Pehlivan Karakas, Fatma;Sahin, Gunce;Ucar Turker, Arzu;Verma, Sandeep Kumar research published 《 Impacts of heavy metal, high temperature, and UV radiation exposures on Bellis perennis L. (common daisy): Comparison of phenolic constituents and antioxidant potential (enzymatic and non-enzymatic)》, the research content is summarized as follows. Bellis perennis L., common daisy, is a beneficial medicinal plant used in folk medicine to treat skin diseases, wounds, sore throats, common colds, inflammation, and rheumatism. The objective of this study was to determine the effect of 4 different abiotic stress applications [mercury chloride (HgCl2), copper sulfate (CuSO4), high temperature (45°C) and UV radiation (UV)] on individual phenolic constituents, non-enzymic antioxidant activities (free radical scavenging activity, total phenol-flavonoid content, and proline level), and enzymic antioxidant activities [superoxide dismutase (SOD) and catalases (CAT)] in common daisy callus cultures. Quantification of the 21 phenolic substances was performed using LC-ESI-MS/MS anal. and all applied stress applications increased specific phenolic constituents comparing with control (no exposure to any stress). Heavy metal stresses (HgCl2 and CuSO4) and high temperature application enhanced the quantity of the selected phenolic substances more than UV-radiation stress. The most noticeable increased quantities were observed with high temperature stress in p-coumaric acid (51.74-fold), and with CuSO4 stress in luteolin (17.76-fold) and apigenin (15.62-fold). Chlorogenic acid was the most prevalent phenolic compound in all tested callus extract including control. Although the first highest total phenol content was obtained with HgCl2 exposure and then followed by high temperature stress, the first highest total flavonoid content was obtained with high temperature stress and then UV radiation. The antioxidant capacity was significantly enhanced (2.11 fold increase) with CuSO4 stress application. Stress-related enzymic antioxidant activity (SOD and CAT) was elevated the most with HgCl2 stress. Similarly, the highest proline content was obtained with HgCl2 treatment. These findings showed the ability of the phenolic compounds to protect common daisy against harmful heavy metals, high temperature and UV-radiation stresses. The adaptation of the callus under abiotic stress conditions developed here may be useful for the production of optimized phenolic compounds of common daisy and the exploitation of other medicinal species of interest.

Paul, Dipayan;Mahanta, Saurov;Tag, Hui;Das, Sanjib Kumar;Das Gupta, Debmalya;Tanti, Bhaben;Ananthan, Rajendran;Das, Ranjan;Jambhulkar, Sanjay;Hui, Pallabi Kalita research published 《 Identification of tyrosine kinase inhibitors from Panax bipinnatifidus and Panax pseudoginseng for RTK-HER2 and VEGFR2 receptors, by in silico approach》, the research content is summarized as follows. Breast and stomach cancer is reported as a leading cause for human mortality across the world. The overexpression of receptor tyrosine kinase (RTK) proteins, namely the human epidermal growth factor receptor2 (HER2) and the vascular endothelial growth factor receptor2 (VEGFR2), is reported to be responsible for development and metastasis of breast and stomach cancer. Although several synthetic tyrosine kinase inhibitors (TKIs) as drug candidates targeting RTK-HER2 and VEGFR2 are currently available in the market, these are expensive with the reported side effects. This confers an opportunity for development of alternative novel tyrosine kinase inhibitors (TKIs) for RTK-HER2 and VEGFR2 receptors from the botanical sources. In the present study, we characterized 47 bioactive phytocompounds from the methanol extracts of the rhizomes of Asiatic traditional medicinal herbs-Panax bipinnatifidus and Panax pseudoginseng, of Indian Himalayan landraces using HPLC, GC-MS and high-sensitivity LC-MS tools. We performed mol. docking and mol. dynamics simulation anal. using Schrodinger suite 2020-3 to confirm the TKI phytocompounds showing the best binding affinity towards RTK-HER2 and VEGFR2 receptors. The results of mol. docking studies confirmed that the phytocompound (ligand) luteolin 7-O-glucoside (IHP15) showed the highest binding affinity towards receptor HER2 (PDB ID: 3PP0) with docking score and Glide g score (G-Score) of – 13.272, while chlorogenic acid (IHP12) showed the highest binding affinity towards receptor VEGFR2 (PDB ID: 4AGC) with docking score and Glide g score (G-Score) of – 10.673. Mol. dynamics (MD) simulation anal. carried out for 100 ns has confirmed strong binding interaction between the ligand and receptor complex [luteolin 7-O-glucoside (IHP15) and HER2 (PDB ID: 3PP0)] and is found to be stabilized within 40 to 100 ns of MD simulation, whereas ligand-receptor complex [chlorogenic acid (IPH12) and VEGFR2 (PDB ID: 4AGC)] also showed strong binding interaction and is found to be stabilized within 18-30 ns but slightly deviated during 100 ns of MD simulation. In silico ADME-Tox study using SwissADME revealed that the ligands luteolin 7-O-glucoside (IHP15) and chlorogenic acid (IHP12) have passed majority parameters of the common drug discovery rules. The present study has confirmed luteolin 7-O-glucoside (IHP15) and chlorogenic acid (IHP12) as potential tyrosine kinase inhibitors (TKIs) which were found to inhibit RTKs-HER2 and VEGFR2 receptor proteins, and thus paving the way for development of alternative potential TKIs (drug mols.) for treatment of HER2- and VEGFR2-pos. breast and stomach cancer.

