Synthesis of compound 2 is shown in Figure 3. The synthesis began with converting chloroacetonitrile (13) to the sulfonic acid and then to the sulfonyl chloride (14) in 13% yield over two steps. Sulfonyl chloride (14) was found to be relatively unstable and therefore was quickly reacted with amine-TEG (7) to give sulfonamide-TEG (15) in 86% yield.
