Development of well-defined multivalent systems with densely packed multiple functional groups located within a single mol. structure provides an excellent opportunity to generate catalysts of enhanced activity. A series of mono-, di- and trinuclear Pd(II) complexes based on polyketonate ligands allied with 2,2′-bipyridine was designed, synthesized and fully characterized in both solution and solid state. The mono-, di- and tritopic β-diketonate mols. serve as scaffolds for Pd(II) active sites which, in the two latter cases, are forced into close proximity. Application of the complexes as catalysts in Suzuki-Miyaura cross-coupling as a model reaction revealed significant differences in reaction yields and a trend in reactivity reflecting their nuclearity.

Kuno, Goshi;Sakaguchi, Kota;Matsumoto, Akikazu research published Colloidal Crystal Thin Films with Square Lattice Nanoprotrusions Formed by Self-Assembly via Spin-Coating and Heating the research content is summarized as follows. Colloidal crystals with non-close-packed structures, such as body-centered cubic (bcc) structures, are of high interest. Nanosized protrusions, prepared from colloidal crystals, are interesting for biomimetic applications. Herein, self-assembly approaches for synthesizing colloidal crystals with square lattice nanoprotrusions are presented. In the first approach, spin-coating and subsequent heating of colloidal silica and acrylic monomer results in a bcc-like arrangement; the number of stacked colloid layers and the colloid d. of these compounds are important in the formation of this structure. Moreover, prolonged heating exposes colloids from acrylic monomer, with square lattice nanoprotrusions formed. In the second approach, the protrusions are fabricated in a shorter time by replacing some of the acrylic monomer with volatile media. Only certain volatile media, such as a solvent with a structure similar to diethylene glycol, exhibit the formation of square lattice protrusions. These bottom-up approaches contribute toward high-throughput techniques for fabricating metastable square lattice colloidal arrangements.

Fisetin is a polyphenolic flavonoid reported to have antioxidant, anti-inflammatory, and anti-cancer activities. However, it loses its importance as an effective phytochem. due to its poor water solubility and lower bioavailability. In the present study, the self-nanoemulsifying drug delivery system (SNEDDS) of fisetin was developed in order to improve its pharmacol. activity. The developed SNEDDS of fisetin was evaluated for improving the rotenone-induced behavioral changes in the rats, and its efficacy was compared with naive fisetin. It was noticed that fisetin loaded in the SNEDDS formulation significantly (p < 0.001) ameliorated the rotenone-induced alteration in the body weight, grip strength, beam walk, postural instability, etc., in rats when compared to the effect of naive fisetin. Naive fisetin significantly (p < 0.05) ameliorated the effect of rotenone on the level of dopamine only at a higher dose. Whereas, SNEDDS of fisetin produced a significant (p < 0.05) effect at both dose levels when compared with the diseased group as well as also produced a significant (p < 0.05) effect when compared with the naive fisetin group. The results of histopathol. examination revealed about the neuroprotective effect of SNEDDS loaded with fisetin as observed through the protection of neuronal damage. From this study, it was concluded that SNEDDS improved the anti-Parkinsonian activity of fisetin by improving the behavioral alteration produced by rotenone due to enhancement in its solubility and bioavailability.

Kuleshova, Olena;Asako, Sobi;Ilies, Laurean research published Ligand-Enabled, Iridium-Catalyzed ortho-Borylation of Fluoroarenes the research content is summarized as follows. A terpyridine derivative and an iridium complex catalyze the C-H borylation of a stoichiometric amount of a fluoroarene with high ortho-selectivity and tolerance of functional groups such as bromide, chloride, ester, ketone, amine, and in situ-borylated hydroxyl. Complex drug mols. such as haloperidol can be selectively borylated ortho to the F atom. The terpyridine ligand undergoes rollover cyclometalation to produce an N,N,C-coordinated iridium complex, which may either selectively borylate the fluoroarene by itself or undergo reductive elimination to produce a borylated ligand.