The malignant transformation of melanocytes causes several thousand deaths each year, making melanoma an important public health concern. Melanoma is the most aggressive skin cancer, which incidence has regularly increased over the past decades. We described here the preparation of new compounds based on the 1-(3,4-dihydroxyphenyl)imidazo[1,2-a]quinoxaline structure. Different positions of the quinoxaline moiety were screened to introduce novel substituents in order to study their influence on the biol. activity. Several alkylamino or alkyloxy groups were also considered to replace the methylamine of our first generation of Imiqualines. Imidazo[1,2-a]pyrazine derivatives were also designed as potential minimal structure. The investigation on A375 melanoma cells displayed interesting in vitro low nanomolar cytotoxic activity. Among them, 9d (EAPB02303) is particularly remarkable since it is 20 times more potent than vemurafenib, the reference clin. therapy used on BRAF mutant melanoma. Contrary to the first generation, EAPB02303 does not inhibit tubulin polymerization, as confirmed by an in vitro assay and a mol. modelisation study. The mechanism of action for EAPB02303 highlighted by a transcriptomic anal. is clearly different from a panel of 12 well-known anticancer drugs. In vivo EAPB02303 treatment reduced tumor size and weight of the A375 human melanoma xenografts in a dose-dependent manner, correlated with a low mitotic index but not with necrosis.

Peptide-Peptide Nucleic Acid (PNA) conjugates targeting essential bacterial genes have shown significant potential in developing novel antisense antimicrobials. The majority of efforts in this area are focused on identifying different PNA targets and the selection of peptides to deliver the peptide-PNA conjugates to Gram-neg. bacteria. Notably, the selection of a linkage strategy to form peptide-PNA conjugate plays an important role in the effective delivery of PNAs. Recently, a unique Cysteine- 2-Cyanoisonicotinamide (Cys- CINA) click chem. has been employed for the synthesis of cyclic peptides. Considering the high selectivity of this chem., we investigated the efficiency of Cys-CINA conjugation to synthesize novel antimicrobial peptide-PNA conjugates. The PNA targeting acyl carrier protein gene (acpP), when conjugated to the membrane-active antimicrobial peptides (polymyxin), showed improvement in antimicrobial activity against multidrug-resistant Gram-neg. Acinetobacter baumannii. Thus, indicating that the Cys-CINA conjugation is an effective strategy to link the antisense oligonucleotides with antimicrobial peptides. Therefore, the Cys-CINA conjugation opens an exciting prospect for antimicrobial drug development.

