Effects of Soil-Transmitted Helminths and Intestinal Protozoan Infections on Haemoglobin Levels among School-Aged Children in Belo and Bui, North West Cameroon: A Cross-Sectional Study

Introduction. Soil-transmitted helminths (STH) remain a public health problem worldwide especially in low-income countries. Soil-transmitted helminths are known to affect both growth and haemoglobin levels. In this study, we determine the prevalence of intestinal parasites and explored its association with anaemia in school-aged children (SAC). Methodology. A cross-sectional study was carried out amongst pupils selected from 18 primary schools in four communities of the North West Region. Stool samples were examined using direct wet mount and formol-ether concentration techniques, while finger prick was performed for the measurement of haemoglobin with a haemoglobinometer. Results. The results showed significant variation of STHs, intestinal protozoan prevalence, and anaemia between age, sex, and communities. The prevalence of Ascaris lumbricoides, Trichuris trichiura, and hookworms was 15.1%, 0.8%, and 2%, respectively, with an overall STH prevalence of 18.3% across the four communities. Soil-transmitted helminths were more prevalent in Mbessa (30.2%) in comparison to Nkor (26.7%), Djottin (17.5%), and Kumbo (9.6%) communities. The prevalence of Entamoeba histolytica/dispar was 25.4% and Giardia duodenalis (6.9%), with an overall intestinal protozoa prevalence of 31% across the four communities. The prevalence of anaemia in the study population was 23.1%. Conclusion. Soil-transmitted helminths, intestinal protozoan infections, and anaemia are prevalent in the study area. Anaemia was observed to be associated with STH, intestinal protozoa, and the communities under study. This study recommends continuous deworming and health education on personal sanitation and environmental sanitation.

(15-crown-5)BiI3 as a Building Block for Halogen Bonded Supramolecular Aggregates

We present the synthesis and characterization of (15-crown-5)BiI3 (1) and (15-crown-5)BiI3 center dot 0.5TIE (2), a halogen bonded adduct with tetraiodoethylene (TIE), a typical halogen bond donor. Single crystal structure analysis of 2 suggests a halogen bonding interaction between crown ether complex and TIE. The two compounds’ thermal, optical and vibrational properties are investigated in comparison, with differences pointing towards a notable interaction between the two building blocks in 2. Our results show that crown ether complexes of main group metal halides can be employed as halogen bond acceptors for the synthesis of new supramolecular aggregates.

Decabromodiphenyl ether disturbs hepatic glycolipid metabolism by regulating the PI3K/AKT/GLUT4 and mTOR/PPAR gamma/RXR alpha pathway in mice and L02 cells

Decabromodiphenyl ether (BDE-209) is a persistent environmental pollutant that poses great risks to human health and has been associated with glucose and lipid metabolism. However, the mechanisms by which BDE-209 disturbs glycolipid metabolism in the liver remain unclear. Therefore, this study sought to confirm the effects of BDE-209 on glycolipid metabolism in mice livers and L02 cells to elucidate potential mechanisms of action. In vivo BDE-209 exposure caused histological damage and lipid accumulation, elevated glucose, low-density lipoprotein, total cholesterol, and triglyceride levels, and decreased glycogen and high-density lipoprotein levels in mice livers. Moreover, in vitro BDE-209 exposure not only induced L02 cells cytotoxicity (i.e., reduced cell viability and increased LDH leakage and ROS generation) but also increased glucose and triglyceride concentrations in L02 cells. Furthermore, IdF-1, an activator of the PI3K-AKT pathway, markedly inhibited BDE-209-incluced glucose concentration increase in L02 cells and antagonized the inhibitory effect of BDE-209 on the PI3K/AKT/GLUT4 pathway by counteracting the changes in the expression levels of p-IRS, AKT, Pl3K, p-AKT, and GLUT4. Moreover, GW9662, a PPAR gamma inhibitor, blocked lipid accumulation and the upregulation of the mTOR/PPAR gamma/RXRa. pathway in L02 cells induced by BDE-209 by relieving the increases in p-mTOR, PPAR gamma, and RXR alpha protein expression levels. In summary, this study revealed that BDE-209 disrupted glycol ipid metabolism by inhibiting the Pl3K/AKT/GLUT4 pathway and activating the mTOR/PPAR gamma/RXR alpha pathway. (C) 2020 Elsevier B.V. Discovery of degradable niclosamide derivatives able to specially inhibit small cell lung cancer (SCLC)

Small cell lung cancer (SCLC) is exceedingly tough to treat and easy to develop resistance upon long use of the first-line drug carboplatin or radiotherapy. Novel medicines effective and specific against SCLC are greatly needed. Herein, we focused on the discovery of such a medicine by exploring a drug niclosamide with repurposing strategy. Initial screening efforts revealed that niclosamide, an anthelmintic drug, possessed the in vitro anticancer activity and an obvious sensitivity towards SCLC. This observation inspired the evaluation for two different kinds of niclosamide derivatives. 2 with a degradable ester as a linker exhibited the comparable activity but slightly inferior selectivity to SCLC, by contrast, the cytotoxicities of 4 and 5 with non-degradable ether linkages completely disappeared, clearly validating the importance of 2-free hydroxyl group or 2-hydroxyl group released in the antitumor activity. Mechanism study unfolded that, similar to niclosamide, 2 inhibited growth of cancer cells via p 53 activation and subsequent underwent cytochrome c dependent apoptosis. Further structural modification to afford phosphate sodium 8 with significantly enhanced aqueous solubility (22.1 mg/mL) and a good selectivity towards SCLC demonstrated more promising druggability profiles. Accordingly, niclosamide as an attractive lead hold a huge potential for developing targeted anti-SCLC drugs.

