Characterization of Olfactory Receptor Repertoires in the Endangered Snow Leopard Based on the Chromosome-Level Genome

Olfaction is a complicated process that begins with the specific binding of volatile odorant molecules to dedicated olfactory receptors (ORs) in the olfactory epithelium and plays a pivotal role in the survival of mammals. The OR subgenome of the snow leopard has remained largely unexplored, and thus, investigation of the OR system would shed light on the evolutionary dynamics of the snow leopard OR repertoires and genetic evidence for environmental adaptation. In this study, we conducted genome-wide identification and characterization of OR genes in the snow leopard and compared them to all other Panthera species. A total of 213, 294, 624, 305, and 253 functional OR genes were identified in the snow leopard, lion, jaguar, leopard, and tiger, respectively. The phylogenetic relationships of functional Panthera OR genes were illustrated, which comprised 69 families and 350 subfamilies distributed in two classes (Class I and Class II). Comparative analysis of the five Panthera species indicated 115 shared and 5 snow leopard-specific clusters. The potential odorant specificity of certain snow leopard OR genes was identified by similarities to human protein sequences and we identified odorants such as eugenol methyl ether that had the most OR genes. Since our references for odorants were from human studies, possible odorants from snow leopard-specific OR genes need further investigation. The lowest number of OR genes for the snow leopard among Panthera species possibly revealed the association between OR gene family contraction and high-altitude adaptation, which needed further and deeper investigation. This systematic study of OR genes in the snow leopard will provide a solid foundation for further study of olfactory function and variation in the snow leopard.

A low-volume air sampling method for legacy and novel brominated flame retardants in indoor environment using a newly developed sorbent mixture

Brominated flame retardants (BFRs) are a series of stable and outstanding flame retardants bringing human exposure risks in indoor environment. However, sampling methods now available for BFRs are solvent-consuming and relatively complicated. This study provides a new option of low-volume air sampling device using cartridges with a sorbent mixture for different types of legacy and novel BFRs. In this study, we found that HC-C-18 sorbent is most suitable for polybrominated diphenyl ethers (PBDEs) and novel BFRs (NBFRs) enrichment, and that NH2 for hexabromocyclododecanes (HBCDs). The sorbent mixture was optimized using a complex of HC-C-18 and NH2 sorbents with elution recovery of 69.4% +/- 7.9-117% +/- 10%, pumping-through recovery of 84.5% +/- 7.9-127% +/- 36%, and breakthrough recovery of 70.8% +/- 3.4-118% +/- 6% for PBDEs, NBFRs, and HBCDs in indoor air. A sequential elution was also achieved using hexane for PBDEs and NBFRs and ethyl acetate for HBCDs. The method was validated with field sampling at nine student dormitory rooms. For legacy BFRs, all the isomers of HBCDs were detected in the air of nine rooms with the median concentrations of 91, 33, and 25 pg/m(3) for (+/-)alpha-HBCD, (+/-)beta-HBCD, (+/-)gamma-HBCD, respectively, while PBDEs were hardly detected. In contrast, NBFRs were detected at total concentrations of 15-811 pg/m(3). Pentabromotoluene (PBT) was the most frequently detected NBFRs with a median concentration of 4 pg/m(3), followed by EHTBB at 56 pg/m(3) and HBBZ at 21 pg/m(3). For the risk assessment, the total hazard index value for air inhalation of BFRs was estimated at 6.1=10(-4)-0.35, which are consistently lower than 1, indicating no immediate health risk, while their long-term effects remain worth concerns.

