Ryan, Philip;Shi, Yun;von Itzstein, Mark;Rudrawar, Santosh research published 《 Novel bisubstrate uridine-peptide analogs bearing a pyrophosphate bioisostere as inhibitors of human O-GlcNAc transferase》, the research content is summarized as follows. Protein O-linked β-D-N-acetylglucosamine (O-GlcNAc) modification (O-GlcNAcylation), an essential post-translational as well as co-translational modification, is the attachment of β-D-N-acetylglucosamine to serine and threonine residues of nucleocytoplasmic proteins. An aberrant O-GlcNAc profile on certain proteins has been implicated in metabolic diseases such as diabetes and cancer. Inhibitors of O-GlcNAc transferase (OGT) are valuable tools to study the cell biol. of protein O-GlcNAc modification. In this study we report novel uridine-peptide conjugate mols. composed of an acceptor peptide covalently linked to a catalytically inactive donor substrate analog that bears a pyrophosphate bioisostere and explore their inhibitory activities against OGT by a radioactive hOGT assay. Further, we investigate the structural basis of their activities via mol. modeling, explaining their lack of potency towards OGT inhibition.

Ruokolainen, Joonas;Hyttinen, Marko;Sorvari, Jouni;Pasanen, Pertti research published 《 Exposure of cleaning workers to chemical agents and physical conditions in swimming pools and spas》, the research content is summarized as follows. Swimming pools and spas require a high hygiene level, and therefore constant cleaning. In this study, cleaning worker′s exposure to volatile organic compounds (VOCs), trichloramine (TCA), and particulate matter (PM) in the swimming pools and spas were evaluated. Also, statistical methods were employed to determine what activities affect the exposure to disinfection byproducts (DBPs). The study was conducted in 32 swimming pools and spas. The measurement locations were pool areas, bathrooms, and locker rooms, both during cleaning and opening hours. During the cleaning, the total volatile organic compound (TVOC) concentrations were low, on average 96, 251, and 91 μg/m3 for locker rooms, bathrooms, and pool areas, resp. Similarly, during the opening hours, the TVOC concentrations were on average 78, 125, and 83 μg/m3, for locker rooms, bathrooms, and pool areas, resp. This is in line with previous studies investigating cleaning work in other environments. The most prevalent compounds during the cleaning were 2-(2-butoxyethoxy)ethanol (DEGBE), 2-(2-ethoxyethoxy)ethanol (DEGEE), 2-butyl-1-octanol, trichloromethane (chloroform), decamethylcyclopentasiloxane (D5), and carbon tetrachloride. The most prevalent compounds during the opening hours were D5, D-limonene, carbon tetrachloride (bathrooms and pool areas), and trichloromethane (bathrooms and pool areas). The TCA concentrations during the cleaning in the bathrooms and pool areas were on average 60 and 67 μg/m3, resp., and during the opening hours, 28 and 122 μg/m3, resp. The use of disinfectants was found to increase the TCA concentration in the bathrooms, while the other cleaning products did not. Even though the TCA concentrations were below the WHO′s guideline and the Finnish occupational exposure limit value of 500 μg/m3, the measured TCA levels were occasionally high enough to pose a risk of irritative symptoms. The PM concentrations were low, both in the real-time monitoring (aerodynamic diameter, Dae ≤ 15 μm) and inhalable dust samples (Dae ≤ 100 μm). Highest measured inhalable dust concentration was 350 μg/m3, well below the Finnish occupational limit value of 5,000 μg/m3 for organic inhalable dust.

Category: ethers-buliding-blocks, Sinapinic acid is a chemical compound that is the dihydroxybenzoic acid derivative of sinapic acid. It has been shown to have anti-inflammatory properties in vitro and in vivo. Sinapinic acid inhibits the activity of various enzymes, such as cyclooxygenase (COX), lipoxygenase (LOX), and 5-lipoxygenase-activating protein (FLAP). It also decreases levels of adhesion molecules and downregulates inflammatory response genes. Sinapinic acid has been shown to reduce inflammation by inhibiting the formation of proinflammatory mediators, such as prostaglandin E2 or leukotriene B4, in endothelial cells and mammary epithelial cells.
Sinapic acid is a phenylpropanoid hydroxycinnamic acid with diverse biological activities. Sinapic acid inhibits collagen-induced human platelet aggregation by up to 70% in vitro (IC50 = 1.03 mM). It scavenges 2,2-diphenyl-1-picrylhydrazyl (DPPH; ) and 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonate) (ABTS) free radicals with IC50 values of 8.3 and 5.4 μg/ml, respectively. Sinapic acid (200 μM) reduces colony formation of SW480 human colon carcinoma cells by 4-fold. It also inhibits colony formation of E. coli, S. enteritidis, and S. aureus on agar (MICs = 2.2, 2, and 1.8 mM, respectively). In vivo, sinapic acid (4 mg/kg, p.o.) increases the time spent in the open arms of the elevated plus maze by approximately 15% in mice, an effect that can be blocked by the GABAA receptor antagonists flumazenil and bicuculline. Sinapic acid is also commonly used as a matrix in protein mass spectrometry.

