(HA-SA) tend to self-assemble into polydisperse colloid particles and the particle properties of HA-SA, such as particle size, zeta potential as well as the interfacial activity were studied in detail. The results indicate that the properties of HA-SA particles prepared with different esterification reaction time are quite different due to the different graft amounts of SA. The graft amount of SA increased with the prolongation of esterification reaction time, and the interfacial activity of the obtained HA-SA particles also increased gradually, but an excessively high graft amount would make the polymer too hydrophobic and thus lead to the interfacial activity reduce slightly. Most of the isatin derivatives were proved to be excellent in the inhibition of aldose reductase (ALR2) with IC50 values at submicromolar level, and (E)-2-(5-(4-methoxystyryl)-2,3-dioxoindolin-1-yl) acetic acid (9g) was identified as the most effective with an IC50 value of 0.015 μM. Moreover, compounds 9a-h with styryl side chains at the C5 position of isatin showed potent antioxidant activity. Particularly, the phenolic compound 9h demonstrated similar antioxidant activity with the well-known antioxidant Trolox. The growing epidemic of diabetes means that there is a need for therapies that are more efficacious, safe, and convenient. Here, we report the efficient synthesis of a novel disulfide dimer of human insulin tethered at the N-terminus of its B-chain through placement of a cysteine residue. The resulting peptide was shown to bind to both the insulin receptor isoform B and insulin-like growth factor-1 receptor with comparable affinity to native insulin. In in vivo insulin tolerance tests, the dimer was equipotent to Actrapid insulin and possessed a sustained duration of action greater than that of Actrapid and Glargine. While the secondary structure of our dimeric insulin was similar to that of insulin, it was more resistant to proteolysis. More importantly, our analog was produced in quant. yield from a monomeric thiol insulin scaffold. Our results suggest that this dimer has significant potential to address the clin. needs in the treatment of diabetes.