Effects of SiO2 nanoparticles on the uptake of tetrabromobisphenol A and its impact on the thyroid endocrine system in zebrafish larvae

The coexistence of nanoparticles and organic toxicants in the environment modifies pollutant bioavailability and toxicity. This study investigated the influence of silicon dioxide nanoparticles (n-SiO2) on the uptake of tetrabromobisphenol A (TBBPA) and its impact on the thyroid endocrine system in zebrafish larvae. Zebrafish (Danio rerio) embryos were exposed to TBBPA at different concentrations (50, 100, and 200 mu g/L) alone or in combination with n-SiO2 (25 mg/L) until 120 h post-fertilization (hpf). Chemical measurements showed that both TBBPA and n-SiO2 were bioconcentrated in zebrafish larvae, and the uptake of TBBPA was enhanced by n-SiO2. Furthermore, zebrafish larvae exposed to 200 mu g/L TBBPA alone exhibited significantly increased T4 contents and decreased T3 contents, whereas n-SiO2 treatment alone did not have a detectable effect. Furthermore, the thyroid hormone levels changed more upon treatment with 200 mu g/L TBBPA combined with 25 mg/L n-SiO2 than upon TBBPA treatment alone. Alterations in gene transcription along the related hypothalamic-pituitary-thyroid (HPT) axis were observed, and expression of the binding and transport protein transthyretin (TTR) was significantly decreased for both TBBPA alone and co-exposure with n-SiO2. Thus, the current study demonstrates that n-SiO2, even at the nontoxic concentrations, increases thyroid hormone disruption in zebrafish larvae co-exposed to TBBPA by promoting its bioaccumulation and bioavailability.

Solvent-free functionalization of graphene oxide powder and paper with aminobenzo-crown ethers and complexation with alkali metal cations

Graphene oxide powder and graphene oxide paper were covalently functionalized with the 4′-aminobenzo-15-crown-5 (AB15C5) and 4′-aminobenzo-18-crown-6 (AB18C6) ethers by a solvent-free gas-phase treatment. Then, we attempted the complexation with potassium and sodium cations. A comparative characterization in the functionalized graphene oxide structures was carried out using Fourier-transform infrared (FTIR), Raman and X-ray photoelectron spectroscopy (XPS), thermogravimetric and differential thermal analysis (TGA and DTA, respectively), scanning electron microscopy (SEM), and atomic force microscopy (AFM). The wettability of graphene oxide paper (GOP) samples was analyzed by measuring the contact angle. The degree of covalent functionalization in GO powder with aminobenzo-crown ethers was estimated at 27% (AB15C5) and 19% (AB18C6). The amount of organics attached to GOP samples was approximately 7%. After the interaction with potassium and sodium cations, the functionalized GOP samples showed an increase in the contact angle, followed by a decrease in surface free energy. The complexation with alkali metal cations suggests an uptake of about 3%. However, the cation selectivity for alkali metal ions was dependent on the ions’ radius. Surface free energy measurements suggest that complexation with potassium effectively takes place when using AB18C6.

A highly sensitive and selective electrochemical chiral sensor was developed based on the competitive supramolecular interaction of carbon nanofibers (CNFs) embedded sulfobutyl ether-b-cyclodextrin (SBE-beta-CD) with optically active cationic drugs at glassy carbon electrode (GCE). The difference in intermolecular hydrogen bonding/stability constant/enantioselectivity coefficient and Gibbs free energy of anionic host SBE-b-CD with enantiomers of amlodipine (R-/S-AML), clenbuterol (R-/S-CBL) and metoprolol (R-/S-MET) as a guest paved the way for efficient discrimination. The proposed sensing platform (CNFs-SBE-beta-CD/GCE) could recognize the aforementioned enantiomers based on the discernible difference of peak potential (S/R-AML (Delta E-p = 135 mV), R/S-MET (Delta E-p = 99 mV) and R/S-CBL (Delta E-p = 111 mV). The binding mechanisms and thermodynamic study of the enantiospecific behavior have been investigated using the host-guest chemistry approach inside the nanocavity and results suggest that S-AML, RMET, and R-CBL show stronger stability constants than their antipodes. Formation of the diastereomeric complex was taken as a measure of enantioselectivity and experimental results indicated that anionic SBE-b-CD is a better chiral ligand than neutral cyclodextrins. The fabricated sensor could be a useful low-cost electrochemical tool for molecular recognition of a variety of cationic species not only of drugs but also from other sources. Pressurized extraction of high-quality blackberry (Rubus spp. Xavante cultivar) seed oils