Sinapic acid is an hydroxycinnamic acid derivative that occurs naturally in Brassicaceae species.
cis-Sinapic acid, also known as cis-sinapate or synapitic acid, belongs to the class of organic compounds known as hydroxycinnamic acids. Hydroxycinnamic acids are compounds containing an cinnamic acid where the benzene ring is hydroxylated. cis-Sinapic acid is considered to be a practically insoluble (in water) and relatively neutral molecule. Within the cell, cis-sinapic acid is primarily located in the cytoplasm. Outside of the human body, cis-sinapic acid can be found in common pea and pulses. This makes cis-sinapic acid a potential biomarker for the consumption of these food products.
Cis-sinapic acid is a 3-(4-hydroxy-3,5-dimethoxyphenyl)prop-2-enoic acid in which the double bond has cis-configuration. It has been isolated from the shoots of alfalfa. It has a role as a plant metabolite., 530-59-6.

Ethers can again be classified into two varieties: if the alkyl or aryl groups are the same on both sides of the oxygen atom, 530-59-6, formula is C11H12O5, Name is 3,5-Dimethoxy-4-hydroxycinnamic acid. Then it is a simple or symmetrical ether, whereas if they are different, the ethers are called mixed or unsymmetrical ethers. Category: ethers-buliding-blocks.

Ruiz-Alcaraz, Antonio Jose;Martinez-Sanchez, Maria Antonia;Garcia-Penarrubia, Pilar;Martinez-Esparza, Maria;Ramos-Molina, Bruno;Moreno, Diego A. research published 《 Analysis of the anti-inflammatory potential of Brassica bioactive compounds in a human macrophage-like cell model derived from HL-60 cells》, the research content is summarized as follows. Chronic inflammatory diseases are major causes of global morbidity and mortality. Acute inflammation is meant to protect the body against foreign agents, but it also plays a major role in tissue repairment. Several mediators are involved in this process, including pro-inflammatory cytokines produced by macrophages. Occasionally, if the inflammatory response is not resolved, the acute inflammatory process can evolve into a chronic inflammation. Natural compounds from vegetables are considered as an important source of active agents with potential to treat or prevent inflammatory related pathologies and could be used as an alternative of the therapeutic agents currently in use, such as non-steroidal anti-inflammatory drugs (NSAIDs), which present several side effects. In this research work we evaluated in vitro the anti-inflammatory activity of a series of ten phytochems. present in Brassica, measured as the potential of those compounds to reduce the production of key pro-inflammatory cytokines (TNF-α, IL-6 and IL-1β) by a human macrophage-like cell model of HL-60 cellsMost of the tested phytochems. (including the most representative bioactive mols. of the major classes of compounds present in cruciferous foods such as glucosinolates, isothiocyanates, hydroxycinnamic acids, flavonols and anthocyanins) demonstrated significant anti-inflammatory activity at micromolar level in the absence of cytotoxic effects in this human macrophage-like cell model. These data confirm that phytochems. commonly obtained from Brassica may be potential therapeutic leads to treat or prevent human chronic inflammation and related diseases.

