A transcriptomic view to wounding response in young Scots pine stems

We studied the stress response of five-year-old Scots pine xylem to mechanical wounding using RNA sequencing. In general, we observed a bimodal response in pine xylem after wounding. Transcripts associated with water deficit stress, defence, and cell wall modification were induced at the earliest time point of three hours; at the same time, growth-related processes were down-regulated. A second temporal wave was triggered either at the middle and/or at the late time points (one and four days). Secondary metabolism, such as stilbene and lignan biosynthesis started one day after wounding. Synthesis and characteristics of novel TPA-containing electrochromic poly(ether sulfone)s with dimethylamino substituents

The PESs were readily soluble in commonly used laboratory organic solvents and could be solution-cast into tough and amorphous films with moderate levels of glass-transition temperature around 220 degrees C and thermal stability without significant weight loss up to 400 degrees C under nitrogen or air atmosphere. The Nernst equation method was used to explore the number of electrons transferred at each oxidation step of the targeted two monomers. Improving the enantioselectivity of halohydrin dehalogenase for the synthesis of (R)-benzyl glycidyl ether via biocatalytic azidolysis

Halohydrin dehalogenases (HHDHs) are valuable biocatalysts for the synthesis of enantiopure benzyl glycidyl ether (BGE) and its derivatives, which are important synthetic intermediates for anti-cancer and anti-obesity drugs. However, all the reported HHDHs exhibit low enantioselectivity. In this study, we screened site-saturation mutagenesis libraries of AbHHDH at positions R89, A136, V137, P178, N179, F180, I181, Y186 and F187 for mutants with enhanced enantioselectivity toward BGE. The four improved variant R89V, R89Y, R89K and V137I were identified, and the double mutant R89Y/V1371 showed 2.9-fold higher enantioselectivity than the wild type. The regions of HHDH containing the identified mutations were analyzed by homology modeling to explain the changes of enantioselectivity. Kinetic resolution of 20 to 100 mM BGE using whole cells of Escherichia coli expressing the mutant R89Y/V137I resulted in (R)-BGE yields of 42 to 32.5%, with cc >99%. This study improves our understanding of the enantioselectivity of HHDHs and contributes improved biocatalysts for the kinetic resolution of BGE. (C) 2020 Elsevier B.V. All rights reserved.

Low-Viscosity Ether-Functionalized Ionic Liquids as Solvents for the Enhancement of Lignocellulosic Biomass Dissolution

Due to the substantial usage of fossil fuels, the utilization of lignocellulosic biomass as renewable sources for fuels and chemical production has been widely explored. The dissolution of lignocellulosic biomass in proper solvents is vital prior to the extraction of its important constituents, and ionic liquids (ILs) have been found to be efficient solvents for biomass dissolution. However, the high viscosity of ILs limits the dissolution process. Therefore, with the aim to enhance the dissolution of lignocellulosic biomass, a series of new ether-functionalized ILs with low viscosity values were synthesized and characterized. Their properties, such as density, viscosity and thermal stability, were analyzed and discussed in comparison with a common commercial IL, namely 1-butyl-3-methylimidazolium chloride (BMIMCl). The presence of the ether group in the new ILs reduces the viscosity of the ILs to some appreciable extent in comparison to BMIMCl. 1-2(methoxyethyl)-3-methylimidazolium chloride (MOE-MImCl), which possesses the lowest viscosity value among the other ether-functionalized ILs, demonstrates an ability to be a powerful solvent in the application of biomass dissolution via the sonication method. In addition, an optimization study employing response surface methodology (RSM) was carried out in order to obtain the optimum conditions for maximum dissolution of biomass in the solvents. Results suggested that the maximum biomass dissolution can be achieved by using 3 weight% of initial biomass loading with 40% amplitude of sonication at 32.23 min of sonication period.