This study reports the extraction of oil from organic blackberry (Rubus spp. Xavante cultivar) performed using supercritical CO2 (scCO(2)), compressed propane, pi-hexane and petroleum ether. The extractions with compressed propane showed higher extraction yield and extraction rate, when compared to those performed with scCO(2). All the oils obtained represent a rich source of unsaturated fatty acid (around 93 %) composed mainly by linoleic acid. Furthermore, the extraction using scCO(2 )resulted an oil with high total phenolic compounds (113.49 mg GAE 100 g(-1)) and antioxidant activity as given by the ABTS method (2701.74 mu mol TE 100 g(-1)). On the other hand, compressed propane showed a higher efficiency in extracting beta-carotene and the oil obtained with such solvent presented a higher thermal stability (224.70 degrees C), rather than the oil obtained with scCO(2) (198.83 degrees C). Study on the influence of the external conditions and internal components on foam performance in gas recovery

In the lifting process of drainage gas recovery, external conditions and internal components of the foam are complex. We used molecular dynamics simulation to construct a series of fatty alcohol polyoxyethylene ether sulfate (AES) foam models that could represent various foams in different lifting stages from well to ground. The effect of temperature and pressure on foam stability was investigated. We found that there are two main reasons for foam instability in the first-half stage of the lifting process. Firstly, high temperature aggravated the molecule movement; some CH4 molecules entered the foam film and formed a molecular transport channel. Secondly, the interaction between the CH4 phase and condensate oil leads to some alkane molecules entering the gas-liquid interface. Green extraction of perilla volatile organic compounds by pervaporation

Volatile organic compounds (VOCs) present in perilla essential oil are of high interest in medicinal and food processing. In this work, pervaporation was implemented to extract the valuable perilla VOCs from dilute aqueous solutions as a green process. Three representative VOCs of perilla (i.e., limonene, linalool, and perillaldehyde) having different functional groups were selected as model components, and poly(ether-block-amide) (PEBA) and polydimethylsiloxane (PDMS) membranes were prepared for the VOC extraction studies. The influences of operating conditions (i.e., feed concentration and temperature) on the pervaporation performance of the membranes were investigated. In binary VOC/water mixtures, an increase in the feed concentration increased the VOC flux and decreased the separation factor. The VOC flux also increased significantly with temperature, mainly due to an augmented driving force for permeation. The impact of the coupling effects in multicomponent permeation was evaluated by comparing the pervaporation performance of VOCs in binary VOC/water and quaternary VOCs/water systems. Results show that the VOC permeation behavior was affected by the presence of other VOCs, depending on the permeant-permeant and membrane-permeant interactions. Based on pervaporation separation index, the PEBA membrane showed a better overall separation efficiency than the PDMS membrane for the extraction of perilla VOCs. Since pervaporation does not involve any chemical solvents and operates at moderate temperatures, it provides a green process for extracting valuable perilla VOCs.

Fabrication of PBI/SPOSS hybrid high-temperature proton exchange membranes using SPAEK as compatibilizer

Because of the harsh operating circumstances of high temperature over 100 degrees C and strong acidity, it is of great significance to explore some new methods on the developments of high-performance membrane materials using in high-temperature proton exchange membrane fuel cells (HT-PEMFCs). In current work, with the assistance of newly designed sulfonated poly (aryl ether ketone) (SPAEK) compatibilizer, some hydrophilic and acidophilic polyhedral-oligosilsesquioxane nanoparticles bearing sulfuric acid groups (SPOSS) were successfully incorporated into a soluble arylether-type polybenzimidazole (Ph-PBI) matrix via an in situ sol-gel process to obtain a new family of hybrid membranes with an improved overall performance. As a result, these hybrid membranes exhibited enhanced acid absorption ability and doping levels at 120 degrees C, improved acid retention ability and attractive mechanical-dimensional stability. More importantly, the proton conductivity of a phosphoric acid doped hybrid membrane reached 126 mS cm(-1) at 200 degrees C. This membrane was also suitable for membrane electrode assemblies (MEAs), and a maximum power density of 300 mW cm(-2) could be achieved at 160 degrees C without humidity.
