Masson-Makdissi, Jeanne;Prieto, Liher;Abel-Snape, Xavier;Lautens, Mark research published 《 Enantio- and Diastereodivergent Sequential Catalysis Featuring Two Transition-Metal-Catalyzed Asymmetric Reactions》, the research content is summarized as follows. This study demonstrated the feasibility and inherent benefits of combining two distinct asym. transition-metal-catalyzed reactions in one pot. The reported transformation feature a Pd-catalyzed asym. allylic alkylation and a Rh-catalyzed enantioselective 1,4-conjugate addition, effectively converting simple allyl enol carbonate precursors into enantioenriched cyclic ketones such as I [R = Me, (CH2)2CN, (CH2)2CO2Me, CH2OTBS; Ar = C6H5, 2-MeC6H4, 2-naphthyl, etc.] with two remote stereocenters. Despite the anticipated challenges associated with controlling stereoselectivity in such a complex system, the products were obtained in enantiomeric excesses ranging up to >99% ee, exceeding those obtained from either of the individual asym. reactions. In addition, since the stereoselectivity of both steps was under catalyst control, this one-pot reaction was enantio- and diastereodivergent, enabling facile access to all stereoisomers from the same set of starting materials.

Marasinghege, Chalani;Broadfoot, Ross;Bottle, Steven;Bartley, John;Doherty, William O. S.;Rackemann, Darryn W. research published 《 Investigation on the effect of the heating surface temperature of 1st evaporator on sucrose loss and the degradation of sugarcane juice constituents》, the research content is summarized as follows. To enable more energy efficient sugar production from sugarcane, higher process temperatures in the 1st evaporator is typically utilized, as this allows higher temperature bleed vapors to be used rather than process steam for other heating duties in the factory. This invariably increases juice degradation and the temperature difference (ΔT) between the heated surface and juice can be even higher where process steam used for heating is superheated. This study investigated the effect of high surface temperatures (139-168°C) and residence times (0-60 min) on both factory and synthetic juice degradation using a specially designed laboratory evaporator rig. The results show that ΔT is inversely proportional to the heat transfer coefficient At up to 148°C and short residence time, there were minor differences in the juice degradation profile. However, at higher temperatures there were significant sucrose loss (≤2.5%), amino acid degradation, lignin (associated with fiber in juice) depolymerization to hydroxycinnamic acids, isomerization of trans-to cis-aconitic acid, and the formation of alcs. Strategies to minimize juice degradation in evaporators are highlighted.

Mao, Runyu;Xi, Shiyi;Shah, Sayali;Roy, Michael J.;John, Alan;Lingford, James P.;Gade, Gerd;Scott, Nichollas E.;Goddard-Borger, Ethan D. research published 《 Synthesis of C-mannosylated glycopeptides enabled by Ni-catalyzed photoreductive cross-coupling reactions》, the research content is summarized as follows. The biol. functions of tryptophan C-mannosylation are poorly understood, in part, due to a dearth of methods for preparing pure glycopeptides and glycoproteins with this modification. To address this issue, efficient and scalable methods are required for installing this protein modification. Here, we describe unique Ni-catalyzed cross-coupling conditions that utilize photocatalysis or a Hantzsch ester photoreductant to couple glycosyl halides with (hetero)aryl bromides, thereby enabling the α-C-mannosylation of 2-bromo-tryptophan, peptides thereof, and (hetero)aryl bromides more generally. We also report that 2-(α-D-mannopyranosyl)-L-tryptophan undergoes facile anomerization in the presence of acid: something that must be considered when preparing and handling peptides with this modification. These developments enabled the first automated solid-phase peptide syntheses of C-mannosylated glycopeptides, which we used to map the epitope of an antibody, as well as providing the first verified synthesis of Carmo-HrTH-I, a C-mannosylated insect hormone. To complement this approach, we also performed late-stage tryptophan C-mannosylation on a diverse array of peptides, demonstrating the broad scope and utility of this methodol. for preparing glycopeptides.

In particular, the peel resulted rich of flavan-3-ols and proanthocyanidins, whereas seeds contained high amount of organic acids, including ferulic, citric and sinapic acids. Concerning functional properties, both edible and non-edible portions showed a significant prevention of lipid peroxidation in a cell-based model. Moreover, the results suggested that the antioxidant protection involved both redox active properties and gene expression modulation. Concerning redox active properties, peel extracts showed an antioxidant activity 7/12-fold higher than the edible portion, while seed extracts were more active in increasing catalase gene expression. The obtained results confirmed that black sapote is a good source of antioxidant phytochems. and its non-edible portions have a great potential in the production of functional foods and supplements.

