Constitutive androstane receptor (CAR) activation has found to ameliorate diabetes in animal models. However, no CAR agonists are available clinically. Therefore, a safe and effective CAR activator would be an alternative option. In this study, sixty courmarin derivatives either synthesized or purified from Artemisia capillaris were screened for CAR activation activity. Chemical modifications were on position 5,6,7,8 with mono-, di-, tri-, or tetra-substitutions. Among all the compounds subjected for in vitro CAR activation screening, 6,7-diprenoxycoumarin was the most effective and was selected for further preclinical studies. Chemical modification on the 6 position and unsaturated chains were generally beneficial. Electron-withdrawn groups as well as long unsaturated chains were hazardous to the activity. Mechanism of action studies showed that CAR activation of 6,7-diprenoxycoumarin might be through the inhibition of EGFR signaling and upregulating PP2Ac methylation. To sum up, modification mimicking natural occurring coumarins shed light on CAR studies and the established screening system provides a rapid method for the discovery and development of CAR activators. In addition, one CAR activator, scoparone, did showed anti-diabetes effect in db/db mice without elevation of insulin levels.

Aryl-aryl cross-coupling constitutes one of the most widely used procedures for the synthesis of high-value materials that range from pharmaceuticals to organic electronics and conducting polymers. The assembly of (hetero)biaryl scaffolds generally requires multiple steps; coupling partners must be functionalized before the key bond-forming event is considered. The development of selective C-H arylation processes in arenes-which sidestep the need for prefunctionalized partners-is thus crucial for streamlining the construction of these key architectures. Here we report an expedient, one-pot assembly of (hetero)biaryl motifs using photocatalysis and two non-prefunctionalized arene partners. The approach is underpinned by the functionalization of a C-H bond in an arene coupling partner using the interrupted Pummerer reaction. A unique pairing of the organic photoredox catalyst and the intermediate dibenzothiophenium salts enables highly selective reduction in the presence of sensitive functionalities. The utility of the metal-free, one-pot strategy is exemplified by the synthesis of a bioactive natural product and the modification of complex molecules of societal importance.

