On June 12, 2017, Zhu, Lei; Qi, Xiaotian; Li, Yingzi; Duan, Meng; Zou, Lufeng; Bai, Ruopeng; Lan, Yu published an article.Product Details of 1417036-28-2 The title of the article was Ir(III)/Ir(V) or Ir(I)/Ir(III) Catalytic Cycle? Steric-Effect-Controlled Mechanism for the para-C-H Borylation of Arenes. And the article contained the following:

A potential energy surface for iridium-catalyzed C-H-borylation of aromatic compounds was calculated; while bulky phosphine ligands favor Ir(I)-Ir(III) C-H activation, small amine ligands promote formation of triboryl Ir(V) intermediates. D. functional theory method N12 was used to study the mechanism of the [Ir(cod)OH]2/Xyl-MeO-BIPHEP-catalyzed para-selective C-H borylation reaction. The results revealed that the use of a bulky diphosphine ligand such as Xyl-MeO-BIPHEP was unfavorable for the previously proposed iridium(III)/iridium(V) catalytic cycle because it resulted in considerable steric repulsion in the hepta-coordinated iridium(V) intermediate. Inspired by this steric effect, we have proposed a novel iridium(I)/iridium(III)-based catalytic cycle for this transformation and shown that it can be used to account for the exptl. results. The iridium(I)/iridium(III) catalytic cycle induced by this steric effect consists of several steps, including: (i) the oxidative addition of the C-H bond of the substrate to an active iridium(I) boryl complex; (ii) the reductive elimination of a C-B bond; (iii) the oxidative addition of B2pin2 to an iridium(I) hydride complex; and (iv) the reductive elimination of a B-H bond. Notably, the computed regioselectivity of this reaction was consistent with the exptl. observations. The high para-selectivity of this reaction was also explained using structural anal. and a 2D contour model, which revealed that the strong steric repulsion between the diphosphine ligand and the meta-substituents resulted in a higher energy barrier for meta-C-H activation. The experimental process involved the reaction of 2-(3-Methoxy-4-methylphenyl)-4,4,5,5-tetramethyl-1,3,2-dioxaborolane(cas: 1417036-28-2).Product Details of 1417036-28-2

N-Alkyl-N-(2-(1-arylvinyl)aryl)cinnamamides were converted into natural product inspired scaffolds via iridium photocatalyzed intramol. [2+2] photocycloaddition The protocol had a broad substrate scope, while operating under mild reaction conditions. Tethering four components forming a trisubstituted cyclobutane core builds rapidly high mol. complexity. This approach allowed the design and synthesis of a variety of tetrahydrocyclobuta[c]quinolin-3(1H)-ones, in yields ranging between 20-99%, and with excellent regio- and diastereoselectivity. Moreover, it was demonstrated that the intramol. [2+2]-cycloaddition of 1,7-enynes-after fragmentation of the cyclobutane ring-leads to enyne-metathesis-like products. Zou, Xiaoliang;Li, Yinwu;Ke, Zhuofeng;Xu, Senmiao research published 《 Chiral Bidentate Boryl Ligand-Enabled Iridium-Catalyzed Enantioselective Dual C-H Borylation of Ferrocenes: Reaction Development and Mechanistic Insights》, the research content is summarized as follows. Ferrocenes with planar chirality are an important class of privileged scaffolds for diverse chiral ligands and organocatalysts. The development of efficient catalytic asym. methods under mild reaction conditions is a long-sought goal in this field. Though many transition-metal-catalyzed asym. C-H activation methods were recorded during the last decade, most of them are related to C-C bond-forming reactions. Owing to the useful attribute of the C-B bond, the authors herein report an amide-directed Ir-catalyzed enantioselective dual C-H borylation of ferrocenes. The key to the success of this transformation relies on a chiral bidentate boryl ligand and a judicious choice of a directing group. The current reaction could tolerate a vast array of functionalities, affording a variety of chiral borylated ferrocenes with good to excellent enantioselectivities (35 examples, up to 98% enantiomeric excess). The authors also demonstrated the synthetic utility by preparative-scale reaction and transformations of a borylated product. Finally, from the observed exptl. data, the authors performed DFT calculations to understand its reaction pathway and chiral induction, which reveals that Me C(sp3)-H borylation is crucial to conferring high enantioselectivity through an amplified steric effect caused by an interacted B-O fragment in the transition state.

