Zhou, Jingwei;Li, Lamei;Wang, Songping;Yan, Ming;Wei, Wentao research published 《 Catalyst-free photodecarbonylation of ortho-amino benzaldehydes》, the research content is summarized as follows. A catalyst-free photodecarbonylation of the aldehydes I (R = H, 2-F, 4-Br, 3-CN, etc.; R1 = Me, Et, n-Bu, Bn; R2 = Me, prop-2-en-1-yl, Bn, etc.; R1R2 = -(CH2)4-, -(CH2)5-, -(CH2)6-, etc.) based on the conformational-selectivity design strategy has been discovered. It revealed that decarbonylation can be easily achieved with visible light irradiation by introducing a tertiary amine into the ortho-position of the aldehyde group. A diverse array of tertiary amines II is tolerated by our photodecarbonylation under mild conditions. Furthermore, the (QM) computations of the mechanism and the experiments on well-designed special substrates revealed that our photodecarbonylation depends on the conformational specificity of the aldehyde group and tertiary amine, and occurs through an unusual [1,4]-H shift and a subsequent [1,3]-H shift.

The pandemic of acute respiratory disease in 2019 caused by highly pathogenic and infectious SARS-CoV-2 has seriously endangered human public safety. The 6-helix bundle (6-HB) heptad repeat 1-heptad repeat 2 (HR1-HR2 complex) formation occurring in the process of spike protein-mediated membrane fusion could serve as a conserved and potential target for the design of fusion inhibitors. Based on the HR2 domain of 6-HB, we designed and synthesized 32 stapled peptides using an all-hydrocarbon peptide stapling strategy. Owing to the improved proteolytic stability and higher helical contents, the optimized stapled peptides termed SCH2-1-20 and SCH2-1-27 showed better inhibitory activities against pseudo and authentic SARS-CoV-2 compared to the linear counterpart. Of note, SCH2-1-20 and SCH2-1-27 were proved to interfere with S protein-mediated membrane fusion. Structural modeling indicated similar binding modes between SCH2-1-20 and the linear peptide. These optimized stapled peptides could serve as potent fusion inhibitors in treating and preventing SARS-CoV-2, and the corresponding SAR could facilitate further optimization.

Jindou kumquat (Fortunella hindsii var. chintou Swingle) with very small fruits belongs to genus Fortunella. However, there is a lack of relevant research on the characteristic secondary metabolites in Jindou compared with other Fortunella fruits. Here, non-targeted and targeted metabolomics based on UPLC-Q-TOF-MS and UPLC-QqQ-MS/MS was performed to profile and quantitate the phytochems. of 16 varieties including five species in the genus Fortunella. Results showed the types and contents of phytochems. in different varieties were quite distinct from each other. Twenty four of the screened metabolites in Jindou different from the other varieties were undoubtedly or tentatively identified. Targeted quant. anal. verified that coumarins and furanocoumarins could be considered to be the dominant characteristic metabolites in Jindou among other Fortunella fruits including auraptene, bergapten, imperatorin, xanthotoxin and umbelliferone. This study makes relatively comprehensive research into the secondary metabolites of Jindou and provides data as a basis for further evaluation and utilization of Fortunella germplasms.

Total twenty-nine [1,2,4]triazolo[4,3-a]pyrazine derivatives were designed and synthesized. The target compounds, especially 4aa, showed potent activity to inhibit c-Met both in an enzyme assay and a cellular assay. The comprehensive screening for the inhibition of 60 different kinases revealed that 4aa could selectively inhibit c-Met while had no effect on other kinases, indicating 4aa is an excellent c-Met selective inhibitor. The flow cytometry studies found that 4aa had a similar behavior to the pos. control SGX-523 in terms of causing the tumor cell apoptosis and blocking cell-cycle progression. More importantly, 4aa showed much better pharmacokinetic properties than SGX-523. Altogether, the findings suggested the target compounds may be potential anti-tumor drug candidates.

Zhao, Hua;Martin, Caden J.;Larm, Nathaniel E.;Baker, Gary A.;Trujillo, Tyler C. research published 《 Enzyme activation by water-mimicking dual-functionalized ionic liquids》, the research content is summarized as follows. Biocatalytic synthesis represents a green alternative to metal-catalyzed reactions. However, enzymes typically display much lower catalytic activities in nonaqueous solvents than in aqueous media. To mimic the aqueous environment for enzyme activation, this study designed a series of sixteen dual-functionalized ionic liquids (ILs) that contain both glycol ether (hydrogen-bond acceptor) and tert-alc. (hydrogen-bond donor) groups. These “water-like” ILs enabled high transesterification activities for immobilized Candida antarctica lipase B (CALB) known as Novozym 435 and immobilized Bacillus licheniformis protease (known as subtilisin A), resp. Several water-mimicking ILs containing 2-3 vol% water significantly increased the CALB activity by 1.8-fold of that in tert-butanol, and 1.6-fold of that in diisopropyl ether (both organic solvents are highly enzyme-compatible). To a smaller degree, subtilisin was activated by these ionic solvents up to 1.2-fold (with 100% selectivity at 2 vol% water) than by diisopropyl ether. Fluorescence emission spectra suggested that the characteristic emission maximum peaks were maintained in “water-like” ILs in most cases.

