Psoralen is a furocoumarin natural product that intercalates within DNA and forms covalent adducts when activated by ultraviolet radiation. It is well known that this property contributes to psoralen’s clinical efficacy in several disease contexts, which include vitiligo, psoriasis, graft-versus-host disease and cutaneous T-cell lymphoma. Given the therapeutic relevance of psoralen and its derivatives, we attempted to synthesize psoralens with even greater potency. In this study, we report a library of 73 novel psoralens, the largest collection of its kind. When screened for the ability to reduce cell proliferation, we identified two derivatives even more cytotoxic than 4 ‘-aminomethyl-4,5 ‘,8-trimethylpsoralen (AMT), one of the most potent psoralens identified to date. Using MALDI-TOF MS, we studied the DNA adduct formation for a subset of novel psoralens and found that in most cases enhanced DNA binding correlated well with cytotoxicity. Generally, our most potent derivatives contain positively charged substituents, which we believe increase DNA affinity and enhance psoralen intercalation. Thus, we provide a rational approach to guide efforts toward further optimizing psoralens to fully capitalize on this drug class’ therapeutic potential. Finally, the structure-activity insights we have gained shed light on several opportunities to study currently underappreciated aspects of psoralen’s mechanism.

Cleavage of ether bonds is a crucial but challenging step for lignin valorization. To efficiently realize this transformation, the development of robust catalysts or catalytic systems is required. In this study, montmorillonite (MMT)-supported Ru (denoted as Ru/MMT) is fabricated as a dual-functional heterogeneous catalyst to cleave various types of ether bonds through transfer hydrogenolysis without using any additional acids or bases. The prepared Ru/MMT material is found to efficiently catalyze the cleavage of various lignin models and lignin-derived phenols; cyclohexanes (fuels) and cyclohexanols (key intermediates) are the main products. The synergistic effect between electron-enriched Ru and the acidic sites on MMT contributes to the excellent performance of Ru/MMT. Systematic studies reveal that the reaction proceeds through two possible reaction pathways, including the direct cleavage of ether bonds and the formation of intermediates with one hydrogenated benzene ring, for all examined types of ether bonds, namely, 4-O-5, alpha-O-4, and beta-O-4.

Cleavage of C-O bonds in aryl ether through the catalytic transfer hydrogenolysis (CTH) is an encouraging method for the valorization of lignin into fine chemicals and liquid fuels. In this contribution, the CTH of typical lignin alpha-O-4 model compound 4-(benzyloxy)phenol (BOP) and lignin extracted from ash wood have been explored over 0.45 wt% Pt/HNbWO6/CNTs catalyst. The effect of solvents was elaborated and the highest catalytic activity was achieved in the CTH of BOP using isopropanol as the solvent (98.1%). This was because isopropanol can provide more hydrogen atoms to promote CTH reaction due to the weakest interaction between isopropanol and BOP among all H-donor solvents. The favorable results obtained in the model compound study inspired us to afford Pt/HNbWO6/CNTs catalyst in the CTH of lignin. The relative oxygen content of lignin was sharply decreased after the CTH process, and the higher heating value (HHV) significantly increased from 21.4 MJ/kg to 30.8 MJ/kg. GPC, FT-IR, GC-MS, and 2D-NMR results revealed that lignin was successfully depolymerized into small molecule compounds over Pt/HNbWO6/CNTs catalyst through the CTH process. Thereinto, phenolic compounds were the major components in the liquid products in which most of them were monophenols. The results obtained in this work shed light on that Pt/HNbWO6/CNTs was a favorable catalyst in the depolymerization of lignin into valuable phenolic compounds via the CTH method. With the aid of direct heating through microwave irradiation in non-aqueous media, nanocrystalline tungsten(vi) oxide is achievable in 30 minutes at 200 degrees C, faster and at a lower temperature than conventional synthesis methods. Forming in a platelet morphology, these particles are as small as 20 nm with a BET surface area of 37 m(2) g(-1) WO3. These nanoplatelets are active for the photocatalytic oxidation of the 1 degrees alcohols benzyl alcohol (rate constant, k of 2.6 x 10(-3) h(-1)) and 5-(hydroxymethyl)-2-furfural (k of 0.01 h(-1)) using 10 mg of WO3 with 2 mL of 0.250 M substrate in acetonitrile and a 150 mW cm(-2) 460 nm blue LED source. As expected, these rate constants are larger than those observed for commercially prepared, micron-sized WO3. XPS analysis shows that during catalysis, the concentration of W5+ on the surface increases, but the nanoplatelets are stable under these reaction conditions. The overall morphology and size of the particles are retained through the reactions. Moreover, the nanoplatelets are recyclable-showing no loss in activity for four reaction cycles.

