Investigating environmental pollution is important to understand its impact on endangered species such as green turtles (Chelonia mydas). In this study, we investigated the accumulation and potential toxicity of selected persistent organic pollutants (POPs) and naturally occurring MeO-PBDEs in liver, fat, kidney and muscle of turtles (n = 30) of different gender, size, year of death, location and health status. Overall, POP concentrations were low and accumulation was highest in liver and lowest in fat which is likely due to the poor health of several animals, causing a remobilization of lipids and associated compounds. PCBs and p,p’-DDE dominated the POP profiles, and relatively high MeO-PBDE concentrations (2′-MeO-BDE 68 up to 192 ng/g lw, 6-MeO-BDE 47 up to 79 ng/g lw) were detected in all tissues. Only few influences of factors such as age, gender and location were found. While concentrations were low compared to other marine wildlife, biological toxicity equivalences obtained by screening the tissue extracts using the micro-EROD assay ranged from 2.8 to 356 pg/g and the highest values were observed in muscle, followed by kidney and liver. This emphazises that pollutant mixtures found in the turtles have the potential to cause dioxin-like effects in these animals and that dioxin-like compounds should not be overlooked in future studies. An efficient and simple approach for the direct synthesis of aryl and heteroaryl sulfonic esters was developed using DMS and DES as alkoxysulfonylation reagents. The reaction is operationally simple and scalable. This protocol does not require solvent, expensive catalysts, base, ligand additives or other reagents. A wide range of sulfonic esters were synthesized in moderate to good chemical yields. This method has the advantage of low cost, facile and tolerated a wide range of substrates.

Magnetic biopolymer hybrid nanoparticles have gained a lot of attention in the field of biomedical applications. Here we present the synthesis of hyaluronic acid (HA)-coated magnetic clusters of superparamagnetic nanoparticles. Firstly, the superparamagnetic magnetite nanoparticles coated with oleic acid were synthesized and characterized in terms of size, composition and magnetic properties. Secondly, magnetic nanoclusters were prepared and wrapped with HA grafted with an ethylene glycol-based copolymer. The resulting nanostructures and nanobeads were characterized using electron microscopy, dynamic light scattering, thermogravimetric analysis and magnetic measurements. The clustering of magnetite nanoparticles facilitates magnetic purification, separation, rapid attraction and controlled positioning by high field gradient micro-magnet arrays. In addition, clusters could be formed in the presence of a fluorescent drug model, which demonstrates the possibility of dual functionalization of our hybrid nanosystems: magnetic responsiveness and drug encapsulation.

Psoralen is a furocoumarin natural product that intercalates within DNA and forms covalent adducts when activated by ultraviolet radiation. It is well known that this property contributes to psoralen’s clinical efficacy in several disease contexts, which include vitiligo, psoriasis, graft-versus-host disease and cutaneous T-cell lymphoma. Given the therapeutic relevance of psoralen and its derivatives, we attempted to synthesize psoralens with even greater potency. In this study, we report a library of 73 novel psoralens, the largest collection of its kind. When screened for the ability to reduce cell proliferation, we identified two derivatives even more cytotoxic than 4 ‘-aminomethyl-4,5 ‘,8-trimethylpsoralen (AMT), one of the most potent psoralens identified to date. Using MALDI-TOF MS, we studied the DNA adduct formation for a subset of novel psoralens and found that in most cases enhanced DNA binding correlated well with cytotoxicity. Generally, our most potent derivatives contain positively charged substituents, which we believe increase DNA affinity and enhance psoralen intercalation. Thus, we provide a rational approach to guide efforts toward further optimizing psoralens to fully capitalize on this drug class’ therapeutic potential. Finally, the structure-activity insights we have gained shed light on several opportunities to study currently underappreciated aspects of psoralen’s mechanism.

