The rationale of this study was to investigate the role of Acacia jacquemontii Et acetate extract (AJEAE) in the downregulation of hyperglycemia. The current study was performed in two parts, in vitro, through characterization (high-performance liquid chromatog.), estimation of total phenolic content, total flavonoid content, antioxidant (2,2-diphenyl-1-picrylhydrazylassay), and α-amylase inhibitory activities of the studied extract, and in vivo using Wistar rats in which animals were divided into five groups NC, DC, GL, AJEAE 250 mg/kg, and AJEAE 500 mg/kg. The effects of AJEAE on fasting plasma glucose, plasma insulin, HOMA-IR, oral glucose tolerance test, glycated Hb (HBA1c), lipid profile, inflammatory cytokines (Interleukin-6, tumor necrosis factor-alpha), and oxidative stress markers (lipid peroxidation, nitic oxide, superoxide dismutase, catalase, glutathione peroxidase) were evaluated. Our findings confirmed the presence of quercetin, kaempferol, gallic acid, vanillic acid, syringic acid, M-coumaric acid, sinapic acid, chlorogenic acid, cinnamic acid, and ferulic acid in AJEAE. The VAFC was formulated using P 50% + B 50% (F1), P 45%+B 45%+LCT 10% (F2), and P 45%+B 45%+LCT 8%+Vita E 2% (F3). Revealed that the soly of the VAFC was significantly increased in F3 formulation. Likewise, total phenolic content, including single phenolic acids, total flavonoid including rutin and quercetin, total anthocyanin including single anthocyanins, and antioxidant capacity of the VAFC were significantly increased in F3 formulation. The developed QCT-AgNPs hydrogel was characterized for hydrodynamic diameter, %entrapment efficiency (%EE), surface morphol., texture anal., in-vitro drug release, skin irritation study, ex-vivo permeation study (confocal study), and antimicrobial efficacy. The optimized formulation showed hydrodynamic diameter of ∼44.1 nm with smooth spherical surface morphol. and ∼92.09% of QCT was entrapped in QCT-AgNPs hydrogel matrixes. The antimicrobial study revealed superior therapeutic efficacy of QCT-AgNPs hydrogel in comparison to marketed (MRKT) gel on S. aureus and E. coli. Moreover, in-vivo results demonstrated that QCT-AgNPs hydrogel significantly (p < 0.001) reduced the wound gap and increased % re-epithelialization compared with diabetic control after 18 d of post treatment in excisional diabetic wound model.
