A multitude of metrics exist to assign scores to synthetic routes within computer-aided synthesis planning (CASP) tools. A quantitative scoring method is necessary to identify the most promising synthetic approaches to a molecule. However, current CASP tools are limited in their capacity to evaluate reaction selectivity and are unable to fully account for the effect of side products on the purification sequences associated with chemical syntheses. We develop a novel quantitative metric called ExtractionScore for evaluating synthetic routes based on the predicted identities of side products as well as the separability of major and side products by liquid-liquid extraction based on chemical property prediction. By comparing industrially practiced routes to a collection of 200 pharmaceutically relevant compounds with routes suggested by state-of-the-art CASP software, we show that ExtractionScore may improve retrosynthetic recommendations by incorporating information about the formation of side products.

A combination of carbon ions/photons irradiation and hyperthermia as a novel therapeutic approach for the in-vitro treatment of pancreatic cancer BxPC3 cells is presented. The radiation doses used are 0-2 Gy for carbon ions and 0-7 Gy for 6 MV photons. Hyperthermia is realized via a standard heating bath, assisted by magnetic fluid hyperthermia (MFH) that utilizes magnetic nanoparticles (MNPs) exposed to an alternating magnetic field of amplitude 19.5 mTesla and frequency 109.8 kHz. Starting from 37 degrees C, the temperature is gradually increased and the sample is kept at 42 degrees C for 30 min. For MFH, MNPs with a mean diameter of 19 nm and specific absorption rate of 110 +/- 30 W/g(Fe3)o(4) coated with a biocompatible ligand to ensure stability in physiological media are used. Irradiation diminishes the clonogenic survival at an extent that depends on the radiation type, and its decrease is amplified both by the MNPs cellular uptake and the hyperthermia protocol. Significant increases in DNA double-strand breaks at 6 h are observed in samples exposed to MNP uptake, treated with 0.75 Gy carbon-ion irradiation and hyperthermia. The proposed experimental protocol, based on the combination of hadron irradiation and hyperthermia, represents a first step towards an innovative clinical option for pancreatic cancer.

Introduction: Gestational exposure to chemical mixtures, which is prevalent among pregnant women, may be associated with adverse childhood neurodevelopment. However, few studies have examined relations between gestational chemical mixture exposure and children’s cognitive abilities. Methods: In a cohort of 253 pregnant women and their children from Cincinnati, OH (enrolled 2003-2006), we quantified biomarker concentrations of 43 metals, phthalates, phenols, polybrominated diphenyl ethers, organophosphate and organochlorine pesticides, polychlorinated biphenyls, perfluoroalkyl substances, and environmental tobacco smoke in blood or urine. Using k-means clustering and principal component (PC) analysis, we characterized chemical mixtures among pregnant women. We assessed children’s cognitive abilities using the Wechsler Preschool and Primary Scale of Intelligence-III and Wechsler Intelligence Scale for ChildrenIV at ages 5 and 8 years, respectively. We estimated covariate-adjusted differences in children’s cognitive ability scores]=cross clusters, and with increasing PC scores and individual biomarker concentrations. Results: Geometric mean biomarker concentrations were generally highest, intermediate, and lowest among women in clusters 1, 2, and 3, respectively. Children born to women in clusters 1 and 2 had 5.1 (95% CI: 9.4,-0.8) and 2.0 (95% CI: 5.5, 1,4) lower performance IQ scores compared to children in cluster 3, respectively. PC scores and individual chemical biomarker concentrations were not associated with cognitive abilities. Conclusions: In this cohort, combined prenatal exposure to phenols, certain phthalates, pesticides, and perfluoroalkyl substances was inversely associated with children’s cognition, but some individual chemical biomarker concentrations were not. Additional studies should determine if the aggregate impact of these chemicals on cognition is different from their individual effects.