In this paper, the liquefied products obtained from corn stalk using polyhydric alcs. 1,2-propanediol (PG) mixed diethylene glycol (DEG) through acid catalysis under atm. pressure were characterized by various anal. technologies. The results indicated that 39 kinds of organic compounds were present in bio-oil, among which alcs. were the most, phenols were the second, and their relative contents were 70.7% and 25.6%, resp. There were also some organic acids, ethers, esters, and ketones. More than 80% of these compounds had a carbon number less than 25. Carbon NMR spectra (13C-NMR) showed that different chem. shifts δ (ppm) corresponded to various carbon types. The chem. composition of the residue from liquefaction was complex and contained a certain amount of large mol. substances that were difficult to degrade. It required more severe pyrolysis conditions than those of corn stalk. Results from X-Ray Diffraction (XRD) indicated the destruction of crystalline structure of carbohydrates and the cellulose mols. were cracked, indicating that the cellulose was degraded and the degree of liquefaction was high.

530-59-6, Sinapinic acid is a chemical compound that is the dihydroxybenzoic acid derivative of sinapic acid. It has been shown to have anti-inflammatory properties in vitro and in vivo. Sinapinic acid inhibits the activity of various enzymes, such as cyclooxygenase (COX), lipoxygenase (LOX), and 5-lipoxygenase-activating protein (FLAP). It also decreases levels of adhesion molecules and downregulates inflammatory response genes. Sinapinic acid has been shown to reduce inflammation by inhibiting the formation of proinflammatory mediators, such as prostaglandin E2 or leukotriene B4, in endothelial cells and mammary epithelial cells.
Sinapic acid is a phenylpropanoid hydroxycinnamic acid with diverse biological activities. Sinapic acid inhibits collagen-induced human platelet aggregation by up to 70% in vitro (IC50 = 1.03 mM). It scavenges 2,2-diphenyl-1-picrylhydrazyl (DPPH; ) and 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonate) (ABTS) free radicals with IC50 values of 8.3 and 5.4 μg/ml, respectively. Sinapic acid (200 μM) reduces colony formation of SW480 human colon carcinoma cells by 4-fold. It also inhibits colony formation of E. coli, S. enteritidis, and S. aureus on agar (MICs = 2.2, 2, and 1.8 mM, respectively). In vivo, sinapic acid (4 mg/kg, p.o.) increases the time spent in the open arms of the elevated plus maze by approximately 15% in mice, an effect that can be blocked by the GABAA receptor antagonists flumazenil and bicuculline. Sinapic acid is also commonly used as a matrix in protein mass spectrometry.
Sinapic acid analytical standard provided with w/w absolute assay, to be used for quantitative titration.
Sinapic acid is an hydroxycinnamic acid derivative that occurs naturally in Brassicaceae species.
cis-Sinapic acid, also known as cis-sinapate or synapitic acid, belongs to the class of organic compounds known as hydroxycinnamic acids. Hydroxycinnamic acids are compounds containing an cinnamic acid where the benzene ring is hydroxylated. cis-Sinapic acid is considered to be a practically insoluble (in water) and relatively neutral molecule. Within the cell, cis-sinapic acid is primarily located in the cytoplasm. Outside of the human body, cis-sinapic acid can be found in common pea and pulses. This makes cis-sinapic acid a potential biomarker for the consumption of these food products.
Cis-sinapic acid is a 3-(4-hydroxy-3,5-dimethoxyphenyl)prop-2-enoic acid in which the double bond has cis-configuration. It has been isolated from the shoots of alfalfa. It has a role as a plant metabolite., Quality Control of 530-59-6

Ethers can again be classified into two varieties: if the alkyl or aryl groups are the same on both sides of the oxygen atom, 530-59-6, formula is C11H12O5, Name is 3,5-Dimethoxy-4-hydroxycinnamic acid. Then it is a simple or symmetrical ether, whereas if they are different, the ethers are called mixed or unsymmetrical ethers. Quality Control of 530-59-6.

Zhang, Xin;Zheng, Yuanrong;Zhou, Ran;Ma, Ming research published 《 Comprehensive identification of molecular profiles related to sensory and nutritional changes in Mongolian cheese during storage by untargeted metabolomics coupled with quantification of free amino acids》, the research content is summarized as follows. Non-targeted metabolomics was used to study metabolites with low mol. weight which may contribute to quality deterioration of Mongolian cheese during storage. Microbiol. anal., pH, FAAs (free amino acids), volatile compounds, and sensory evaluation of the cheese during storage were also studied. A total of 278 metabolites were identified in Mongolian cheese, of which 51 metabolites were used as differential metabolites, including amino acids, peptides, organic acids, lipids, and carbohydrates. Bitter amino acids, bitter peptide (Phe-Ile), and organic acids (sinapic acid, butyric acid) increased during storage. Metabolic pathway anal. showed that differential metabolites were mainly related to amino acid metabolism, such as β-alanine metabolism and glycine, serine, and threonine metabolism Moreover, accompanied with the increased contents of short-chain fatty acids, 2-undecanone and Et esters, strength of odor and unpleasant smell increased but overall acceptability decreased during Mongolian cheese storage. This research provides suitable strategies for quality control of Mongolian cheese during shelf life.
