Magnetic biopolymer hybrid nanoparticles have gained a lot of attention in the field of biomedical applications. Here we present the synthesis of hyaluronic acid (HA)-coated magnetic clusters of superparamagnetic nanoparticles. Firstly, the superparamagnetic magnetite nanoparticles coated with oleic acid were synthesized and characterized in terms of size, composition and magnetic properties. Secondly, magnetic nanoclusters were prepared and wrapped with HA grafted with an ethylene glycol-based copolymer. The resulting nanostructures and nanobeads were characterized using electron microscopy, dynamic light scattering, thermogravimetric analysis and magnetic measurements. The clustering of magnetite nanoparticles facilitates magnetic purification, separation, rapid attraction and controlled positioning by high field gradient micro-magnet arrays. In addition, clusters could be formed in the presence of a fluorescent drug model, which demonstrates the possibility of dual functionalization of our hybrid nanosystems: magnetic responsiveness and drug encapsulation.

Composite worm-like micelles (CWMs) that encapsulate a large number of superparamagnetic magnetite nanoparticles (SMNPs) within the core while still having a small width should have a high magnetic resonance sensitivity and simultaneously a desirable circulation behaviour in biological systems. However, the preparation of such CWMs is still a challenge. Herein we report the fabrication of CWMsviatwo-stage self-assembly of SMNPs guided by a block copolymer and DNA, respectively. First, poly(ethylene glycol)-b-poly(4-vinylpyridine) co-assembled with SMNPs to form spherical composite micelles, guiding the clustering of multiple SMNPs within the core of the micelles. Then, at the second stage, DNA co-assembled with and guided the linear aggregation and fusion of the micelles, resulting in the CWM encapsulating a large number of SMNPs in the linear core. As a proof of concept, CWMs with aT(2)relaxivity as high as 704 mM(-1)s(-1)and a low MRI detection limit of 2.2 mu g mL(-1)(similar to 0.02 nM CWMs) were prepared; it is significant that theT(2)relaxivity of the SMNPs assembled in the CWMs is around 20 times that of the same SMNPs in the same amount but individually dispersed in the suspension. Additionally, although heavily loaded with SMNPs, the CWMs have a width as small as 20.5 nm and a core-shell structure with PEG as the shell. These features should make the CWMs very promising in their use as ultra-sensitive magnetic resonance imaging probes.

A quinoxaline-based porous organic polymer (Q-POP) as a mesoporous organic copolymer was developed as a new platform for the immobilization of CuNPs and copper nanocrystals. The prepared materials were characterized by FT-IR, XRD, N-2 adsorption-desorption isotherms, ICP, TGA, SEM, HR-TEM, EDX, and single-crystal X-ray crystallography. The obtained catalyst presented extraordinary catalytic activity towards Ullmann C-O coupling reactions with high surface area, hierarchical porosity, and excellent thermal and chemical stability. Due to its high porosity, and synergistic effect of copper nanocrystals incorporated in the polymer composite, the as-synthesized catalyst was successfully utilized for the Ullmann C-O coupling reaction of phenols and different aryl halides to prepare various diaryl ether derivatives. All types of aryl halides (except aryl fluorides) were screened in the Ullmann C-O coupling reaction with phenols to produce diaryl ethers in good to excellent yields (70-97 %), and it was found that aryl iodides have the best results. Besides, due to the strong interactions between CuNPs, N, and O-atoms of quinoxaline moiety existing in the polymeric framework, the copper leaching from the support was not observed. Furthermore, the catalyst was recycled and reused for five consecutive runs without significant activity loss.