To study if the antibacterial activity of fingolimod could be further improved and to explore in-depth structure-activity relationships, 28 novel fingolimod derivatives I [R1 = 4-PhCH2C6H4, 4-OBnC6H4, 5-octyl-2-pyridyl, etc.; n = 1, 2], II [R2 = octyl, decyl] and III [R3 = H, octyl, decyl; R4 = H, octyl; Z = CH, N; m = 1, 2] were synthesized and evaluated their efficacy against Staphylococcus aureus grown in planktonic/single cell and biofilms. The most effective derivatives were tested on preformed S. aureus biofilms and against Gram-neg. bacteria Acinetobacter baumannii and Pseudomonas aeruginosa, using fingolimod as the reference compound Seven derivatives were more effective against S. aureus, while five other derivatives showed improved activity against P. aeruginosa and/or A. baumannii, with no apparent change in cytotoxicity on human cells. The most interesting derivatives, compounds I [R1 = 3-octylphenyl, n = 2] and II [R2 = decyl], displayed a broader spectrum of antibacterial activity, possibly exerted by the change of the para-hydrocarbon chain to a meta position for compound I [R1 = 3-octylphenyl, n = 2] and by an addnl. hydroxyl group for compound II [R2 = decyl].

Zhu, Zixi;Boger, Dale L. research published 《 N1/N4 1,4-Cycloaddition of 1,2,4,5-Tetrazines with Enamines Promoted by the Lewis Acid ZnCl2》, the research content is summarized as follows. The second example of selective N1/N4 1,4-cycloaddition (vs C3/C6 1,4-cycloaddition) of 1,2,4,5-tetrazines with preformed or in-situ generated enamines promoted by the Lewis acid ZnCl2 and with an expanded scope was described. The reaction constituted a formal [4+2] cycloaddition across two nitrogen atoms (N1/N4 vs C3/C6) of a 1,2,4,5-tetrazine followed by retro [4+2] cycloaddition loss of a nitrile and aromatization to provide 1,2,4-triazines. Optimization of reaction parameters, simplification of its implementation through in-situ enamine generation from ketones, definition of the enamine reaction scope for 3,6-bis(thiomethyl)-1,2,4,5-tetrazine, exploration of the 1,2,4,5-tetrazine scope, and representative applications of the product 1,2,4-triazines were detailed. The work established and further extended a powerful method for efficient one-step regioselective synthesis of 1,2,4-triazines under mild reaction conditions directly now from easily accessible ketones. It extended the substrate scope of a solvent (hexafluoroisopropanol) hydrogen bonding-promoted reaction that recently reported with aryl-conjugated enamines, permitting the use of simple ketone-derived enamines and expanding the generality of the remarkable reaction. The reaction was regioselective with respect to the site of reaction with unsym. ketones and provided exclusively a single 1,2,4-triazine regioisomer consistent with previously established stepwise mechanism of formal N1/N4 1,4-cycloaddition, overcoming the challenges observed in conventional approaches to 1,2,4-triazines.