Infant total diet study in France: Exposure to substances migrating from food contact materials

A total diet study (TDS) was conducted in France to assess the health risks related to the chemicals in food of non-breastfed children under three years of age (Infant TDS). For the first time, substances coming from food contact materials, such as bisphenol A (BPA), bisphenol A diglycidyl ether (BADGE) and its derivatives, some phthalates, and some ink photoinitiators, were targeted because of growing interest in these substances. Food samples were collected to be representative of the whole diet of non-breastfed children aged 1-36 months, and prepared as consumed prior to analysis. Dietary exposure was assessed for 705 representative children under three years of age. Generally, the substances from food contact materials were detected in few samples: 38% for BPA, 0% for BADGE and its derivatives, 0-35% for phthalates, 1.9% for benzophenone, and 0% for the other ink photo initiators. Regarding exposure levels, the situation was deemed tolerable for BADGE and its hydrolysis products, di-isodecyl phthalate, dibutyl phthalate, butyl benzyl phthalate, bis(2-ethylhexyl) phthalate, and di-isononyl phthalate, benzophenone, and 4-methylbenzophenone. Only for BPA, the exposure levels of some children exceeded the lowest toxicological value established by the French Agency for Food, Environmental and Occupational Health & Safety at 0.083 mu g.kg bw(-1).d(-1). The temporary tolerable daily intake of the European Food Safety Authority (EFSA), set at 4 mu g.kg bw(-1).d(-1), was never exceeded. However, actual exposure to BPA was probably overestimated, as well as the associated risk, because the foods were sampled prior to the recent regulations banning BPA in food packaging. This study is the first worldwide to provide an estimate of infant food contamination levels and exposures of children under 3 years of age, based on a TDS approach. It therefore provides key data on the exposure of this particularly sensitive population to substances released from food contact materials, and presents useful data for studies evaluating exposure to mixtures or aggregated exposure.

Flame-responsive aryl ether nitrile structure towards multiple fire hazards suppression of thermoplastic polyester

Multiple fire hazards (heat, smoke, dripping) caused by thermoplastic polymers pose integrated risks. Halogen or phosphorus flame-retardants tend to increase toxic, smoke or dripping hazards due to their flame-retardant mechanism. The physical blending flame-retardants into matrixes also presents a migration dilemma with causing potential environmental threats. Herein, we propose a novel multi-hazards inhibition strategy by chemical-incorporating aryl ether nitrile structures into poly(ethylene terephthalate)(PET), which is a typical thermoplastic polymer and a major contributor of multiple fire hazards. Through flame-responsive cyclotrimerization and aliphatic fragment capture, the flammability risks and multi-hazards (heat, smoke, toxicity, dripping) are significantly suppressed. The limiting oxygen index of the modified PET increases from 21.0 to 31.0. The peak of heat release, total smoke release, and carbon monoxide production decrease by 49.0 %, 31.1 %, and 52.6 %, respectively. The dripping hazards are eliminated, and the UL-94 rating reaches to V-0 level with no dripping production. Hence, this state-of-art strategy supplies a new approach for the fire hazards suppression of thermoplastic polymers.

Photodynamic and Cold Atmospheric Plasma Combination Therapy Using Polymeric Nanoparticles for the Synergistic Treatment of Cervical Cancer

Integrating multi-modal therapies into one platform could show great promise in overcoming the drawbacks of conventional single-modal therapy and achieving improved therapeutic efficacy in cancer. In this study, we prepared pheophorbide a (Pheo a)/targeting ligand (epitope analog of oncoprotein E7, EAE7)-conjugated poly(gamma-glutamic acid) (gamma-PGA)/poly(lactide-co-glycolide)-block-poly(ethylene glycol) methyl ether (MPEG-PLGA)/hyaluronic acid (PPHE) polymeric nanoparticles via self-assembly and encapsulation method for the photodynamic therapy (PDT)/cold atmospheric plasma (CAP) combinatory treatment of human papillomavirus (HPV)-positive cervical cancer, thereby enhancing the therapeutic efficacy. The synthesized PPHE polymeric nanoparticles exhibited a quasi-spherical shape with an average diameter of 80.5 +/- 17.6 nm in an aqueous solution. The results from the in vitro PDT efficacy assays demonstrated that PPHE has a superior PDT activity on CaSki cells due to the enhanced targeting ability. In addition, the PDT/CAP combinatory treatment more effectively inhibited the growth of cervical cancer cells by causing elevated intracellular reactive oxygen species generation and apoptotic cell death. Moreover, the three-dimensional cell culture model clearly confirmed the synergistic therapeutic efficacy of the PDT and the CAP combination therapy using PPHE on CaSki cells. Overall, these results indicate that the PDT/CAP combinatory treatment using PPHE is a highly effective new therapeutic modality for cervical cancer.

