of E.coli (95.33 %). All modified CSNPs showed their capacities to inhibit S.aureus bacterial growth (>98 %). PLA packaging films containing CSNPs-g-pSMA inhibited the growth of natural microorganism on bread slices. Different chemical functions of the CSNPs derivatives provided different gas permeability and mechanical properties of the PLA films. The CSNPs derivatives would be promising antioxidant and antimicrobial additives for bioplastics to be further used as bio-based active food packaging.

Their enhanced performances are attributed to excellent microphase separations caused by block copolymer structures and multiple sulfonated side-chains integrating both rigid aromatic and flexible aliphatic units. Meanwhile, crosslinking further improves the balance among the properties of the membranes.
