Like the kidney transamidinase, it catalyzes reversible arginine-ornithine and canavanine-ornithine transamidinations. The concept of an enzyme-amidine intermediate in transamidination reactions is supported by (a) the occurrence of the arginine-ornithine exchange reaction, coupled with specificity considerations, and (b) the fact that the formamidine moiety of the intermediate can be trapped with HONH2 to form hydroxyguanidine. Formamidine disulfide is a potent inhibitor of transamidinase. It is suggested that its inhibitory action results from the formation of a mixed disulfide with an essential sulfhydryl group on the enzyme. A sensitive colorimetric assay for hydroxyguanidine is described.