Raw glycerol re-valuing through etherification with isobutylene: process design and techno-economical assessment

This study evaluates the techno-economic feasibility to produce tert-butyl ethers of glycerol in Colombia using glycerol obtained from biodiesel production. The major drawback of this process is related to the high costs associated to the isobutylene reagent used for this reaction. Three possible scenarios, using isobutylene: glycerol molar ratios of 2:1 and 4:1, have been evaluated by simulation and techno-economical assessment using pure isobutylene, C-4 fraction and a mixture of C-4 and isobutylene as well as a refining stage for crude glycerol. The desired product, a mixture of di- and tri- ethers of glycerol (h-GTBE) was obtained in high amounts for the three scenarios (92-99 wt.%) and a direct relation between production cost and raw material cost was established. The prices for h-GTBE were higher than the proposed selling prices; however, the formation of byproduct (C-4 fraction) can make the process competitive. h-GTBE production did not have a competitive price for any scenario at high isobutylene: glycerol molar ratio. The production price was lowered by around 20% for all scenarios when crude glycerol was considered. Finally, competitive price of h-GTBE (0.96 $/kg) and NPV positive was obtained using a mixture of C-4 and isobutylene and crude glycerol. Improvement of the wound healing properties of hydrogels with N-acetylcysteine through their modification with methacrylate-containing polymers

Hydrogels with antioxidant activity have shown to significantly improve the standard of care, because they promote efficient wound healing, i.e. regeneration. N-Acetylcysteine (NAC) is an antioxidant amino acid derivative that promotes complete tissue restoration. However, NAC has anticoagulant properties that may also hinder blood coagulation, which is crucial for hydrogels for wound healing applications. To take advantage of the regenerative activity of NAC while avoiding hampering the hemostasis stage during wound healing, we modified gelatin-NAC with the methacrylate-containing polymers 2-hydroxyethyl methacrylate (H) and poly(ethylene glycol) methyl ether methacrylate (P) to produce Gel-HP-NAC. These hydrogels clotted more blood and faster than Gel and Gel-NAC hydrogels, while maintaining fluid absorption properties adequate to promote wound healing. Similarly, there were more viable human skin fibroblasts after 10 days cultured in Gel-HP-NAC compared with Gel and Gel-NAC. A mouse full-thickness skin wound model demonstrated that Gel-HP-NAC hydrogels improved the wound healing process as compared to the untreated group as proved by the increased wound closure rates and re-epithelialization. Histology of the biopsied tissues indicated more organized collagen deposits on the wounds treated with either Gel-HP-NAC or Gel-NAC than untreated wounds. Our results show that modification of NAC-containing hydrogels through methacrylate-containing polymers improved their wound healing properties, including blood-clotting, and demonstrate the potential of Gel-HP-NAC hydrogels for wound treatment and tissue regeneration.

Amphiphilic micelle-forming PDMS-PEGBEM comb copolymer self-assembly to tailor the interlamellar nanospaces of defective poly (ethylene oxide) membranes

An amphiphilic comb copolymer was synthesized via free-radical polymerization using two macromonomers: monomethacryloxypropyl-terminated poly(dimethyl siloxane) (PDMS-MA, 10,000 g mol(-1)) and poly(ethylene glycol)-behenyl ether methacrylate (PEGBEM, 1500 g mol(-1)). The PDMS-PEGBEM comb copolymer underwent self-assembly to form micellar structures in ethanol/tetrahydrofuran solvent. The micelle-forming PDMS-PEGBEM copolymer was incorporated into the low-cost, commercially available polymer, and non-selective polymer poly(ethylene oxide) (PEO, 106 g mol-1), resulting in highly selective all-polymeric membranes. The alignment of the PDMS-PEGBEM micelles in the interlamellar region of PEO and the corresponding increase in the domain spacing were observed by transmission electron microscopy and small-angle X-ray scattering, confirming the successful encapsulation of PDMS-PEGBEM in the interlamellar region. The PDMS functions as a rubbery filler between the crystallites, whereas PEGBEM has good affinity towards PEO because of their chemical similarity. The CO2/N-2 and CO2/CH4 selectivities of PEO membranes was significantly enhanced from 0.78 and 0.71 to 41.7 and 13.3, respectively, on the loading of 10 wt.% of the copolymer. The highest CO2 permeability reached 240 barrer at 50 wt.% copolymer loading, with the CO2/N-2 and CO2/CH4 selectivities of 34.6 and 12.2, respectively. Herein, the evolution of the interlamellar nanospaces and dual-phase region, as well as the microstructures of the membranes are reported.