Rostami, Amir;Baluchnejadmojarad, Tourandokht;Roghani, Mehrdad research published 《 Sinapic acid ameliorates paracetamol-induced acute liver injury through targeting oxidative stress and inflammation》, the research content is summarized as follows. Acetaminophen (paracetamol, APAP) overdose is the principal cause of acute liver injury (ALI) that leads to liver failure typified with oxidative stress, mitochondrial and lysosomal dysfunction and with few antidotes for this condition. Therefore, more effective therapeutics are urgently required. Sinapic acid is a phenolic phytochem. with significant antioxidant, anti-inflammatory and hepatoprotective potential. This study was conducted to evaluate hepatoprotective effect of this phytochem. in acetaminophen-induced model of ALI. Male C57BL/6 mice were treated p.o. with sinapic acid (10 or 50 mg/kg) 3 times at 72, 24, and 1 h before APAP (300 mg/kg; i.p.) challenge. Functional factors of liver dysfunction were determined along with hepatic assessment of oxidative stress and inflammatory indexes and histopathol. anal. was also conducted. Sinapic acid (50 mg/kg) properly decreased serum levels of ALT, ALP, and AST besides reducing liver level of ROS, MDA, IL-6, TNF-α, NF-kB, and MPO and improved sirtuin 1, HO-1, Nrf2, SOD activity, and MMP with no significant effect on IL-1β and catalase activity in addition to decreasing activity of lysosomal enzymes including cathepsin B and β-galactosidase. Also, sinapic acid at the higher dose ameliorated liver histopathol. changes due to APAP and properly reversed NF-kB and Nrf2 immunoreactivity. These findings show that sinapic acid pretreatment effectively protects liver against adverse and hepatotoxic effect of APAP through its antioxidant- and anti-inflammatory potential linked to NF-kB/Nrf2/HO-1 signaling and also via regulation of sirtuin 1, mitochondrial integrity, and lysosomal stabilization.

Abstract: This study explored the design of supersaturable self-microemulsifying drug delivery systems (S-SMEDDS) to address poor solubility and oral bioavailability of a novel benzimidazole derivative anticancer drug (BI). Firstly, self-microemulsifying drug delivery systems SMEDDS made of Miglyol 812, Kolliphor RH40, Transcutol HP, and ethanol were prepared and loaded with the BI drug. Upon dispersion, the systems formed neutrally charged droplets of around 20 nm. However, drug precipitation was observed following incubation with simulated gastric fluid (pH 1.2). Aiming at reducing this precipitation and enhancing drug payload, supersaturable systems were then prepared by adding 1% hydroxypropyl cellulose as precipitation inhibitor. Supersaturable systems maintained a higher amount of drug in a supersaturated state in gastric medium compared with conventional formulations and were stable in simulated intestinal medium (pH 6.8). In vitro cell studies using Caco-2 cell line showed that these formulations reduced in a transient manner the transepithelial elec. resistance of the monolayers without toxicity. Accordingly, confocal images revealed that the systems accumulated at tight junctions after a 2 h exposure. In vivo pharmacokinetic studies carried out following oral administration of BI-loaded S-SMEDDS, SMEDDS, and free drug to healthy mice showed that supersaturable systems promoted drug absorption compared with the other formulations. Overall, these data highlight the potential of using the supersaturable approach as an alternative to conventional SMEDDS for improving oral systemic absorption of lipophilic drugs.

(1-aza-12-crown-4)CH2C(CF3)2OH ({RO1F}H), (MeOCH2CH2)2NCH2C(CF3)2OH ({RO2F}H), and (MeOCH2CH2)(Me)NCH2C(CF3)2OH ({RO3F}H) have been synthesized and used to prepare the heteroleptic calcium amido complexes {ROxF}Ca(N(SiMe2R)2) (4–7; x = 2, 3, R = H, Me). The ability to form stable complexes varies with the chelating and electron-donating ability of the aminoether alkoxide ligand, as exemplified by the authors’ failure to isolate cleanly the elusive {RO1F}Ca(N(SiMe2R)2). X-ray diffraction studies show that, in the solid state, intramol. Ca···F interactions help reach coordinative saturation in the dimeric [{RO2F}Ca(N(SiMe3)2)]2 ([4]2), [{RO2F}Ca(N(SiMe2H)2)]2 ([5]2), [{RO3F}Ca(N(SiMe3)2)]2 ([6]2), and [{RO3F}Ca(N(SiMe2H)2)]2 ([7]2), which crystallized free of solvent coligands. Similar stabilizing K···F patterns were found in the polymetallic potassium fluoroalkoxides [{RO1F}K]2 ([1]2), [{RO2F}K]4 ([2]4), and [{RO3F}K]4 ([3]4); [2]4 and [3]4 form heterocubanes in the solid state. Examination of the XRD data for [1]2-[7]2 shows that metal···F interactions can be favored over binding of Oether atoms for calcium and potassium. Pulse-gradient spin-echo NMR spectroscopy shows that the complexes remain aggregated in aromatic solvents. The solvent-free salts [{ROxF}Ca+]·[H2N{B(C6F5)3}2–] (x = 1 (8), 2 (9), 3 (10)) were obtained by treating 4–7 with [H(OEt2)2+]·[H2N{B(C6F5)3}2–] or by reacting Ca(N(SiMe3)2)2 with [{ROxF}HH+]·[H2N{B(C6F5)3}2–]; the solid-state structures of [8·H2O]2 and [9·H2O]2 again showed Ca···F contacts. Complexes 5–7 are promising catalysts for the regiospecific anti-Markovnikov hydrophosphination of styrene with diphenylphosphine, affording TOF values as high as 52 molsubst molCa-1 h-1 with up to 400 equiv of substrates within 1-2 h at 60°.