Name: 3,5-Dimethoxy-4-hydroxycinnamic acid, Sinapinic acid is a chemical compound that is the dihydroxybenzoic acid derivative of sinapic acid. It has been shown to have anti-inflammatory properties in vitro and in vivo. Sinapinic acid inhibits the activity of various enzymes, such as cyclooxygenase (COX), lipoxygenase (LOX), and 5-lipoxygenase-activating protein (FLAP). It also decreases levels of adhesion molecules and downregulates inflammatory response genes. Sinapinic acid has been shown to reduce inflammation by inhibiting the formation of proinflammatory mediators, such as prostaglandin E2 or leukotriene B4, in endothelial cells and mammary epithelial cells.
Sinapic acid is a phenylpropanoid hydroxycinnamic acid with diverse biological activities. Sinapic acid inhibits collagen-induced human platelet aggregation by up to 70% in vitro (IC50 = 1.03 mM). It scavenges 2,2-diphenyl-1-picrylhydrazyl (DPPH; ) and 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonate) (ABTS) free radicals with IC50 values of 8.3 and 5.4 μg/ml, respectively. Sinapic acid (200 μM) reduces colony formation of SW480 human colon carcinoma cells by 4-fold. It also inhibits colony formation of E. coli, S. enteritidis, and S. aureus on agar (MICs = 2.2, 2, and 1.8 mM, respectively). In vivo, sinapic acid (4 mg/kg, p.o.) increases the time spent in the open arms of the elevated plus maze by approximately 15% in mice, an effect that can be blocked by the GABAA receptor antagonists flumazenil and bicuculline. Sinapic acid is also commonly used as a matrix in protein mass spectrometry.
Sinapic acid analytical standard provided with w/w absolute assay, to be used for quantitative titration.
Sinapic acid is an hydroxycinnamic acid derivative that occurs naturally in Brassicaceae species.
cis-Sinapic acid, also known as cis-sinapate or synapitic acid, belongs to the class of organic compounds known as hydroxycinnamic acids. Hydroxycinnamic acids are compounds containing an cinnamic acid where the benzene ring is hydroxylated. cis-Sinapic acid is considered to be a practically insoluble (in water) and relatively neutral molecule. Within the cell, cis-sinapic acid is primarily located in the cytoplasm. Outside of the human body, cis-sinapic acid can be found in common pea and pulses. This makes cis-sinapic acid a potential biomarker for the consumption of these food products.
Cis-sinapic acid is a 3-(4-hydroxy-3,5-dimethoxyphenyl)prop-2-enoic acid in which the double bond has cis-configuration. It has been isolated from the shoots of alfalfa. It has a role as a plant metabolite., 530-59-6.

Plants have served for centuries as sources of compounds useful for human health such as antioxidant, anti-diabetic and antitumor agents. They are also rich in nutrients that improve the human diet. Growing demands for these compounds make it important to seek new sources for them. Hippophae rhamnoides L. is known as a plant with health-promoting properties. In this study we investigated the chem. composition and biol. properties of bioactive components of ethanol extracts from leaves and twigs of H. rhamnoides L. Chem. components such as the total content of phenolic compounds, vitamins and amino acids and the antioxidant activities of these compounds in cellular and cell-free systems were assessed. The results suggest that the studied extracts are rich in bioactive compounds with potent antioxidant properties. Cytotoxicity and hemotoxicity assays showed that the extracts had low toxicity on human cells over the range of concentrations tested. Interaction with human serum albumin was investigated and conformational changes were observed Our results indicate that leaf and twig extracts of H. rhamnoides L. should be considered as a non-toxic source of bioactive compounds which may be of interest to the food, pharmaceutical and cosmetic industries.