Bulk chemicals produced from renewable resources are receiving considerable attention due to the increase in sustainable practices. In this study, a novel, efficient, and sustainable strategy has been proposed to produce versatile petroleum based monophenols from the natural aromatic polymer, lignin, via the selective cleavage of its specific chemical linkages by a cost-effective catalyst, Ni/MgO. The results demonstrate that 93.4% of lignin is converted, yielding 15.0% monophenols in the presence of 20% Ni/MgO. Importantly, 42.3% of these volatile chemicals are found to be 4-ethyl phenol (the yield is 6.31%), a widely used fine chemical currently obtained from the petrochemical industry. Further comparative investigations on the raw and recovered lignin using heteronuclear single quantum correlation-nuclear magnetic resonance (HSQC-NMR), Fourier transform infrared (FT-IR), quantitative C-13 NMR, and the alkaline nitrobenzene oxidation method reveal that the selective breakage of the ester bond between the p-hydroxyphenyl structural unit and the other fragments followed by decarboxylation is responsible for the good performance of the 4-ethyl phenol formation process. Therefore, the results of this work would provide new insights for producing value-added petrochemicals from renewable lignin.

This study was designed to assess the effect of lactational polybrominated diphenyl ether (PBDE) exposure on early physical development of both healthy and fetal growth restriction (FGR) infants. Z scores of head circumference-for-age (ZHC), length-for-age (ZLEN), weight-for-age (ZWEI) and weight-for-length (ZWFL) were calculated according to the WHO Child Growth Standards. FGR infants had a higher PBDE exposure level and faster growth speed from 42 days to 6 months compared with healthy infants. Exposure of lower brominated BDEs (BDE28 similar to 154) and the sum of 18 BDE congeners (BDE28 similar to 209) negatively associated with ZHC [mean difference estimate (95 % CI): -0.71 (-1.22, -0.22) and -0.81 (-1.31, -0.33)] in FGR boys. Both BDE153 and BDE196 exposure had a significant correlation with ZLEN of boys in FGR group [mean difference estimate (95 % CI): -0.28 (-0.48, -0.07) and -0.52 (-0.91, -0.14)]. Each 10 ng/g lipid increase in the concentrations of BDE154 were respectively associated with 0.16 increase in ZWEI among boys in healthy group. No significant association was found in girls. Lactational PBDE exposure had effects on the early growth of both FGR and healthy infants. The encouragement of breastfeeding should therefore be considered in conjunction with PBDE contamination levels.

In this paper, we report the synthesis of Fe3O4@AMCA-MIL53(Al)-NH2-Co-II NPs based on the metal-organic framework structures as a magnetically separable and environmentally friendly heterogeneous nanocatalyst. Magnesium-stabilized amorphous calcium carbonate (Mg-ACC), amorphous magnesium calcium silicate hydrate (MCSH), and hydroxyapatite (HAp) are prepared by a precipitation method. By cold-pressing these particles, it is possible to produce porous bulk discs with a narrow pore size distribution. These porous inorganic discs (Mg-ACC, MCSH, and HAp) are investigated as stationary phases to study the chromatographic behavior and adsorption ability of rhodamine B, methylene blue, and ribonuclease. The adsorption affinities of different biomolecules can be easily observed and evaluated through this method. Furthermore, by infiltrating fabricated opaque porous discs with benzyl ether, which has a similar refractive index as the used inorganic particles (Mg-ACC, MCSH, and HAp), their optical properties significantly change and the discs become translucent. Moreover, by infiltrating the MCSH discs with a light-curing polymer, translucent composites with good surface hardness are fabricated. By doping particles with ions such as Ni2+, Co2+ , Fe3+, and Eu3+, the color and UV-visible spectrum of the bulk discs can be adjusted. Typically, by using iron-doped MCSH particles as the inorganic matrix, nanocomposites, which show a steep UV-absorption edge at 400 nm, are fabricated. Our work provides a simple and economical method to evaluate the affinity of biomolecules to inorganic materials and a novel way to fabricate translucent hard composite materials. The fabricated nanocomposite discs show a great UV shielding effect and superior surface hardness compared to polymethyl methacrylate and commercial sunglasses, suggesting their potential as new sunglass materials.