Background: Neurotoxic effects of Polybrominated Diphenyl Ethers (PBDEs) at low levels have not been well studied in human population, and whether the associations can be explained by thyroid hormones (THs) remains unclear. Objectives: We examined the associations of prenatal PBDE exposures with THs in cord plasma and neurobehavior of children at 2 and 4 years among general population in China. Methods: Participants were mother-child pairs in the Shanghai-Minhang Birth Cohort Study. Nine PBDE congeners and THs (thyroid stimulating hormone, total thyroxine, free thyroxine, total triiodothyronine, and free triiodothyronine) were determined in cord plasma. Child Behavior Checklist (CBCL/1.5-5) were completed by caregivers to assess children’s neurobehavioral development at 2 and 4 years. In the final analyses, 199 and 307 mother-child pairs at 2 and 4 years were included to examine associations of PBDEs with CBCL scores using Pearson-scale-adjusted Poisson regressions, and 339 subjects were included in linear regression models to investigate the associations between PBDEs and THs. Results: BDE-47 had the highest detection rate of 83.82% with the median concentration of 0.19 ng/g lipid, followed by BDE-28, -99, -100 and -153 with detection rates nearly 50%. We found positive associations between prenatal PBDE concentrations and children’s neurobehavior, including Somatic Complaints, Withdrawn, Sleep Problems and Internalizing Problems in girls, and Somatic Complaints and Attention Problems in boys. We also observed inverse associations of the sum of BDE-47, – 28, – 99, -100 and -153 with THs. However, by adding THs to the models examining associations between PBDEs and CBCL, the main results didn’t measurably change. Conclusions: This study adds new knowledge that prenatal PBDEs at low levels may be related to long-lasting behavioral abnormalities in children and reduced THs in cord plasma. However, the hypothesis that the neurotoxic impact of PBDEs may be explained by alterations in cord THs was not supported.

We report on the shape, composition (from Pt95Zn5 to Pt77Zn23), and surface chemistry of Pt-Zn nanoparticles obtained by reduction of precursors M2+(acac) (-)(2) (M2+: Pt2+ and Zn2+) in oleylamine, which serves as both solvent and ligand. We show first that the addition of phenyl ether or benzyl ether determines the composition and shape of the nanoparticles, which point to an adsorbate-controlled synthesis. The organic (ligand)/inorganic (nanoparticles) interface is characterized on the structural and chemical level. We observe that the particles, after washing with ethanol, are coated with oleylamine and the oxidation products of the latter, namely, an aldimine and a nitrile. After exposure to air, the particles oxidize, covering themselves with a few monolayer thick ZnO film, which is certainly discontinuous when the particles are low in zinc. Pt-Zn particles are unstable and prone to losing Zn. We have strong indications that the driving force is the preferential oxidation of the less noble metal. Finally, we show that adsorption of CO on the surface of nanoparticles modifies the oxidation state of amine ligands and attribute it to the displacement of hydrogen adsorbed on Pt. All the structural and chemical information provided by the combination of electron microscopy and X-ray photoelectron spectroscopy allows us to give a fairly accurate picture of the surface of nanoparticles and to better understand why Pt-Zn alloys are efficient in certain electrocatalytic reactions such as the oxidation of methanol.

Addition of DMSO is found to greatly accelerate the aqueous hydrolysis of organohalides to alcohols, providing a neutral, more efficient, milder and more economic process. Mechanistic studies using O-18-DMSO and O-18-H2O showed that, contrary to the opinion that DMSO works as a dipolar solvent to enhance water’s nucleophilicity, the accelerating effect comes from a complete oxygen transfer from DMSO to organohalides through generation of ROS+Me2.X- salts through C-O bond formation, followed by O-S bond disassociative hydrolysis of ROS+Me2.X- with water. This method is applicable to a wide range of organohalides and thus may have potential for practical industrial application, owing to easy recovery of DMSO from the H2O/DMSO mixture by regular vacuum rectification.