A semi-batch reactor was used to carry out the pyrolysis of Ageratum conyzoides. The outcome of pyrolysis parameters vis temperature (350-600 degrees C); heating rate (25-100 degrees C min(-1)) and sweeping gas (N-2) flow rate (0.1-0.5 L min(-1)) were observed on the product yield. The fixed particle size of 0.425 mm was maintained throughout the pyrolysis process. The maximum pyrolytic-oil yield of 37.55 mass% was achieved at a temperature of 525 degrees C under a constant heating rate and sweeping gas flow rate of 75 degrees C min(-1) and 0.2 L min(-1). Biomass, biochar, pyrolytic-oil and pyrolysis-gas were characterized through CHNS and O, FT-IR, TG and DTG, SEM-EDX, BET, XRD, H-1-NMR and GC-MS analysers. The A. conyzoides pyrolytic-oil and biochar has an empirical formula of CH1.32O0.82 and CH0.82O0.44 and a high heating value of 17.79 MJ kg(-1) and 22.93 MJ kg(-1). Presence of lower and higher hydrocarbon compounds in pyrolytic-oil makes it a suitable feedstock for the production of various chemicals. High alkalinity and carbonaceous nature of biochar make it suitable for the modification of soil or can be utilized as a solid fuel. The pyrolysis-gas has a gross calorific value of 5.32 MJ m(-3) and can be utilized as an alternative gaseous energy source.

The use of copper for reducing nosocomial infections or healthcare-acquired infections (HAI) has been carried out in intensive care units (ICU) by replacing some objects generally made of stainless steel or other materials with solid pieces of copper. The authors’ proposal for a sustainable use of copper consists of introducing it in a lamina + adhesive format. This proposal has been tested in an ICU at the Ceuta Hospital in Spain. It has been found to provide an equally efficient solution as antibacterial material than the usual solid format, but with only a layer of 50 microns of copper, which is a high-cost and limited resource. After that intervention, some improvements are also proposed: a standardization of the pieces chosen to cover with a lamina of copper for saving material; and another method of replacement aiming to lower the time that the ICU cannot be used. To ensure that the proposed bonding method is harmless to human health and the adhesive does not interfere with the indoor environment by releasing toxic chemicals, the lamina + adhesive sheet has been further tested. The results and proposals are briefly shown.

The Arctic is subject to long-range atmospheric deposition of globally-distilled semi-volatile organic compounds (SVOCs) that bioaccumulate and biomagnify in lipid-rich food webs. In addition, locally contaminated sites may also contribute SVOCs to the arctic environment. Specifically, Alaska has hundreds of formerly used defense (FUD) sites, many of which are co-located with Alaska Native villages in remote parts of the state. The purpose of this study was to investigate the extent of SVOC contamination on Alaska’s St. Lawrence Island through the analysis of sentinel fish, the ninespine stickleback (Pungitius pungitius), collected from Troutman Lake located within the watershed of an FUD site and adjacent to the Yupik community of Gambell. We measured the concentrations of legacy and emerging SVOCs in 303 fish samples (81 composites), including polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), organophosphate esters (OPEs) and their diester metabolites, and per- and poly-fluoroalkyl substances (PFAS). PBDEs and PCBs were the most abundant SVOC groups found in stickleback with Sigma PBDE and Sigma PCB median concentrations of 25.8 and 10.9 ng/g ww, respectively, followed by PFAS (median Sigma PFAS 7.22 ng/g ww). Sigma OPE and Sigma OPE metabolite concentrations were lower with median concentrations of 4.97 and 1.18 ng/g ww, respectively. Chemical patterns and distributions based on correlations and comparison with SVOC concentrations in stickleback from other parts of the island suggest strong local sources of PCBs, PBDEs, and PFAS on St. Lawrence Island. Casein kinase II (CK2) is an intensively studied enzyme, involved in different diseases, cancer in particular. Different scaffolds were used to develop inhibitors of this enzyme. Here, we report on the synthesis and biological evaluation of twenty phenolic, ketonic, and para-quinonic indeno[1,2-b]indole derivatives as CK2 inhibitors. The most active compounds were 5-isopropyl-1-methyl-5,6,7,8-tetrahydroindeno[1,2-b]indole-9,10-dione 4h and 1,3-dibromo-5-isopropyl-5,6,7,8-tetrahydroindeno[1,2-b]indole-9,10-dione 4w with identical IC50 values of 0.11 mu M. Furthermore, the development of a QSAR model based on the structure of indeno[1,2-b]indoles was performed. This model was used to predict the activity of 25 compounds with naphtho[2,3-b]furan-4,9-dione derivatives, which were previously predicted as CK2 inhibitors via a molecular modeling approach. The activities of four naphtho[2,3-b]furan-4,9-dione derivatives were determined in vitro and one of them (N-isopentyl-2-methyl-4,9-dioxo-4,9-dihydronaphtho[2,3-b]furan-3-carboxamide) turned out to inhibit CK2 with an IC50 value of 2.33 mu M. All four candidates were able to reduce the cell viability by more than 60% after 24 h of incubation using 10 mu M.