UV-curable inks, coatings, and adhesives are being increasingly used in food packaging systems. When exposed to UV energy, UV-photoinitiators (PI’s) present in the formulations produce free radicals which catalyze polymerization of monomers and pre-polymers into resins. In addition to photopolymerization, other free radical reactions occur in these systems resulting in the formation of chemically varied photolytic decomposition products, many of which are low molecular weight chemical species with high migration potential. This research conducted model experiments in which 24 commonly used PI’s were exposed to UV-energy at the typical upper limit of commercial UV-printing press conditions. UV-irradiated PI’s were analyzed by gas chromatography-mass spectrometry (GC-MS) and electrospray-mass spectrometry (ESI-MS) in order to identify photolytic decomposition products. Subsequently, migration studies of 258 UV-cure food packaging samples were conducted using GC-MS; PI’s and photolytic decomposition products were found in nearly all samples analyzed. One hundred-thirteen photolytic decomposition products were identified. Eighteen intact PI’s and 21 photolytic decomposition products were observed as migrants from the 258 samples analyzed, and these were evaluated for frequency of occurrence and migratory concentration range. The most commonly observed PI’s were 2-hydroxy-2-methylpropiophenone and benzophenone. The most commonly observed photolytic decomposition products were 2,4,6-trimethylbenzaldehyde and 1-phenyl-2-butanone. This compilation of PI photolytic decomposition data and associated migration data will aid industry in identifying and tracing non-intentionally added substances (NIAS) in food packaging materials.

A highly efficient Ni/N-C catalyst was synthesized by facile pyrolysis of a Ni-containing metal-organic framework (Ni-MOF), and its catalytic hydrogenolysis performance towards C-O bonds in lignin was evaluated in detail using diphenyl ether (DPE) as a model compound. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) analyses show that the flower-like nanosheets of the Ni-MOF shrink, forming a loose and ordered spherical structure during pyrolysis. Under the optimal conditions, DPE was completely converted and the selectivity to monomers (benzene, cyclohexanol and cyclohexane) reached 99.1%. During the catalytic hydrogenolysis conversion (CHC) of DPE, the direct cleavage of the C-aromatic-O bond affording benzene and phenol is the major reaction pathway, and a low H2 pressure is crucial to increase the monomer selectivity. Furthermore, Ni/N-C-450 shows high hydrogenolysis activity for other lignin-derived aryl ethers, such as benzyl phenyl ether, dibenzyl ether, dinaphthalene ether, benzyl 2-naphthyl ether and 3-methoxyphenol.

Pearl-farming leads to significant plastic pollution in French Polynesia (FP) as the end of life of most farming gear is currently poorly managed. Plastic debris released into the aquatic environment accumulates, with potentially detrimental effects on the lagoon ecosystem and pearl oyster Pinctada margaritifera, a species of ecological, commercial and social value. Here, we tested the effects of leachates from new (N) and aged (A) plastic pearl-farming gear (spat collector and synthetic rope) obtained after 24 h and 120 h incubation, on the embryo-larval development of the pearl oyster using an in-vitro assay. Embryos were exposed for 24 h and 48 h to a negative control (0) and the leachate from 0.1, 1, 10 and 100 g of plastic. L-1. After 24 h exposure to leachate at 100 g.L-1, effects were observed on embryo development (-38% to -60% of formed larvae) and mortality (+72% to +82%). Chemical analyses of plastic gear indicated the presence of 26 compounds, consisting of organic contaminants (PAHs) and additives (mainly phthalates). Screening of leachates demonstrated that these compounds leach into the surrounding seawater with an additional detection of pesticides. Higher levels of phthalates were measured in leachates obtained from new (6.7-9.1 mg.L-1) than from aged (0.4-0.5 mg.L-1) plastics, which could be part of the explanation of the clear difference in toxicity observed after 48 h exposure at lower concentrations (0.1-10 g.L-1), associated with mortality ranging from 26 to 86% and 17-28%, respectively. Overall, this study suggests that plastic gear used in the pearl-farming industry releases significant amounts of hazardous chemicals over their lifetime, which may affect pearl oyster development that call for in-situ exploration.