M.;Alhumaydhi, Fahad A.;Suleria, Hafiz Ansar Rasul research published 《 LC-ESI-QTOF-MS/MS Characterization and Estimation of the Antioxidant Potential of Phenolic Compounds from Different Parts of the Lotus (Nelumbo nucifera) Seed and Rhizome》, the research content is summarized as follows. Edible lotus (Nelumbo nucifera G.) is widely consumed in Asian countries and treated as a functional food and traditional medicinal herb due to its abundant bioactive compounds Lotus rhizome peels, rhizome knots, and seed embryos are important byproducts and processing waste of edible lotus (Nelumbo nucifera G.) with com. significance. Nevertheless, the comprehensive phenolic profiling of different parts of lotus is still scarce. Thus, this study aimed to review the phenolic contents and antioxidant potential in lotus seeds (embryo and cotyledon) and rhizomes (peel, knot, and pulp) grown in Australia. In the phenolic content and antioxidant potential estimation assays by comparing to the corresponding reference standards, the lotus seed embryo exhibited the highest total phenolic content (10.77 ± 0.66 mg GAE/gf.w.), total flavonoid content (1.61 ± 0.03 mg QE/gf.w.), 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity (9.66 ± 0.10 mg AAE/gf.w.), 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) scavenging activity (14.35 ± 0.20 mg AAE/gf.w.), and total antioxidant capacity (6.46 ± 0.30 mg AAE/g), while the highest value of ferric ion reducing antioxidant power (FRAP) activity and total tannin content was present in the lotus rhizome knot (2.30 ± 0.13 mg AAE/gf.w.). A total of 86 phenolic compounds were identified in five parts of lotus by liquid chromatog. coupled with electrospray ionization quadrupole time-of-flight mass spectrometry (LC-ESI-QTOF-MS/MS), including phenolic acids (20), flavonoids (51), lignans (3), stilbenes (2), and other polyphenols (10). The most phenolic compounds, reaching up to 68%, were present in the lotus seed embryo (59). Furthermore, the lotus rhizome peel and lotus seed embryo exhibit significantly higher contents of selected polyphenols than other lotus parts according to high-performance liquid chromatog. (HPLC) quantification anal. The results highlighted that byproducts and processing waste of edible lotus are rich sources of phenolic compounds, which may be good candidates for further exploitation and utilization in food, animal feeding, and pharmaceutical industries.

In vivo, sinapic acid (4 mg/kg, p.o.) increases the time spent in the open arms of the elevated plus maze by approximately 15% in mice, an effect that can be blocked by the GABAA receptor antagonists flumazenil and bicuculline. Sinapic acid is also commonly used as a matrix in protein mass spectrometry.
Sinapic acid analytical standard provided with w/w absolute assay, to be used for quantitative titration.
Sinapic acid is an hydroxycinnamic acid derivative that occurs naturally in Brassicaceae species.
cis-Sinapic acid, also known as cis-sinapate or synapitic acid, belongs to the class of organic compounds known as hydroxycinnamic acids. Hydroxycinnamic acids are compounds containing an cinnamic acid where the benzene ring is hydroxylated. cis-Sinapic acid is considered to be a practically insoluble (in water) and relatively neutral molecule. Within the cell, cis-sinapic acid is primarily located in the cytoplasm. Outside of the human body, cis-sinapic acid can be found in common pea and pulses. This makes cis-sinapic acid a potential biomarker for the consumption of these food products.
Cis-sinapic acid is a 3-(4-hydroxy-3,5-dimethoxyphenyl)prop-2-enoic acid in which the double bond has cis-configuration. It has been isolated from the shoots of alfalfa. It has a role as a plant metabolite., Application In Synthesis of 530-59-6

A series of N-phenyl-7H-pyrrolo[2,3-d]pyrimidin-4-amine derivatives with NF-κB inducing kinase (NIK) inhibitory activity were obtained through structure-based drug design and synthetic chem. Among them, 4-(3-((7H-pyrrolo[2,3-d]pyrimidin-4-yl)amino)-4-morpholinophenyl)-2-(thiazol-2-yl)but-3-yn-2-ol (12f)(I) was identified as a highly potent NIK inhibitor, along with satisfactory selectivity. The pharmacokinetics of 12f and its ability to inhibit interleukin 6 secretion in BEAS-2B cells were better than compound 1 developed by Amgen. Oral administration of different doses of 12f in an imiquimod-induced psoriasis mouse model showed effective alleviation of psoriasis, including invasive erythema, swelling, skin thickening, and scales. The underlying pathol. mechanism involved attenuation of proinflammatory cytokine and chemokine gene expression, and the infiltration of macrophages after the treatment of 12f. This work provides a foundation for the development of NIK inhibitors, highlighting the potential of developing NIK inhibitors as a new strategy for the treatment of psoriasis.