Mandal, Govinda;Moran, Lukas;Pecinka, Lukas;Vanhara, Petr;Havel, Josef research published 《 Matrix enrichment by black phosphorus improves ionization and reproducibility of mass spectrometry of intact cells, peptides, and amino acids》, the research content is summarized as follows. Intact (whole) cell matrix-assisted laser desorption/ionization mass spectrometry (MALDI-TOF MS) is an established method for biotyping in clin. microbiol. as well as for revealing phenotypic shifts in cultured eukaryotic cells. Intact cell MALDI-TOF MS has recently been introduced as a quality control tool for long-term cultures of pluripotent stem cells. Despite the potential this method holds for revealing minute changes in cells, there is still a need for improving the ionization efficiency or peak reproducibility. Here we report for the first time that supplementation by fine particles of black phosphorus to the standard MALDI matrixes, such as sinapinic and α-cyano-4-hydroxycinnamic acids enhance intensities of mass spectra of particular amino acids and peptides, presumably by interactions with aromatic groups within the mols. In addition, the particles of black phosphorus induce the formation of small and regularly dispersed crystals of sinapinic acid and α-cyano-4-hydroxycinnamic acid with the analyte on a steel MALDI target plate. Patterns of mass spectra recorded from intact cells using black phosphorus-enriched matrix were more reproducible and contained peaks of higher intensities when compared to matrix without black phosphorus supplementation. In summary, enrichment of common organic matrixes by black phosphorus can improve discrimination data anal. by enhancing peak intensity and reproducibility of mass spectra acquired from intact cells.

HPLC of Formula: 530-59-6, Sinapinic acid is a chemical compound that is the dihydroxybenzoic acid derivative of sinapic acid. It has been shown to have anti-inflammatory properties in vitro and in vivo. Sinapinic acid inhibits the activity of various enzymes, such as cyclooxygenase (COX), lipoxygenase (LOX), and 5-lipoxygenase-activating protein (FLAP). It also decreases levels of adhesion molecules and downregulates inflammatory response genes. Sinapinic acid has been shown to reduce inflammation by inhibiting the formation of proinflammatory mediators, such as prostaglandin E2 or leukotriene B4, in endothelial cells and mammary epithelial cells.
Sinapic acid is a phenylpropanoid hydroxycinnamic acid with diverse biological activities. Sinapic acid inhibits collagen-induced human platelet aggregation by up to 70% in vitro (IC50 = 1.03 mM). It scavenges 2,2-diphenyl-1-picrylhydrazyl (DPPH; ) and 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonate) (ABTS) free radicals with IC50 values of 8.3 and 5.4 μg/ml, respectively. Sinapic acid (200 μM) reduces colony formation of SW480 human colon carcinoma cells by 4-fold. It also inhibits colony formation of E. coli, S. enteritidis, and S. aureus on agar (MICs = 2.2, 2, and 1.8 mM, respectively). In vivo, sinapic acid (4 mg/kg, p.o.) increases the time spent in the open arms of the elevated plus maze by approximately 15% in mice, an effect that can be blocked by the GABAA receptor antagonists flumazenil and bicuculline. Sinapic acid is also commonly used as a matrix in protein mass spectrometry.

Mallek, Aaron J.;Pentelute, Bradley L.;Buchwald, Stephen L. research published 《 Selective N-arylation of p-aminophenylalanine in unprotected peptides with organometallic palladium reagents》, the research content is summarized as follows. The selective N-arylation of p-aminophenylalanine in polypeptides with pre-formed palladium oxidative addition complexes is described. The depressed pKa of the aniline NH2 group enables chemoselective C-N bond formation on peptides containing multiple other aliphatic amino groups at lysines or the N-terminus via Curtin-Hammett control under mild conditions. Using palladium complexes derived from electron-poor aryl halides, p-aminophenylalanine is fully arylated in aqueous buffer in as little as one hour at micromolar concentrations A complementary protocol using the non-nucleophilic, organic base 1,5-diazabicyclo(4.3.0)non-5-ene (DBN), expands the substrate scope to tolerate electron-rich functional groups provides up to 97% conversion. These procedures enable the chemoselective conjugation of functionally diverse small mol. pharmaceuticals to p-aminophenylalanine containing derivatives of cell-penetrating peptides.

Malik, Uru;Chan, Lai Yue;Cai, Minying;Hruby, Victor J.;Kaas, Quentin;Daly, Norelle L.;Craik, David J. research published 《 Development of novel frog-skin peptide scaffolds with selectivity towards melanocortin receptor subtypes》, the research content is summarized as follows. Melanocortin receptors are pharmaceutically important receptors that are involved in complex physiol. functions. They have been associated with various diseases including obesity, erectile dysfunction, acne, and skin cancer. It has been challenging to transform nonselective endogenous agonist and antagonist ligands into selective and potent ligands. In this study, we investigated naturally occurring peptides derived from frog skin secretions for selectivity and activity toward melanocortin receptors. Three peptides (ORB, ORB2K and ranacyclin-T) were found to have selectivity towards the melanocortin receptor 5 (MC5R). ORB and ORB2K had partial binding affinity at nanomolar concentrations, whereas ranacyclin-T had 57% binding efficiency at 1.6μM. Backbone cyclization of ORB and ORB2K altered the binding efficiency to melanocortin receptors. Our results suggest that these frog-skin peptides could be modified for developing melanocortin-specific ligands and potentially future therapeutics.