Investigating environmental pollution is important to understand its impact on endangered species such as green turtles (Chelonia mydas). In this study, we investigated the accumulation and potential toxicity of selected persistent organic pollutants (POPs) and naturally occurring MeO-PBDEs in liver, fat, kidney and muscle of turtles (n = 30) of different gender, size, year of death, location and health status. Overall, POP concentrations were low and accumulation was highest in liver and lowest in fat which is likely due to the poor health of several animals, causing a remobilization of lipids and associated compounds. PCBs and p,p’-DDE dominated the POP profiles, and relatively high MeO-PBDE concentrations (2′-MeO-BDE 68 up to 192 ng/g lw, 6-MeO-BDE 47 up to 79 ng/g lw) were detected in all tissues. Only few influences of factors such as age, gender and location were found. While concentrations were low compared to other marine wildlife, biological toxicity equivalences obtained by screening the tissue extracts using the micro-EROD assay ranged from 2.8 to 356 pg/g and the highest values were observed in muscle, followed by kidney and liver. This emphazises that pollutant mixtures found in the turtles have the potential to cause dioxin-like effects in these animals and that dioxin-like compounds should not be overlooked in future studies. Human nails have been increasingly used as a biomarker for human exposure to persistent organic pollutants (POPs). In the present study, the fingernails of e-waste-dismantling workers from Longtang town, Qingyuan city, rural residents from Shijiao town, Qingyuan city, and urban residents from Guangzhou city, respectively, were collected from South China to monitor the human burdens of polybrominated diphenyl ether (PBDEs) and polychlorinated biphenyl (PCBs). The median concentrations of Sigma PBDEs in the nails of the e-waste-dismantling workers, and urban and rural residents were 412, 129, and 82.1 ng g(-1), respectively, and the median concentrations of Sigma(20)PCBs were 108, 8.4, and 22.1 ng g(-1), respectively. The levels of PCBs and PBDEs in the nails of e-waste-dismantling workers were significantly higher as compared to those for urban and rural residents (p< 0.05), implying the continuous and greater exposure to these chemicals in the e-waste recycling areas. BDE 209 (92-98%) was the major congener of PBDEs and CB 52 (26-51%) was the main congener of PCB in nail samples. However, no significant gender difference was observed for PBDE and PCB levels in nails from all three investigated areas, and no significant correlation was found between their levels and the age of the participants. The enantiomer fractions (EFs) of CBs 95 and 132 indicated that the external sources (e.g.dust and/or air) were the primary sources for CBs 95 and 132 in human nails from the e-waste area, while the contribution from the internal sources (e.g.serum) could be in a small percentage. The results of this study indicate that human nails can be used as a proper indicator of human exposure to PCBs and PBDEs, and further studies are needed by a comprehensive investigation of the relationships between the PCB and PBDE levels in the nails and serum and/or other internal tissues. In-situ Raman spectroelectrochemistry is a powerful tool to understand the structures and reaction mechanisms of electrochemical interfaces yet usually suffers from a low sensitivity and detection efficiency. Herein, we develop a windowless thin layer electrochemical cell design to improve the detection efficiency of in-situ Raman spectroscopy for practical nanoscale electrocatalysts. This was achieved by perforating a hole in the optical window of the cell, which not only eliminates the refraction of light by the window but also induces a meniscus thin electrolyte layer on the working electrode, thus significantly improving the detection efficiency of Raman spectra. We successfully applied thismethod to conduct in-situ Raman spectroscopy of structural evolutions of two representative Ni-Fe oxide catalysts (layered double hydroxides and spinel oxides) during oxygen evolution reaction (OER) electrocatalysis. It was shown that both catalysts underwent surface transformation to oxyhydroxides and formed active oxygen species at OER relevant potentials. However, the extent of the surface transformation was much lower on the Ni-Fe spinel catalyst, accounting for its lower OER activity. We further show that adding a carbon support can promote the surface transformation of Ni-Fe spinel to oxyhydroxides, thus significantly increasing the electrocatalytic activities. The established windowless thin layer method provides a new way to conduct in-situ electrochemical Raman spectroscopy of a wide range of materials and may be also combined with surface-enhanced Raman spectroscopic methods.

Introduction: Gestational exposure to chemical mixtures, which is prevalent among pregnant women, may be associated with adverse childhood neurodevelopment. However, few studies have examined relations between gestational chemical mixture exposure and children’s cognitive abilities. Methods: In a cohort of 253 pregnant women and their children from Cincinnati, OH (enrolled 2003-2006), we quantified biomarker concentrations of 43 metals, phthalates, phenols, polybrominated diphenyl ethers, organophosphate and organochlorine pesticides, polychlorinated biphenyls, perfluoroalkyl substances, and environmental tobacco smoke in blood or urine. Using k-means clustering and principal component (PC) analysis, we characterized chemical mixtures among pregnant women. We assessed children’s cognitive abilities using the Wechsler Preschool and Primary Scale of Intelligence-III and Wechsler Intelligence Scale for ChildrenIV at ages 5 and 8 years, respectively. We estimated covariate-adjusted differences in children’s cognitive ability scores]=cross clusters, and with increasing PC scores and individual biomarker concentrations. Results: Geometric mean biomarker concentrations were generally highest, intermediate, and lowest among women in clusters 1, 2, and 3, respectively. Children born to women in clusters 1 and 2 had 5.1 (95% CI: 9.4,-0.8) and 2.0 (95% CI: 5.5, 1,4) lower performance IQ scores compared to children in cluster 3, respectively. PC scores and individual chemical biomarker concentrations were not associated with cognitive abilities. Conclusions: In this cohort, combined prenatal exposure to phenols, certain phthalates, pesticides, and perfluoroalkyl substances was inversely associated with children’s cognition, but some individual chemical biomarker concentrations were not. Additional studies should determine if the aggregate impact of these chemicals on cognition is different from their individual effects.