Extraction of Selected Metal Ions with Mixtures of N,N,N’,N’-tetra-n-octyldiglycolamide and 4,4 ‘(5 ‘)-di-t-butylcyclohexano 18-crown-6

Previous work has shown that solvent extraction (SX) and extraction chromatography (EXC) systems containing mixtures of extractants can be utilized to impart selectivity and improved physical characteristics that were not achievable with a single extractant. In an effort to discover EXC materials with unique selectivity and improved physical properties, we studied the extraction of selected metal ions from nitric acid and hydrochloric acid by EXC resins prepared from mixtures of N,N,N’,N’-tetra-n-octyldiglycolamide (TODGA) and 4,4 ‘(5 ‘)-di-t-butylcyclohexano 18-crown-6 (dtBCH18C6). Except for enhanced extraction of Ra and Ba from dilute HNO3, the EXC resins essentially exhibited the metal ion extraction properties expected from materials prepared with the individual extractants. The origin of the Ra/Ba enhancement was studied by solvent extraction variation studies in 1-octanol and n-dodecane and on extraction chromatography resins, indicating that the enhancement may not be a true synergistic interaction between the extractants, but a phase modification by the TODGA related to the co-extraction of HNO3 and water. Additionally, the influence of alkyl substituents on the DGA extractant and potential applications of the mixed extractant system in analytical chemistry and nuclear medicine were explored with extraction chromatography resins prepared from the extractant mixtures.

Extractive removal of micro and trace nitrofen, 2, 4-dichlorophenol and p-nitrotrophenol from water/soil by humic acid ester ether

The amphiphilic humic acid ester ether (HAEE), as a kind of solid-phase extractant with characteristics of easy separation and hydrophilic-hydrophobic amphiphilic property, was prepared and used to extract micro or trace nitrofen, 2,4-dichlorophenol and p-nitrotrophenol (NIPs) from water and soil. Degradation of NIPs and extractant regeneration were carried out by simple photocatalysis. The adsorption equilibrium of the mono- or three mixed NIPs by HAEE in aqueous could be quickly reached within 20 min. The adsorption process was fit to quasi-second-order kinetics model and Friendlich thermodynamics model. The possible adsorption interaction was discussed. Results suggested that the adsorption of NIPs onto HAEE predominated by hydrogen bonding, hydrophobic interaction and pi-pi interaction. The extraction capacity of mixed NIPs (80 mu g/L each component) by HAEE was up to 0.38 mg/g and tended to be multi-layer adsorption, in which p-nitrotrophenol had higher adsorption competitiveness because of lower resistance to HAEE. When HAEE-NIPs were degraded by photo-catalyst Fe-0/F-TiO2 for 8 h, not only the adsorbed NIPs could be totally degraded and mineralized, but also the HAEE could be effectively regenerated. When the NIPs were continuously extracted from 40-year aging soil for three times (regenerative twice) by combined extractant (48 mL H2O + 2 mL n-hexane + 0.1 g HAEE), the total extraction efficiency of NIPs could reach to 84.66%. This research could supplement the theory and technique for harmless treatment of NIPs contaminated water and soil.

Polybrominated diphenyl ethers exert genotoxic effects in pantropic spotted dolphin fibroblast cell lines

Cetaceans accumulate persistent and toxic substances such as polybrominated diphenyl ethers in their tissue. PBDEs are ubiquitous in marine environments, and their exposure to mammals is linked to numerous health effects such as endocrine disruption, neurotoxicity, carcinogenicity, and fetal toxicity. However, the toxicological effects and mechanism of toxicity in cetaceans remains poorly understood. The effects of BDE-47 (0.1-0.5 mu g mL(-1)), BDE-100 (0.1-0.5 mu g mL(-1)), and BDE-209 (0.25-1.0 mu g mL(-1)) exposure on cell viability, oxidative stress, mitochondrial structure, and apoptosis were evaluated using a recently established pantropical spotted dolphin (Stenella attenuata) skin fibroblast cell line (PSD-LWHT) as an in vitro model. However, the production of reactive oxygen species (ROS) increased following exposure to 1.0 mu g mL(-1) PBDE while superoxide anion, hydroxyl radicals, and inducible nitric oxide increased in a dose-dependent manner. At 0.5-1.0 mu g mL(-1), PBDEs significantly reduced the mitochondrial membrane potential. In addition, exposure to BDE-47 and -209 significantly affected mitochondrial structure as well as cell signaling and transduction compared to BDE-100. Although PBDE exposure did not affect cell viability, a significant increase in cell apoptosis markers (Bcl2 and caspase-9) was observed. This study demonstrated that BDE-47, -100, and -209 congeners might cause cytotoxic and genotoxic effects as they play a crucial role in the dysregulation of oxidative stress and alteration of mitochondrial and cell membrane structure and activity in the fibroblast cells. Hence, these results suggest that PBDEs might have adverse health effects on cetaceans inhabiting contaminated marine environments. (C) 2020 Elsevier Ltd. All rights reserved.