Rasagiline mesylate (RSM) is a selective and irreversible monoamine oxidase B inhibitor used for the treatment of Parkinson’s disease (PD). However, its unfavorable biopharmaceutical properties, such as extensive degradation in the gastrointestinal tract and first-pass metabolism are responsible for its low oral bioavailability and suboptimal therapeutic efficacy. Here, we report the feasibility of delivering RSM via the transdermal route using RSM containing microemulsion-based gel (RSM-MEG) to achieve effective management of PD. Our in vitro skin permeation studies of RSM-MEG showed significantly higher (at least ∼1.5-fold) permeation across rat skin compared to the conventional RSM hydrogel. Our skin irritation studies in rabbits showed that RSM-MEG is safe for transdermal application. Finally, using the rat model of rotenone-induced Parkinsonism, we demonstrated that the topical application of RSM-MEG was equally effective in reversing PD symptoms when compared to oral RSM therapy. Thus, our study confirmed the feasibility and potential of transdermal delivery of RSM via simple topical application of RSM-MEG, and this approach could be an alternative therapeutic intervention for the treatment of Parkinson’s disease.

Selenium deficiency in daily diet is a common problem in many countries, thus searching for new dietary sources of this trace element is an important scientific challenge. Selenium biofortified sprouts from Brassicaceae family are good candidates for new dietary selenium source, as they reveal one of the highest capability to synthesize and accumulate this element. As a part of this extensive search, the influence of novel selenium organic compounds on fortification of kale sprouts biol. activity was investigated. The present study is focused on the evaluation of the influence of these compounds on the synthesis of glucosinolates, isothiocyanates, indoles and phenolic acids in kale sprouts, together with the determination of their impact on antioxidant, anti-inflammatory and cytotoxic activity on gastrointestinal, prostate, and thyroid normal and cancer cells. The present study yields the conclusion that fortification of kale sprouts with selenium organic compounds bearing benzoselenoate scaffold influences the production of isothiocyanates, phenolic acids, and enhances the antioxidant properties of fortified sprouts. Notably, fortification with compounds based on benzoselenoate scaffold display chemoprotective properties in various cancer types (gastric, thyroid, and prostate cancer). The present study can facilitate the design of future agrochems. Compounds bearing benzoselenoate scaffold or selenyl phenylpiperazine motif seem to be particularly promising for these purposes.

Formula: C11H12O5, Sinapinic acid is a chemical compound that is the dihydroxybenzoic acid derivative of sinapic acid. It has been shown to have anti-inflammatory properties in vitro and in vivo. Sinapinic acid inhibits the activity of various enzymes, such as cyclooxygenase (COX), lipoxygenase (LOX), and 5-lipoxygenase-activating protein (FLAP). It also decreases levels of adhesion molecules and downregulates inflammatory response genes. Sinapinic acid has been shown to reduce inflammation by inhibiting the formation of proinflammatory mediators, such as prostaglandin E2 or leukotriene B4, in endothelial cells and mammary epithelial cells.
Sinapic acid is a phenylpropanoid hydroxycinnamic acid with diverse biological activities. Sinapic acid inhibits collagen-induced human platelet aggregation by up to 70% in vitro (IC50 = 1.03 mM). It scavenges 2,2-diphenyl-1-picrylhydrazyl (DPPH; ) and 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonate) (ABTS) free radicals with IC50 values of 8.3 and 5.4 μg/ml, respectively. Sinapic acid (200 μM) reduces colony formation of SW480 human colon carcinoma cells by 4-fold. It also inhibits colony formation of E. coli, S. enteritidis, and S. aureus on agar (MICs = 2.2, 2, and 1.8 mM, respectively). In vivo, sinapic acid (4 mg/kg, p.o.) increases the time spent in the open arms of the elevated plus maze by approximately 15% in mice, an effect that can be blocked by the GABAA receptor antagonists flumazenil and bicuculline. Sinapic acid is also commonly used as a matrix in protein mass spectrometry.
Sinapic acid analytical standard provided with w/w absolute assay, to be used for quantitative titration.
Sinapic acid is an hydroxycinnamic acid derivative that occurs naturally in Brassicaceae species.
cis-Sinapic acid, also known as cis-sinapate or synapitic acid, belongs to the class of organic compounds known as hydroxycinnamic acids. Hydroxycinnamic acids are compounds containing an cinnamic acid where the benzene ring is hydroxylated. cis-Sinapic acid is considered to be a practically insoluble (in water) and relatively neutral molecule. Within the cell, cis-sinapic acid is primarily located in the cytoplasm. Outside of the human body, cis-sinapic acid can be found in common pea and pulses. This makes cis-sinapic acid a potential biomarker for the consumption of these food products.