Short-chain oxymethylene dimethyl ethers (OMEs) (molecular formula: H3CO-(CH2O)(n)-CH3, where n = 3-5) have previously been studied as diesel-like fuels and fuel additives. OMEs can be produced from sustainably sourced methanol, and tests indicate that they are neither genotoxic nor mutagenic. In this report, their potential as solvents has been investigated to expand the bio-derived solvent space. According to traditional solvatochromic parameters, a commercial mixture of OME3-5 and its individual components (OME3, OME4, and OME5) have solvation properties similar to problematic industrial ether solvents such as 1,4-dioxane. Peroxide formation, one of the chief dangers of classical ether solvents, was found to occur much more slowly in OMEs than in conventional solvents such as tetrahydrofuran (THE), demonstrating an improved safety profile. The commercial OME3-5 mixture was found to be broadly miscible with organic solvents but immiscible with water, suggesting potential application in aqueous extractions. It performed well in the dissolution of polystyrene and removal of paints and coatings, indicating OME3-5 may suitable to replace dichloromethane in polymer recycling, polymer welding, and cleaning applications. To further demonstrate applicability as a solvent, this mixture was shown to facilitate a model Suzuki coupling reaction at rates similar to cyclopentyl methyl ether, which is currently marketed as a green ether. Finally, OME3-5 proved a suitable solvent for enzymatic polymerization, giving high yields, moderately high degrees of polymerization, and remarkably narrow dispersity values.

Various optical clearing methods have emerged as powerful tools for deep biological imaging. Organic solvent-based clearing methods, such as three-dimensional imaging of solvent-cleared organs (3DISCO), present the advantages of high clearing efficiency and size reduction for panoptic imaging of large samples such as whole organs and even whole bodies. However, 3DISCO results in a rapid quenching of endogenous fluorescence, which has impeded its application. Here, we propose an advanced method named FDISCO to overcome this limitation. FDISCO can effectively preserve the fluorescence of various fluorescent probes and can achieve a long storage time of months while retaining potent clearing capability. We used FDISCO for high-resolution imaging and reconstruction of neuronal and vascular networks. Moreover, FDISCO is compatible with labeling by multiple viruses and enables fine visualization of neurons with weak fluorescence labeling in the whole brain. FDISCO represents an effective alternative to the three-dimensional mapping of whole organs and can be extensively used in biomedical studies.

A mild, atom-economic, and metal-free alpha-C-H amination of ethers using relatively stable nonafluorobutanesulfonyl (nonaflyl, Nf) azide as the aminating reagent to give N-sulfonyl hemiaminals is reported. Rationale: Structural remodeling or damage as a result of disease or injury is often not evenly distributed throughout a tissue but strongly depends on localization and extent of damaging stimuli. Skeletal muscle as a mechanically active organ can express signs of local or even systemic myopathic damage, necrosis, or repair. Conventionally, muscle biopsies (patients) or whole muscles (animal models) are mechanically sliced and stained to assess structural alterations histologically. Three-dimensional tissue information can be obtained by applying deep imaging modalities, e.g. multiphoton or light-sheet microscopy. Chemical clearing approaches reduce scattering, e.g. through matching refractive tissue indices, to overcome optical penetration depth limits in thick tissues. Methods: Here, we optimized a range of different clearing protocols. We find aqueous solution-based protocols employing (20-80%) 2,2′-thiodiethanol (TDE) to be advantageous over organic solvents (dibenzyl ether, cinnamate) regarding the preservation of muscle morphology, ease-of-use, hazard level, and costs. Results: Applying TDE clearing to a mouse model of local cardiotoxin (CTX)-induced muscle necrosis, a complete loss of myosin-II signals was observed in necrotic areas with little change in fibrous collagen or autofluorescence (AF) signals. The 3D aspect of myofiber integrity could be assessed, and muscle necrosis in whole muscle was quantified locally via the ratios of detected AF, forward- and backward-scattered Second Harmonic Generation (fSHG, bSHG) signals. Conclusion: TDE optical clearing is a versatile tool to study muscle architecture in conjunction with label-free multiphoton imaging in 3D in injury/myopathy models and might also be useful in studying larger biofabricated constructs in regenerative medicine.