Jr. research published 《 Solubility of trans-resveratrol in {ethanol (1) + water (2)} mixtures revisited: Correlation, dissolution thermodynamics and preferential solvation》, the research content is summarized as follows. Reported equilibrium mole fraction solubility values of trans-resveratrol in some aqueous-ethanolic mixtures in the temperature interval from (273.2 to 323.2) K were correlated by means of some cosolvency models, like multi-linear models of Jouyban-Acree and Jouyban-Acree-van’t Hoff and non-linear models of the modified Wilson and Buchowski-Ksiazczak. Apparent thermodn. functions of the dissolution processes were computed using the van’t Hoff and Gibbs equations. Gibbs energy and enthalpy were pos. in all cases, while neg. and pos. entropies were observed The plot of enthalpy vs.Gibbs energy of dissolution was non-linear with neg. slopes in the region of 0.00 ≤x1 ≤ 0.60 but pos. slope in the region of 0.60 ≤ x1 ≤ 0.90. Further, by means of the inverse Kirkwood-Buff integrals is observed that trans-resveratrol is preferentially solvated by water in water-rich mixtures but preferentially solvated by ethanol in the composition interval of 0.24 ≤ x1 ≤ 1.00.

Romdhani, Asma;Osorio, Irmis P.;Martinez, Fleming;Jouyban, Abolghasem;Acree, William E. Jr research published 《 Further calculations on the solubility of trans-resveratrol in (Transcutol + water) mixtures》, the research content is summarized as follows. Ha et al. reported the equilibrium solubility of trans-resveratrol in some {Transcutol (1) + water (2)} mixtures at several temperatures with some numerical correlation and apparent dissolution thermodn. analyze. Nevertheless, no attempt was made to evaluate the effect of mixtures polarity on solubility nor preferential solvation of this compound by the solvent components. In this communication the effect of Hildebrand solubility parameters of mixtures on trans-resveratrol solubility was discussed. Moreover, the preferential solvation of trans-resveratrol in {Transcutol (1) + water (2)} mixtures at several temperatures was analyzed based on the inverse Kirkwood-Buff integrals as described in the literature. trans-resveratrol is preferentially solvated by water in water-rich mixtures (0.00 < x1 < 0.14) but preferentially solvated by Transcutol in mixtures with composition 0.14 < x1 < 1.00. These results could be interpreted as a consequence of hydrophobic hydration around the aromatic rings of the solute in the former case. The observed trend in the last case could be a consequence of the acid behavior of trans-resveratrol in front of Transcutol mols. because this cosolvent is more basic than water.

Ulcerative colitis and Crohn′s disease are forms of inflammatory bowel disease whose incidence and prevalence are increasing worldwide. These diseases lead to chronic inflammation of the gastrointestinal tract as a result of an abnormal response of the immune system. Recent studies positioned Cortistatin, which shows low stability in plasma, as a candidate for IBD treatment. Here, using NMR structural information, we design five Cortistatin analogs adopting selected native Cortistatin conformations in solution One of them, A5, preserves the anti-inflammatory and immunomodulatory activities of Cortistatin in vitro and in mouse models of the disease. Addnl., A5 displays an increased half-life in serum and a unique receptor binding profile, thereby overcoming the limitations of the native Cortistatin as a therapeutic agent. This study provides an efficient approach to the rational design of Cortistatin analogs and opens up new possibilities for the treatment of patients that fail to respond to other therapies.