Novel heteroaryl-containing benzamide derivatives were synthesized and screened using an in vitro assay measuring increases in glucose uptake and glucokinase activity stimulated by 10 mM glucose in rat hepatocytes. From a library of synthesized compounds, 3-(4-methanesulfonylphenoxy)-N-[1-(2-methoxy-ethoxymethyl)-1H-pyrazol-3-yl]-5-(3-Me pyridin-2-yl)-benzamide (19e) was identified as a potent glucokinase activator with assays demonstrating an EC50 of 315 nM and the induction of a 2.23 fold increase in glucose uptake. Compound 19e exhibited a glucose AUC reduction of 32% (50 mg/kg) in an OGTT study with C57BL/6J mice compared to 28% for metformin (300 mg/kg). Single treatment of the compound in C57BL/J6 and ob/ob mice elicited basal glucose lowering activity, while in a two-week repeated dose study with ob/ob mice, the compound significantly decreased blood glucose levels with no evidence of hypoglycemia risk. In addition, 19e exhibited favorable pharmacokinetic parameters in mice and rats and excellent safety margins in liver and testicular toxicity studies. Compound 19e was therefore selected as a development candidate for the potential treatment of type 2 diabetes.

Stapled peptides have the ability to mimic α-helixes involved in protein binding and have proved to be effective pharmacol. agents for disrupting protein-protein interactions. DNA-binding proteins such as transcription factors bind their cognate DNA sequences via an α-helix interacting with the major groove of DNA. We previously developed a stapled peptide based on the bacterial alternative sigma factor RpoN capable of binding the RpoN DNA promoter sequence and inhibiting RpoN-mediated expression in Escherichia coli. We have elucidated a structure-activity relationship for DNA binding by this stapled peptide, improving DNA binding affinity constants in the high nM range. Lead peptides were shown to have low toxicity as determined by their low hemolytic activity at 100 μM and were shown to have anti-virulence activity in a Galleria mellonella model of Pseudomonas aeruginosa infection. These findings support further preclin. development of stapled peptides as antivirulence agents targeting P. aeruginosa.

Paprocki, Maciej P.;Rasmussen, Jakob E.;Soerensen, Kasper K.;Jensen, Knud J. research published 《 Safety-catch linkers for Fmoc solid-phase synthesis of peptide thioesters and hydrazides by amide-to-imide activation》, the research content is summarized as follows. The use of C-terminal peptide thioesters and hydrazides in synthetic protein chem. has inspired the search for optimal solid-phase peptide synthesis (SPPS) strategies for their assembly. However, peptide thioesters are not directly accessible by conventional Fmoc-SPPS (Fmoc = 9-fluorenylmethoxycarbonyl) owing to the nucleophilicity of the secondary amine required for Fmoc removal. Here, we report the mild and effective activation of the pGlu linker and a new safety-catch linker that was used for the convenient synthesis of peptide thioesters and hydrazides via efficient amide-to-imide activation followed by nucleophilic displacement.