Synthesis of three kinds of multi-armed cardanol-based acrylic resins based on different routes for UV/EB-cured coatings

In this work, we describe three kinds of multi-armed cardanol-based acrylic resins to fabricate UV/EB-cured coatings. Hydroxyethyl cardanol ether reacts with methyl etherified melamine formaldehyde resin to synthesize multi-arm cardanol-based oligomer (HF). HF was used to synthesize multi-arm cardanol-based epoxy acrylate (AEHF), cardanol acrylate (AHF), and cardanol-based polyurethane acrylate (ISHF) based on three different routes. The three synthetic routes were based on homogeneous catalytic system by peroxyacids through epoxidation and acrylate reactions, electrophilic addition reaction, and mercapto-olefin photo-click reaction. The structure and properties of AEHF, AHF, and ISHF were characterized by FR-IR, H-1-NMR and GPC. The three kinds of cardanol-based acrylic resins were used to prepare UV/EB-cured coatings, and the performance of the coatings was tested systematically. The hardness of AHF and ISHF coating has a high pencil hardness (3H), both adhesion of the three coatings can be improve to 0 grade after addition of phosphate resin. Compared with UV curing, EBcured was not require photo-initiators, the storage modulus and the pencil hardness of the coatings were higher. The three kinds of cardanol-based acrylic resins can be applied in UV/EB-cured coatings for metal protection and other fields.

Dysregulated Lipid Metabolism Precedes Onset of Psychosis

BACKGROUND: A key clinical challenge in the management of individuals at clinical high risk for psychosis (CHR) is that it is difficult to predict their future clinical outcomes. Here, we investigated if the levels of circulating molecular lipids are related to adverse clinical outcomes in this group. METHODS: Serum lipidomic analysis was performed in 263 CHR individuals and 51 healthy control subjects, who were then clinically monitored for up to 5 years. Machine learning was used to identify lipid profiles that discriminated between CHR and control subjects, and between subgroups of CHR subjects with distinct clinical outcomes. RESULTS: At baseline, compared with control subjects, CHR subjects (independent of outcome) had higher levels of triacylglycerols with a low acyl carbon number and a double bond count, as well as higher levels of lipids in general. CHR subjects who subsequently developed psychosis (n = 50) were distinguished from those that did not (n = 213) on the basis of lipid profile at baseline using a model with an area under the receiver operating curve of 0.81 (95% confidence interval = 0.69-0.93). CHR subjects who became psychotic had lower levels of ether phospholipids than CHR individuals who did not (p < .01). CONCLUSIONS: Collectively, these data suggest that lipidomic abnormalities predate the onset of psychosis and that blood lipidomic measures may be useful in predicting which CHR individuals are most likely to develop psychosis. Physiological Responses and Phytotoxicities of Lythrum salicaria to Decabromodiphenyl Ether (BDE-209)

Decabromodiphenyl ether (BDE-209), a member of a major group of brominated flame retardants, is detected in aquatic environments at considerable levels and induces physiological and toxic effects on aquatic plants. In this study, the physiological responses induced by and the toxic effects of BDE-209 at different concentrations (0, 0.2, 0.5 and 1.0 mg L-1) in Lythrum salicaria were examined. OJIP transient curves indicated that BDE-209 treatment negatively affected photosystem II (PSII) grouping. Additionally, the results showed that BDE-209 inhibited seedling development and elevated reactive oxygen species (ROS), phosphorylated histone H2AX (gamma-H2AX), malondialdehyde (MDA) levels and antioxidative enzyme activities in the roots and shoots of L. salicaria. The results revealed that BDE-209 exposure contributed to ROS accumulation, which was considered as the probable toxicity mechanism. The current results provided an insight into the development of L. salicaria with high BDE-209 tolerance.