Electrocatalytic hydrogenation (ECH) provides a green route to upgrade oxygenated bio-oil under mild conditions, but is still challenged with the issues of low working current density (<60 mA cm(-2)) and low faradaic efficiency (usually 20-60%) that seriously hinder its practical applications. Herein, we present a dual-catalyst electrochemical route that achieves extremely high faradaic efficiency (>99% for many chemicals) and high working current density (up to 800 mA cm(-2)) in the hydrogenation of model bio-oil compounds. More importantly, efficient deoxygenation to alkanes, often thought to be very difficult in conventional ECH, was achieved in the aqueous electrolysis. The dual-catalyst system consists of a suspended noble-metal catalyst and soluble polyoxometalate (POM). The theoretical calculations indicate that the POM functions as a superacid, changing the common hydrogenation route to a carbocation mechanism and resulting in effective electrolytic deoxygenation of oxygenates. Because no current flows through the catalyst, even a non-conductive catalyst can be used, which provides a great opportunity for extension to general applications.

Organophosphate esters (OPEs) are commonly used as flame retardants (FRs) and plasticizers. The usage of OPEs has increased recently due to the ban of several brominated flame retardants, but information on levels in the environment, including the indoor environment is still limited. We investigated the occurrence and distribution of 12 OPEs in urban house dust from Vancouver, Canada; Istanbul, Turkey; and Cairo, Egypt. The medianSOPE concentration was 41.4 mu g/g in the Vancouver samples while median levels in Istanbul and Cairo were significantly lower. The median composition profiles of OPEs in Vancouver and Cairo were dominated by tris (2-butoxyethyl) phosphate (TBOEP), accounting for 56 and 92% of total OPEs respectively while it showed a detection frequency of only 14% in Istanbul. Tris (2-chloropropyl) phosphate (TCPP) was the most abundant chlorinated OPE representing 20 and 36% of the total OPEs in Vancouver and Istanbul respectively, but was below the detection limit in the Cairo dust samples. Consistent with other studies, SOPE concentrations were similar to 1 to 2 orders of magnitude higher than PBDEs and currently used flame retardants in the same dust samples. The mean estimated daily intakes (EDI) of SOPE from dust were 115, 38 and 9 ng/kg/bw/day in Vancouver, Cairo and Istanbul respectively for toddlers where adults were similar to 10 times lower. The total toddler OPE intake ranged from 115 to 2900, 38 to 845 and from9 to 240 ng/kg bw/day across the three cities. TBOEP had the largest contribution to the EDI in both toddler and adults, where toddler TBOEP exposures via dust represented 4% to 80%, 2% to 44% and 0.1% to 6% of the Reference Doses (RfD) in the mean and high intake scenarios for toddlers in Vancouver, Cairo and Istanbul respectively. Here, we have successfully synthesised and purified multifunctional PLGA-based nanoparticles by the co-encapsulation of an anticancer drug (tetrandrine) and a magnetic material (Fe3O4). The obtained Tet-Fe3O4-PLGA NPs had a uniform spherical shape with a particle size of approximately 199nm and a negative surface charge of -18.0mV, displaying a high encapsulation efficiency. Furthermore, TEM studies provided representative images of the purification process of the magnetic nanoparticles with MACS((R)) technology. The MFM and VSM results indicated that both the Fe3O4 NPs and Tet-Fe3O4-PLGA NPs were superparamagnetic. The DSC spectrum demonstrated that Tet was successfully encapsulated within the PLGA-based nanoparticles. Significantly, the release studies revealed NPs had a relatively slower release rate than free Tet after 8h’s initial burst release, which had decreased from 98% to 65% after 24h. In vitro cellular studies revealed that NPs could effectively penetrate into A549 cells and A549 multicellular spheroids to exert cytotoxicity, displaying a significantly high anti-proliferation effect. Moreover, western blot demonstrated that the co-loaded NPs had a higher anticancer activity by injuring lysosomes to activate the mitochondria pathway and induce A549 cell apoptosis. The magnetic characteristics and high anticancer activity support the use of Tet/Fe3O4 co-loaded PLGA-based nanoparticles as a promising strategy in the treatment of lung cancer.

We report the synthesis, characterization, and spectroscopic investigations of a new responsive-at-metal cyclometalated platinum(II) complex. With mild chemical oxidants and reductants, it was possible to obtain the same complex in three different oxidation states and each of these complexes was structurally characterized by single-crystal X-ray diffraction. We discovered that the platinum(II) complex displays strong solvatochromism in the solid state, which can be attributed to modulation of Pt…Pt interactions that results in switching between optical and photoluminescent states. Incorporating responsive-at-metal species as dynamic components in nanostructured materials might facilitate response amplification, sensing, actuation, or self-healing processes.