The dependence of polymer film transparency on different kinds of planar orientations of nanoporous-crystalline (NC) phases of syndiotactic polystyrene (sPS) and poly(2,6-dimethyl-1,4-phenylene)oxide (PPO) is described. The two polymers exhibit opposite behavior: film transparency is improved for sPS and PPO by orientations of the chain axes of the NC phases being parallel (c(parallel to)) and perpendicular (c(perpendicular to)) to the film plane, respectively. This behavior can be rationalized by the negative and positive birefringence of sPS and PPO chains, respectively. In fact, to maximize transparency, the refractive index of the NC phase along the perpendicular to the film plane has to be increased to come closer to that of the corresponding amorphous phase. This can be pursued by controlling the orientation of the NC phases and hence of the associated refractive index ellipsoids.

Studies of environmentally benign catalytic methods are of great value in modern chemical synthesis, especially the chemo-selective construction of chemical bonds under green conditions. This work elucidates such preferential synthesis of C-N bond over C-O bond via selective N-arylation of 3-aminophenols using 1,3-bis-[2-hydroxyphenyl] imidazolium chloride (IHPHCl) and copper iodide as catalyst (1 mol %) in aqueous medium. Presence of chelating group (-OH) on IHPHCl enhances N-selectivity. Overall this is a simple and green method for selective N-arylation of 3-aminophenols with good substrate scope and yields (60-88 %). GC-MS, HRMS and other spectroscopic techniques were utilised in detailing the kinetics and mechanistic aspects.

Marine plastic debris, including microplastics (<5 mm in size), comprises a suite of chemical ingredients and sorbed chemical contaminants. Thus, microplastics are a potential, and debated, source of anthropogenic chemicals for bioaccumulation and biomagnification. Several studies have investigated the role of microplastics as a vector of contaminants to marine organisms via modeling exercises, laboratory experiments, and field studies. Here, we examined relationships among chemical contaminants and microplastics in lanternfish (family Myctophidae), an important link in marine food webs, from the North Pacific Ocean as a case study from the field. We compared the body burden of several chemical groups (bisphenol A [BPA], nonylphenol [4-NP], octylphenol [4n-OP], alkylphenol ethoxylates [APEs], pesticides, polychlorinated biphenyls [PCBs], and polybrominated diphenyl ethers [PBDEs]) in fish caught within and outside the North Pacific Subtropical Gyre where plastic is known to accumulate. We also tested whether there was a relationship between chemical concentrations in fish and plastic density at each sampling location. Mean concentrations of common plastic constituents (BPA, 4-NP, 4n-OP, APEs, and total PBDEs) were comparable between myctophids collected within and outside the North Pacific Gyre. Pesticides were higher in lanternfish caught outside the gyre and were associated with lower plastic density. Total PCBs were also higher in fish outside the gyre. In contrast, lower chlorinated PCB congeners were higher in fish residing in the accumulation zone and were correlated with higher plastic density. This finding is consistent with other studies demonstrating an association between lower chlorinated PCBs and plastics in biota and suggests that microplastic may be a transport mechanism for some chemicals in nature. Hierarchically porous P substituted HZSM-5 (H[P, Al]-ZSM-5-AT-0.05 M ) was successfully synthesized by isomorphous substitution method with introduced P atoms into ZSM-5 zeolite frameworks followed by alkali treatment. H[P, Al]-ZSM-5-AT-0.05 M was characterized by XRD, ICP-OES, SEM, FT-IR, N-2 adsorption-desorption, NH3-TPD, XPS, Py-IR, and TG-DTA, respectively. The surface area, pore volume, particle size and P/Al (molar ratio) are 328 m(2).g(-1), 0.23 cm(3).g(-1), 563 nm and 0.027, respectively. The catalytic performance of H[P, Al]-ZSM-5-AT-0.05 M in coupling reaction of methanol with 1-butene was investigated in the fixed-bed reactor. On the condition of reaction temperature at 500 degrees C under atmosphere pressure, the main product is propylene with the selectivity and yield of 33.7% and 31.0%, respectively. Hydrogen transfer index was calculated to 0.24. Moreover, its diffusion limitation was investigated with the self-etherification of benzyl alcohol reaction. The value of Thiele modulus (eta) and effectiveness factor (phi) are 0.08 and 16.9, respectively.