Name: Diethylene Glycol Monoethyl Ether, Diethylene glycol monoethyl ether appears as a colorless, slightly viscous liquid with a mild pleasant odor. Flash point near 190°F. Used to make soaps, dyes, and other chemicals.
Diethylene glycol monoethyl ether is a primary alcohol that is ethanol substituted by a 2-ethoxyethoxy group at position 2. It has a role as a protic solvent. It is a diether, a primary alcohol and a hydroxypolyether. It derives from a diethylene glycol., 111-90-0.

Ethers lack the hydroxyl groups of alcohols. Without the strongly polarized O―H bond, ether molecules cannot engage in hydrogen bonding with each other. 111-90-0, formula is C6H14O3, Name is Diethylene Glycol Monoethyl Ether. Ethers do have nonbonding electron pairs on their oxygen atoms, however, and they can form hydrogen bonds with other molecules (alcohols, amines, etc.) that have O―H or N―H bonds. Name: Diethylene Glycol Monoethyl Ether.

Klupinski, Theodore P.;Moyer, Robert A.;Chen, Po-Hsu Allen;Strozier, Erich D.;Buehler, Stephanie S.;Friedenberg, David A.;Koszowski, Bartosz research published A procedure to detect and identify specific chemicals of potential inhalation toxicity concern in aerosols the research content is summarized as follows. ObjectiveUnderstanding the potential inhalation toxicity of poorly characterized aerosols is challenging both because aerosols may contain numerous chems. and because it is difficult to predict which chems. may present significant inhalation toxicity concerns at the observed levels. We have developed a novel systematic procedure to address these challenges through non-targeted chem. anal. by two-dimensional gas chromatog.-time-of-flight mass spectrometry (GC x GC-TOFMS) and assessment of the results using publicly available toxicity data to prioritize the tentatively identified detected chems. according to potential inhalation toxicity. Materials and MethodsThe procedure involves non-targeted chem. anal. of aerosol samples utilizing GC x GC-TOFMS, which is selected because it is an effective technique for detecting chems. in complex samples and assigning tentative identities according to the mass spectra. For data evaluation, existing toxicity data (e.g. from the U.S. Environmental Protection Agency CompTox Chems. Dashboard) are used to calculate multiple toxicity metrics that can be compared among the tentatively identified chems. These metrics include hazard quotient, incremental lifetime cancer risk, and metrics analogous to hazard quotient that we designated as exposure-(toxicol. endpoint) ratios. Results and DiscussionWe demonstrated the utility of our procedure by detecting, identifying, and prioritizing specific chems. of potential inhalation toxicity concern in the mainstream smoke generated from the machine-smoking of marijuana blunts. ConclusionBy designing a systematic approach for detecting and identifying numerous chems. in complex aerosol samples and prioritizing the chems. in relation to different inhalation toxicol. endpoints, we have developed an effective approach to elucidate the potential inhalation toxicity of aerosols.

Kloevekorn, Philip;Pfaffenrot, Bent;Juchum, Michael;Selig, Roland;Albrecht, Wolfgang;Zender, Lars;Laufer, Stefan A. research published From off-to on-target: New BRAF-inhibitor-template-derived compounds selectively targeting mitogen activated protein kinase kinase 4 (MKK4) the research content is summarized as follows. The mitogen-activated protein kinase (MAP) kinase 4 (MKK4) was found to be a major regulator of liver regeneration and could be a valuable drug target addressing liver related diseases by restoring its intrinsic regenerative capacity. We report on the synthesis and optimization of novel MKK4 inhibitors following a target-hopping strategy from the FDA-approved BRAFV600E inhibitor PLX4032 (8, I). Applying an iterative multi-parameter optimization process we carved out essential structural features yielding in compounds with a low nanomolar affinity for MKK4 and excellent selectivity profiles against the main off-targets MKK7 and JNK1, which, upon relevant inhibition, would totally abrogate the pro-regenerative effect of MKK4 inhibition, as well as against the off-targets MAP4K5, ZAK and BRAF with selectivity factors ranging from 40 to 430 for our best-balanced compounds 70 and 73 (II and III, resp.).
